To detect the molecular changes of malignancy in histologically normal breast (HNB) tissues, we recently developed a novel 117-gene-malignancy-signature. Here we report validation of our leading malignancy-risk-genes, topoisomerase-2-alpha (TOP2A), minichromosome-maintenance-protein-2 (MCM2) and “budding-uninhibited-by-benzimidazoles-1-homolog-beta” (BUB1B) at the protein level. Using our 117-gene malignancy-signature, we classified 18 fresh-frozen HNB tissues from 18 adult female breast cancer patients into HNB-tissues with low-grade (HNB-LGMA; ) and high-grade molecular abnormality (HNB-HGMA; ). Archival sections of additional HNB tissues from these patients, and invasive ductal carcinoma (IDC) tissues from six other patients were immunostained for these biomarkers. Ovarian epithelial cancer is the most lethal gynecologic malignancy. The high mortality is attributed to the fact that most cases typically present in late stage when ovarian cancer (OC) has already spread beyond the ovary. Ovarian epithelial cancer cells are shed into intraperitoneal ascites and easily disseminate throughout the peritoneal cavity with preferential metastasis to the omentum, peritoneum, and local organs. Understanding how ovarian epithelial cells interact with and modulate their microenvironment can provide insight into the molecular mechanism(s) involved with malignant transformation and progression which may eventually identify novel diagnostic, prognostic, and therapeutic targets. Recent research has provided mechanistic insight into the important contributions of the gut microbiota to vertebrate biology, but questions remain about the evolutionary processes that have shaped this symbiosis. In the present study, we showed in experiments with gnotobiotic mice that the evolution of Lactobacillus reuteri with rodents resulted in the emergence of host specialization. To identify genomic events marking adaptations to the murine host, we compared the genome of the rodent isolate L. reuteri 100-23 with that of the human isolate L. reuteri F275, and we identified hundreds of genes that were specific to each strain. One such supplement is XS® Energy Drink which contains B vitamins, caffeine, taurine, and an herbal adaptogen blend. Local regulation of vascular tone plays an important role in cardiovascular control of blood pressure. Aside from chemical or hormonal regulations, this local homeostasis is highly regulated by fluid-shear stress. It was previously unclear how vascular endothelial cells were able to sense fluid-shear stress. The cellular functions of mechanosensory cilia within vascular system have emerged recently. In particular, hypertension is insidious and remains a continuous problem that evolves during the course of polycystic kidney disease (PKD). The basic and clinical perspectives on primary cilia are discussed with regard to the pathogenesis of hypertension in PKD.

Despite the success of potent reverse transcriptase (RT) inhibitors against human immunodeficiency virus type 1 (HIV-1) in combination regimens, the development of drug resistant RTs constitutes a major hurdle for the long-term efficacy of current antiretroviral therapy. Nucleoside β-triphosphate analogs of adenosine and nucleoside reverse transcriptase inhibitors (NRTIs) (3′-azido-2′,3′-dideoxythymidine (AZT), 3′-fluoro-2′,3′-dideoxythymidine (FLT), and 2′,3′-didehydro-2′,3′-dideoxythymidine (d4T)) were synthesized and their inhibitory activities were evaluated against wild-type and multidrug resistant HIV-1 RTs. KX2-391 (KX-01/Kinex Pharmaceuticals), N-benzyl-2-(5-(4-(2-morpholinoethoxy)phenyl)pyridin-2-yl)acetamide, is a highly selective Src substrate binding site inhibitor. To understand better the role of pyridine ring and N-benzylsubstitution in KX2-391 and establish the structure-activity relationship, a number of N-benzyl substituted (2-morpholinoethoxy)phenyl)thiazol-4-yl)acetamide derivatives containing thiazole instead of pyridine were synthesized and evaluated for Src kinase inhibitory activities. The unsubstituted N-benzyl derivative (8a) showed the inhibition of c-Src kinase with GI50 values of 1.34 μM and 2.30 M in NIH3T3/c-Src527F and SYF/c-Src527F cells, respectively. Objective—A single Pro-70 to Thr (p.P70T) mutation of amelogenin is known to result in hypomineralized amelogenesis imperfecta (AI). This study aims to test the hypothesis that the given mutation affects the self-assembly of amelogenin molecules and impairs their ability to conduct the growth of apatite crystals.

Design—Recombinant human full-length wild-type (rh174) and p.P70T mutated amelogenins were analyzed using dynamic light scattering (DLS), protein quantification assay and atomic force microscopy (AFM) before and after the binding of amelogenins to hydroxyapatite crystals. The crystal growth modulated by both amelogenins in a dynamic titration system was observed using AFM.

This study examined the gene transfer efficiency and toxicity of 2-kDa polyethylenimine conjugated to gold nanoparticles (PEI2-GNP) in the human cornea in vitro and rabbit cornea in vivo. PEI2-GNP with nitrogen-to-phosphorus (N/P) ratios of up to 180 exhibited significant transgene delivery in the human cornea without altering the viability or phenotype of these cells. Similarly, PEI2-GNP applied to corneal tissues collected after 12 h, 72 h, or 7 days exhibited appreciable gold uptake throughout the rabbit stroma with gradual clearance of GNP over time. Covalent attachment of carbohydrates to proteins is one of the most common post-translational modifications. At the cell surface, sugar moieties of glycoproteins contribute to molecular recognition events involved in cancer metastasis. We have combined glycan metabolic labeling with mass spectrometry analysis to identify and characterize metastasis-associated cell surface sialoglycoproteins. Our model system used syngeneic prostate cancer cell lines derived from PC3 (N2, nonmetastatic, and ML2, highly metastatic). The metabolic incorporation of AC4ManNAz and subsequent specific labeling of cell surface sialylation was confirmed by flow cytometry and confocal microscopy. The 15-kDa selenoprotein (Sep15) is a thioredoxin-like, endoplasmic reticulum-resident protein involved in the quality control of glycoprotein folding through its interaction with UDP-glucose:glycoprotein glucosyltransferase. Expression of Sep15 is regulated by dietary selenium and the unfolded protein response, but its specific function is not known. In this study, we developed and characterized Sep15 KO mice by targeted removal of exon 2 of the Sep15 gene coding for the cysteinerich UDP-glucose:glycoprotein glucosyltransferase-binding domain. BACKGROUND: Metal-containing proteins comprise a diverse and sizable category within the proteomes of organisms, ranging from proteins that use metals to catalyze reactions to proteins in which metals play key structural roles. Unfortunately, reliably predicting that a protein will contain a specific metal from its amino acid sequence is not currently possible. We recently developed a generally-applicable experimental technique for finding metalloproteins on a genome-wide scale. PURPOSE. This study tested a hypothesis that tissue-selective targeted decorin gene therapy delivered to the stroma with adeno-associated virus serotype 5 (AAV5) inhibits corneal fibrosis in vivo without significant side effects.

METHODS. An in vivo rabbit model of corneal fibrosis was used. Targeted decorin gene therapy was delivered to the rabbit cornea by a single topical application of AAV5 (100 L; 6.5 1012 g/mL) onto the bare stroma for 2 minutes. To evaluate hypolipidemic and anti-oxidant activities of Monascus purpureus fermented Indian variety of rice. Materials and methods: Indian rice variety IR-532-E-576 was fermented with Monascus purpureus MTCC 1090 (Monascaceae) and extracted with water by boiling. Experimentally induced hyperlipidemia was produced by feeding rats with 2% cholesterol enriched diet for 15 days. To compare the serum levels of adiponectin and resistin between patients with polycystic ovary syndrome (PCOS) and healthy control subjects matched for age and body mass index (BMI), and to assess possible correlations of adiponectin and resistin to the hormonal and metabolic parameters of the syndrome. Materials and methods: Fifty five patients with PCOS diagnosis composed of 25 obese, 13 overweight, 17 normal weight subjects, and 49 healthy women matched for age and BMI were included in this study. In childhood, urticarial lesions are sometimes associated with purpura. This form might be identified as atypical, and may also be related to urticarial vasculitis (UV). The aim of this study was to assess the clinicopathologic characteristics of UV in children with atypical urticaria. Materials and methods: Fifteen children with atypical urticaria were evaluated with medical history, physical examination, and laboratory and skin punch biopsy findings. Results: Infections were detected as possible precipitating factor in 12 patients. Complement levels were normal in all. The prepuce was excised after achieving successful hypospadias repair with satisfactory functional and cosmetic appearance. The patients who underwent preputial reconstructions and complicated repairs were excluded. The control group consists of boys without hypospadias. Community acquired acute bacterial cystitis (CA-ABC) is one of the most common infectious presentations in the community. This paper aims to provide a rational approach in the management of CA-ABC based on local Turkish data. Materials and methods: The publications evaluating the microbiological data obtained from CA-ABC cases were searched in both Turkish and international databases. Results: Fosfomycin and nitrofurantain appear to be baseline antibiotics while the empirical use of trimethoprim sulfamethoxazole, amoxicillin clavulanate, and first and second generation cephalosporins seems to be unreliable in Turkey. A law prohibiting smoking in the enclosed areas of public and private workplaces was put into effect in Turkey on 19 May 2008. The objective of this study was to evaluate the effect of this law upon the smoking habits of the employees in this short period of 3 months. The continuous stratified layer of human oral keratinocytes was grown on a cadaveric human dermal matrix in a defined culture medium without a feeder layer. Materials and methods: Human oral keratinocytes from a keratinized oral mucosa biopsy specimen were enzymatically dissociated and cultured in a serum-free defined culture medium, "EpiLife" (Cascade Biologics, Portland, OR, USA), containing a low calcium concentration of 0.06 mM. There is no national screening program for rubella, Toxoplasma gondii, and cytomegalovirus (CMV) infections for pregnant women in Turkey. The purpose of this study was to determine the prevalence of these infections among pregnant women in Denizli province, and to review seroprevalences reported from different regions of Turkey. Materials and methods: Between April 2008 and April 2009, 1268 women were investigated for rubella, 1102 for toxoplasma, and 1000 women for CMV seroprevalences in the first trimester of pregnancy. The effects of dacarbazine, thiocolchicoside, methotrexate, olanzapine, pantoprazole sodium, and 5-fluorouracil on the enzyme activity of glutathione reductase (GR) were studied using human erythrocyte GR enzymes in in vitro studies, respectively. Materials and methods: The enzyme was purified 3333-fold from human erythrocytes in a yield of 44.44% with 4.0 U/mg. The purification procedure involved the preparation of hemolysate, ammonium sulfate precipitation, 2', 5'-ADP Sepharose 4B affinity chromatography, and Sephadex G-200 gel filtration chromatography. There are several studies on tricyclic antidepressant (TCA) intoxication in the literature; however, there is no specific study on amitriptyline, a specific TCA, intoxication. Materials and methods: Forty adult patients who had been admitted to the emergency department due to suicide attempts with amitriptyline were included.
