How well an athlete tolerates pain may aid the athletic trainer in determining a proper rehabilitation protocol for the quickest recovery and return to play after an injury. The present investigation was conducted to determine if there is a difference in pain tolerance between NCAA Division I-A male and female athletes. The subjects consisted of nineteen healthy Division I-A male athletes and nineteen healthy Division I-A female athletes all between the ages of 18 and 23 (N=38). In order to determine the strategies in regulation of bone mass in the rat and better understand the pathophysiological mechanism of the postmenopausal osteoporosis, a series of studies were conducted at the whole animal and cellular level. At the whole animal level, female Sprague-Dawley rats were ovariectomized (Ovx) or sham-operated (control) at 18 weeks and the entire skeleton obtained at 24 weeks (baseline) or after an additional 31 day (28 week) interval on a normal (1.0%) or deficient (0.02%) calcium diet. Ovx rats on the normal calcium diet showed a 42% increase in whole body bone resorption and a 70% …

The Skn7 response regulator has previously been shown to play a role in the induction of stress-responsive genes in yeast, e.g., in the induction of the thioredoxin gene in response to hydrogen peroxide. The yeast Heat Shock Factor, Hsf1, is central to the induction of another set of stress-inducible genes, namely the heat shock genes. These two regulatory trans-activators, Hsf1 and Skn7, share certain structural homologies, particularly in their DNA-binding domains and the presence of adjacent regions of coiled-coil structure, which are known to mediate protein–protein interactions. In the nucleus, transcription factors must contend with the presence of chromatin in order to gain access to their cognate regulatory sequences. As most nuclear DNA is assembled into nucleosomes, activators must either invade a stable, preassembled nucleosome or preempt the formation of nucleosomes on newly replicated DNA, which is transiently free of histones. We have investigated the mechanism by which heat shock factor (HSF) binds to target nucleosomal heat shock elements (HSEs), using as our model a dinucleosomal heat shock promoter (hsp82-ΔHSE1). The Arkansas Rous Sarcoma Regressor (AR) and Progressor (AP) lines of chickens represent an excellent model to study immune responses to tumors. Both lines of chickens initially develop tumors when injected with Rous sarcoma viros (RSV) or with DNA coding for the RSV oncogene v-src (v-src DNA). AR chickens will eventually regress the tumors whereas AP chickens will allow the tumors to progress to a terminal stage. By using both v-src DNA and RSV for tumor induction, we were able to compare the immune response to tumor antigens alone and tumor antigens in combination with viral components, respectively. The original authors of Billman et af. (2000) are joined by three other analysts from the Cowboy Wash research team to respond to the critique of this article by Dongoske et af. (2000). Dongoske and his coauthors state that Billman et af. (2000) failed to test alternative hypotheses or to consider alternative explanations for the findings at 5MTJOOJO and similar sites. The original authors point out that alternative hypotheses were examined and rejected, leaving a violent episode of cannibalism as the most plausible explanation for the remains found at 5MTJOOJO. This investigation examined the utility of three recombinant protein-expression systems (COS cells, insect cells and insect larvae) to cost-effectively produce biologically active human butyrylcholinesterase (BuChE). It was determined that baculovirus-infected insect cells (Sf9 and High 5) expressed 3.5- and 8.2-fold, respectively, more active enzyme than COS-7 cells. Baculovirus-infected cabbage looper (Trichoplusia ni) insect larvae produced over 26 times more than High 5 cells ; in fact, one baculovirus-infected insect larva provided more active protein than 100 ml of insect cell culture. Analysis of the larvally expressed proteins revealed that the vast majority of BuChE expressed was inactive due to …

Abstract
Background
PCR amplification of target molecules involves sequence specific primers that flank the region to be amplified. While this technique is generally routine, its applicability may not be sufficient to generate a desired target molecule from two separate regions involving intron /exon boundaries. For these situations, the generation of full-length complementary DNAs from two partial genomic clones becomes necessary for the family of low abundance genes.
Results

Enalapril and its metabolite Enalaprilat were determined by gas chromatography - mass spectrometry (GC - MS) and high - performance liquid chromatography (HPLC). For HPLC, the detection limit of enalapril and enalaprilat was 10n g/ml in methanol, whereas for the GC - MS, the detection limit was 6.25ng/ml in plasma. These methods were compared and GC - MS was selected for routine analysis. Enalapril and Enalaprilat in plasma were extracted and cleaned up by using Sep - Pak C 18. Captopril is a highly specific competitive inhibitor of the angiotensin-convening enzyme (ACE). This enzyme converts angiotensin I, a relatively inactive decapeptide, to angiotensin II , a potent endogenous vasoconstrictor substance. Captopril is widely used for the treatment of hypertension and congestive heart failure. This drug, which contains a sulphydryl group binds readily to albumin and to other plasma proteins. Captopril is unstable in blood and plasma ex vivo because of the reactivity of sulphydryl group which results in oxidation and rapid formation of disulphides. Therefore a fixative or a stabiliser must be added to each blood sample immediately following collection. …

The nociceptive effect was measured using withdrawal latency in tail flick test in mice rendered diabetic by administering streptozotocin (200 mg/kg, i.p.). The antinociceptive effect of morphine (4 and 8 mg/kg, s.c.) and cromakalim, a KATP channel opener, (0.3, 1 and 2 micrograms, i.c.v.) was significantly reduced in diabetic mice. Moreover, co-administration of cromakalim(0.3 microgram) did not alter the reduced antinociceptive effect of morphine(4 mg/kg) in diabetic mice. Spleenectomy in diabetic mice restored the decrease in antinociceptive effect of morphine and cromakalim. Multiple dose treatment with insulin to maintain euglycaemia for 3 days in diabetic mice prevented the decrease in …

The importance of interactions between Sox and POU transcription factors in the regulation of gene expression is becoming increasingly apparent. Recently, many examples of the involvement of Sox-POU partnerships in transcription have been discovered, including a partnership between Sox-2 and Oct-3. Little is known about the mechanisms by which these factors modulate transcription. To better understand the molecular interactions involved, we mapped the location of the transactivation do-main of Sox-2. This was done in the context of its interaction with Oct-3, as well as its ability to transactivate as a fusion protein linked to the DNA-binding domain of Gal4. Both …

Mycobacterium avium is the most frequent cause of disseminated bacterial infection in patients with human immunodeficiency virus type 1 infection and in rhesus macaques with simian immunodeficiency virus (SIV) infection. This animal model of AIDS was used to test the hypothesis that this frequent association is the result of reciprocal enhancement of replication of both microorganisms. The replication of M. avium and SIV was analyzed in lymphatic tissues obtained from rhesus macaques experimentally inoculated with SIVmac who developed or remained free of overt M. avium infection. Listeria monocytogenes is a gram-positive, food-borne human and animal pathogen. L.innocua is a non-pathogenic species the most closely related to L. monocytogenes. Comparative genomics between a pathogenic and a closely related non-pathogenic species is a powerful approach for identifying genes or regions on the chromosome, which might be involved in virulence. Therefore whole genome sequencing of these two organisms was undertaken. With the rapid advances in the field of biotechnology during the last decade, many peptides and proteins have been produced and evaluated for therapy of various diseases, including cancer. However, rapid clearance and the possibility of immunogenicity after the in vivo administration of these biotechnology-driven products have impeded their marketing. To circumvent these problems, synthetic and natural polymers such as polyethylene glycol (PEG) and dextrans, respectively, have been covalently attached to proteins, and some of these protein-polymer conjugates have shown promising therapeutic results. The study was conducted at Alata Horticultural Research institute between 1994 and 1996. The aim of the study was to obtain a strong colony by way of supersedure and empty frames, feeding with saccharose syrup and encouraging egg laying of queen bees at the end of summer. According to the results, there were significant differences among treatments in terms of the development of colony population, brood rearing activity and honey yield. Treatments had no significant effect on wax building activity and swarming tendency.

This study was carried out to investigate the fattening and carcass characteristics of German Blackheaded Mutton (GBM) x Awassi (A) F1, Hampshire Down (HD) x A (F1) and HD x (HD x A) B1 male lambs. Nine GBM x A (F1), eight (HD) x A (F1) and eight HD x (HD x A) B1 male lambs were used for fattening traits, and six of each breed for carcass traits. This study was conducted to investigate the possibilities of developing terminal male and female lines for lamb production by the crossing of native breeds, that is, Akkaraman, Sakız (Chios) and Kıvırcık breeds. The Akkaraman sheep is a fattailed breed raised in central Anatolia, while Sakız and Kıvırcık sheep aren non fattailed breeds raised in western and northern-western Anatolia. This investigation was carried out to determine the effect of amitraz poisoning on the the formation of methemoglobin and osmotic fragility of erytrocytes. in this study, 12 guinea-pigs, weighing 250-300 gr were used. They were equally divided into two groups, control and experimental. The animals in the experimental group were intraperitoneally injected with amitraz at a dose of 1 mg/kg body weight. This study was carried out to investigate the prevalance of leptospirosis in the urine of cattle slaughtered in three major abattoirs in the east of Turkey. A polymerase chain reaction (PCR) based on a pair of genus-specific primers was used to detect leptospiral DNA in the urine samples of 473 cattle, 284 of which were from Elazig, 112 from Malatya and 77 from Diyarbakir. The detection limit of the method was determined to be approximately five bacteria per ml of urine.
