The non-structural protein 1 (nsp1) of porcine reproductive and respiratory syndrome virus is partly responsible for inhibition of type I interferon (IFN) response by the infected host. By performing alanine-scanning mutagenesis, we have identified amino acid residues in nsp1α and nsp1β~ (the proteolytic products of nsp1) that when substituted with alanine(s) exhibited significant relief of IFNsuppression. A mutant virus (16-SA, in which residues 16-20 of nsp1β were substituted with alanines) encoding mutant nsp1β recovered from infectious cDNA clone was shown to be attenuated for growth in vitro and induced significantly higher amount of type I IFN transcripts in infected macrophages. …

The porcine reproductive and respiratory syndrome virus (PRRSV) glycoprotein 4 (GP4) resembles a typical type I membrane protein in its structure but lacks a hydrophilic tail at the C-terminus, suggesting that GP4 may be a lipid-anchored membrane protein. Using the human decay-accelerating factor (DAF; CD55), a known glycosyl-phosphatidylinositol (GPI) lipid-anchored protein, chimeric constructs were made to substitute the GPI-anchor domain of DAF with the putative lipid-anchor domain of GP4, and their membrane association and lipase cleavage were determined in cells. Using a recombinant vesicular stomatitis virus encoding eGFP fused in-frame with an essential viral replication protein, the phosphoprotein P, we show that during passage in culture, the virus mutates the nucleotide C289 within eGFP of the fusion protein PeGFP to A or T, resulting in R97S/C amino acid substitution and loss of fluorescence. The resultant non-fluorescent virus exhibits increased fitness and growth advantage over its fluorescent counterpart. The growth advantage of the non-fluorescent virus appears to be due to increased transcription and replication activities of the PeGFP protein carrying the R97S/C substitution. Porcine reproductive and respiratory syndrome virus (PRRSV) suppresses tumor necrosis factor-alpha (TNF-α) production at both transcriptional and post-transcriptional levels by its non-structural proteins 1α and 1β (Nsp1α and Nsp1β). To identifY the amino acid residues responsible for this activity, we generated several alanine substitution mutants of Nsp1α and Nsp1β. Examination of the mutant proteins revealed that Nsp1α residues Gly90, Asn91 , Arg97, Argl 00 and Arg124 were necessary for TNF-α promoter suppression, whereas several amino acids spanning the entire Nsp1β ~ were found to be required for this activity. In order to resolve the location and activity of submicroscopic viruses in living cells, viral proteins are often fused to fluorescent proteins (FPs) and visualized by microscopy. In this study, we describe the fusion of FPs to three proteins of pseudorabies virus (PRV) that allowed imaging of capsids in living cells. Included in this study are the first recombinant PRV strains expressing FP–pUL25 fusions based on a design applied to herpes simplex virus type 1 by Homa and colleagues. Staphylococci are a versatile genus of bacteria that are capable of causing acute and chronic infections in diverse host species. The success of staphylococci as pathogens is due in part to their ability to mitigate endogenous and exogenous oxidative and nitrosative stress. Endogenous oxidative stress is a consequence of life in an aerobic environment; whereas, exogenous osidative and nitrosative stress are often due to the bacteria's interaction with host immune systems. Background: A number of constitutively activated signaling pathways play critical roles in the survival and growth of primary effusion lymphoma cells (PELs) including NFkB and PI3/AKT kinase cascades. NFkBis constitutively activated in a number of malignancies, including multiple myeloma, Burkitt’s lymphoma and diffuse large cell B-cell lymphoma. However, its role in primary effusion lymphoma has not been fully explored.

Trans-cranial Direct Current Stimulation has been shown to modulate cortical neuronal activity. Weak constant current is applied to the scalp using electrodes, leading to sub-threshold changes in neuronal membrane potential. Before actual clinical stimulation is performed, such systems are validated by simulating the stimulation and the resulting current flow patterns using finite element solvers. However, the reliability of such models depends upon the accuracy with which the underlying anatomy has been modeled [1]. To evaluate the cephalometric features of patients with obstructive sleep apnea syndrome (OSAS) and to elucidate the relationship between the severity of OSAS and cephalometric and clinical variables. Materials and methods: Ninety-three patients (71M/22W) with OSAS were classified into 3 subgroups according to the apnea-hypopnea index (AHI). Various cephalometric and clinical measurements were carried out for all patients. Results: Inferior airway space (IAS), maximum soft palate thickness (MSPT), SNGoGn°, and distance of hyoid bone to mandibular plane (Hy-MPPerp) were found to be higher in severe OSAS for both sexes. To identify risk factors associated with cigarette smoking during pregnancy and to evaluate the effect of smoking on anthropometric measurements. Materials and methods: This study was carried out prospectively in selected women who gave birth to a healthy, term infant at Şişli Etfal Education and Research Hospital from January 2009 to January 2010. Smoking status during pregnancy was categorized into 3 groups: nonsmoker; smoker; passive smoker. Regression analysis was performed to compare risk factors with cigarette smoking. In the present study, we investigated the extent to which changes in corticosterone level and food and water intake behavior occur in diabetes and explored the effect of Tinospora cordifolia extracts and insulin therapy in streptozotocin-induced diabetic rats. The aim of this study was to evaluate the effectiveness of an original vaginal plication of the uterosacral ligaments as a preventive procedure for recurrent vault prolapse. Materials and methods: In total, 216 women with stage III/IV genital prolapse (POPQ system) underwent vaginal hysterectomy combined with uterosacral ligaments plication. First, the rectum was dissected from the posterior vaginal wall; next, 3 absorbable sutures were placed through both uterosacral ligaments; finally, a fourth suture was placed circularly through both uterosacral ligaments and the posterior vaginal margin. To determine the presence of Entamoeba histolytica/E. dispar and E. moshkovskii in stool samples, tRNA-based short tandem repeat gene polymorphism in E. histolytica isolates, and the relationship between amoeba load and clinical outcome in studies. Materials and methods: This study involved 840 stools samples of individuals having diarrhea/dysentery and individuals who were asymptomatic by using microscopy, culture, E. histolytica antigen ELISA, and conventional/real-time PCR methods. To assess the genotoxic and cytotoxic effects of banding procedure with 5 different orthodontic cements on human oral buccal epithelium cells. Materials and methods: Fifty healthy volunteers (mean age: 14.54 ± 2.37 years) were randomly divided into 5 groups of 10. To define a high-risk group for paraaortic (PA) lymph node metastasis among endometrial carcinoma patients. Materials and methods: Prognostic factors determining PA lymph node metastasis were defined. Using these factors in different combinations, 14 risk groups were formed. A patient with at least one of these factors was considered as high-risk for PA lymph node metastasis. Results: This study included 152 patients. Of these patients, 18 had tumors in the PA region. Lymphovascular space invasion (LVSI) and pelvic lymph node metastasis were independent prognostic factors for PA lymph node involvement. The aims of this study were to estimate the preoperative thyroid volume in patients with a multinodular goiter by the use of ultrasonography (USG) and magnetic resonance imaging (MRI), and then to compare these approaches with the postsurgical total volume measured by Archimedes' principle. Approximately 40 diaphragmatic lipoma cases have been reported in the literature to date. Diaphragmatic lipoma is 2 times more likely to appear on the left rather than the right side. In the present study, diaphragmatic lipoma was evaluated in patients who underwent surgical management in light of the literature data. To retrospectively review the epidemiologic characteristics of patients who underwent surgery for hip fracture. Materials and methods: Patients who underwent surgery for hip fracture in our clinic between 2006 and 2010 were evaluated in terms of their age, sex, localization of fracture, and surgical procedure. Results: The mean age of 419 patients who underwent surgery for hip fracture was 74.18 ± 11.26 (50-106). Of the patients, 55.13% were operated on for intertrochanteric femur fractures, 33.41% for collum femoris fractures, and 11.46% for subtrochanteric femur fractures. Materials and methods: In this study, 608 patients (397 male and 211 female) admitted to a smoking cessation clinic to quit smoking between 1 January 2006 and 1 June 2010 were investigated. Interactions mediated by intercellular adhesion molecule-1 (ICAM-1) enhance both the antigenic and cytokine signals that CD8 T cells perceive, and these interactions have been suggested to be required for the formation of memory CD8 T cells. Following acute lymphocytic choriomeningitis virus (LCMV) infection of ICAM-1-/- mice, we observe elevated numbers of "effector-phenotype" (CD127lo KLRG-1hi IL-2-) virus-specific CD8 T cells well into the memory phase, while the development of memory-phenotype CD8 T cells (CD127hi KLRG-1lo IL-2 producing) is not impaired.
