The aim of this study was to design a simple method for detecting the amyloid A4 protein in the cerebrospinal fluid (CSF) of patients who had severe brain trauma caused by traffic or other accidents and to evaluate its diagnostic and prognostic value. For this purpose, a sandwich type ELISA assay was used. A4, a 4kDa protein of 39-43 amino acids, is a metabolic product of a membrane-spanning, 695-770 aa, precursor molecule, the b-amyloid precursor protein (b-APP). The levels of interleukin-1beta, tumour necrosis factor-alpha, and interleukin 6 in gingival crevicular fluid of phenytoin-treated patients (from sites exhibiting overgrowth and sites not exhibiting overgrowth) and control subjects were investigated. Six epileptic patients exhibiting severe phenytoin-induced gingival overgrowth, 12 patients with chronic gingivitis and 11 subjects with clinically healthy periodontium were included in the study. The cytokine levels were analysed by enzyme-linked immunosorbent assay and expressed as ng/2 sites. The numbers of immunocom-promised individuals increase as programmes for organ transplantation and chemotherapy of malignant disease advance. Pulmonary infections, which progress rapidly, are one of the major clinical problems in these groups. This study was designed to evaluate the microbiology of pneumonia in immunocompromised patients and to determine the role of Legionella pneumophila. Immunocompromised patients receiving antineoplastic chemotherapy, diagnosed as pneumonia with clinical and radiographic findings and routine laboratory tests were included in the study. In this study, the effects of sulfur dioxide (SO_2) ,a common pollutant in urban air, on t-butyl hydroperoxide-induced oxidative stress in rat lung tissues were examined. The aims of this study were to determine the role of interstitial and glomerular myofibroblasts in the progression of various glomerulonephritis, and to assess the value of myofibroblasts in determining long-term renal outcome. The biopsy specimens of 42 primary glomerulonephritis cases were reviewed and stained immunohistochemically with alpha smooth muscle actin antibody. The serum creatine level was compared to the pathological changes. The interstitial alpha smooth muscle actin staining correlated with the serum creatine level at the time of biopsy. The aim of the study was to establish apolipoprotein E genotype in the patients with coronary heart disease (CHD) who are inhabitants of the north-east of Turkey. The study involved 106 patients (70 male, 36 female; ages ranging from 37 to 69 years) who are local people and were angiocardiographically diagnosed for CHD. The risk factors such as smoking, alcohol and obesity were determined and serum triglyceride, total cholesterol, LDL and HDL were measured in the subjects. This study was designed to determine the prevalence of osteopathy in diabetic patients, who are at risk of developing osteopathy, and clarify the relationship between osteopathy and other complications of diabetes mellitus. Thirty-four type I and 66 type II diabetic patients admitted to the endocrinology clinic between 1996 and 1997 and 30 nondiabetic subjects as a control group were studied. Corticosteroids, widely used in the treatment of collagen tissue diseases, increase the number of neutrophils and decrease the total numbers of lymphocytes, monocytes, eosinophils and basophils. In addition, corticosteroids alter important functional activities of both lymphocytes and monocytes. They suppress the bactericidial activities of phagocytes. In this manner, the resistance to infections is reduced. Cytotoxic drugs can have profound effects on the production and function of both phagocyte cells and lymphocytes. In Vivo Interaction Between Cadmium And Essential Trace Elements Copper And Zinc In Rats, H. Hakan Aydin, Canan Çoker, Bi̇ltan Ersöz

The complex in vivo interaction between Cd, a toxic metal and essential trace elements, mainly Zn and Cu, has not been elucidated yet. The objective being the elucidation of this interrelationship, Cd was subcutaneously administered as CdCI_2 (1 mg/kg/day) for 5 consecutive days to Swiss albino male rats (n=10). After 5 days, the rats were decapitated. Cd, Zn and Cu levels were estimated in hepatic, renal, cardiac and skeletal muscle specimens. Cd was found to be significantly elevated in all tissues (p< 0.001). Zn was increased in hepatic and renal tissues (p< 0.001 and p< 0.05) compared to the controls. Cu was also significantly increased in these tissues. Myocardial and skeletal muscle tissues also manifested a significant increase in Cu and Zn in this group. It is concluded that Cd administration alters the Zn and Cu status in vivo. However, the mechanism underlying the interactions between toxic and essential elements should be further investigated.

Absorption of zinc by preterm infants from a synthetic milk labelled extrinsically with 70Zn tracer was determined by faecal monitoring. Six male preterm babies with a mean gestational age of 31±2.9 weeks and a mean birth weight of 1.23±0.24 kg were studied for three balance periods of 72 h with intervals ranging from 6 to 30 days. The thalamus is one of the most vulnerable areas for intracerebral hemorrhages. In this study, we investigated the parameters of thalamic hemorrhage such as risk factors, initial complaints, neurologic findings and the prognostic effects of the presence or absence of ventricular penetration, as well as the volume and size of hematoma. A total of 57 patients with thalamic hemorrhage, whose diagnosis was confirmed by cranial computerized tomography, were evaluated. Recent advances in molecular biology enable researchers to understand the basis of breast cancer much better and show that hereditery breast cancer may result from mutations on several specific gene loci including BRCA1, BRCA2, p53, ATM and PTEN. These genes are tumor supressor genes and although their functions are different, they are all involved in the maintenance of genomic stability after DNA damage. We investigated the properties of action potential after-potentials in cultured dorsal root ganglion neurones from neonatal rats using the whole cell patch clamp technique. The aims of this study were to characterize after-potentials and investigate the possible influences of Ca^{2+}-induced Ca^{2+}release (CICR) on neuronal excitability. The reversal potentials of action potential after-depolarizations and after-hyperpolarizations were estimated. Although evidence for both Ca^{2+}-activated chloride and potassium conductances was obtained, the data indicated that the after-potentials were contaminatied by a variety of distinct ionic events. Apamin was used to attenuate potassium conductances and appeared to enhance the after-depolarizations due to Ca^{2+}-activated chloride conductances. …

Various relations between apolipoprotein E (apo E) and diabetic nephropathy have been reported. In this study, the question of whether there is any relation between apo E genotypes and microalbuminuria in patients with type-2 diabetes mellitus was examined. Twenty-one male and 25 female patients, with ages the range of 37-62 years (mean 51.7±7.5) with type-2 diabetes mellitus were studied. The apo E genotypes were determined by PCR amplification of the 227 bp region followed by CfoI digestion to release specific band patterns. In the present study involving patients with bone metastases arising from prostate cancer, we measured urinary deoxypyridinoline (DPD) as a marker of collagen breakdown activity, serum total and bone-specific alkaline phosphatase activities and serum prostate specific antigen (PSA). This study included 20 patients with benign prostate hyperplasia (BPH) and 23 patients with carcinoma, 11 of had with bone metastases. Aim: We investigated the role of c-erb-B2, hormone receptors and cathepsin D in breast carcinoma progression and their correlation with each other. Methods: Forty-six breast carcinoma patients, operated on in the deparment of surgery and diagnosed in the pathology department, were classified according to their histopathological characteristics; the results of immunostaining with estrogen and progesterone receptors, c-erb-B2 and cathepsin-D were correlated with the stage and the disease-free survival of the patients. Workers in the tobacco industry are exposed to tobacco dust, which has allergenic and irritative effects on airways. In this study, our aim was to determine whether tobacco dust has any effect on lung functions and volumes. A questionnaire about working conditions was applied to 448 workers, some of which had been exposed to tobacco dust while others had not, and all workers underwent measurements of spirometric flow and volume. Forty-eight upper limbs were dissected in order to study the courses and relations of the brachial plexus nerves in the axilla. Connections between the musculocutaneous and median nerves were found in five arms. There was a suprascapular nerve arising from the union of C4 and C5 directly in one cadaver. A branch from the median nerve extending to the brachial artery was determined. In one of the cadavers the radial nerve arose only from the union of posterior divisions of the inferior and middle trunks.

Objective: To determine the mechanism of action of Methotrexate and Vepesid on the HEp-2 cells isolated from human laryngeal cancer cells morphologically and flow cytometrically (G1, G2, S and PI). Materials and Methods: The HEp-2 continuous cell line was used. Cultivation of the cells was realized in EMEM medium with 10% fetal bovine serum at an atmosphere of 37ºC with 5% CO_2. Six different concentrations of Vepesid and Methotrexate were prepared by diluting with deionized water (5 µg/ml, 50 mg/ml, 500 µg/ml). B(a)P is a mutagenic, carcinogenic and teratogenic substance. Paclitaxel and doxorubicin are antineoplastic drugs widely used in cancer treatment. The purpose of this study is to observe the effects of doxorubicin and paclitaxel on p170 glycoprotein in rat liver and kidney tissue after administration of B(a)P. As is well known, p170 glycoprotein is an indicator of drug resistance. We hypothesized that a combination of these antineoplastic drugs would cause lower p170 levels and thus would have a stronger effect than single drug administration. H. influenzae is a fastidious bacteria, so the selection of a reliable method for in-vitro susceptibility tests is particularly important. The E-test is a practical method for detecting in-vitro antimicrobial susceptibility of antibiotics by MIC values. This study was performed using the E-test for direct patient management and in order to obtain epidemiological data on H. influenzae. The detection of antimicrobial agent resistance among fifty-seven H. influenzae isolates from lower respiratory tract infections in childhood was studied by the E-test.
