The green revolution was based on genetic modification of the gibberellin (GA) hormone system with “dwarfing” gene mutations that reduces GA signals, conferring shorter stature, thus enabling plant adaptation to modern farming conditions. Strong GA-related mutants with shorter stature often have reduced coleoptile length, discounting yield gain due to their unsatisfactory seedling emergence under drought conditions. Here we present gibberellin (GA) 3-oxidase1 (GA3ox1) as an alternative semi-dwarfing gene in barley that combines an optimal reduction in plant height without restricting coleoptile and seedling growth. Using large-scale field trials with an extensive collection of barley accessions, we showed that a natural …

Insights Into Early-Stage Inbreeding Depression In Hawaiian Metrosideros, Rosalie Chaleunsouck
Undergraduate Research Symposium Podium Presentations
Inbreeding depression is the reduced biological fitness of offspring resulting from the conception of two genetically related gametes. Selfing, the fusion of gametes from the same individual, is the most extreme form of inbreeding, common in many plant species. Inbreeding depression is most prevalently seen at the seed-development stage (Levin, 1981).

Context: The legume shrub/tree leucaena (Leucaena leucocephala (Lam.) de Wit.) is highly regarded as a cattle fodder, although it is considered an environmental weed in many parts of Australia.
Aims: We investigated the feasibility of developing a forage variety of triploid leucaena through interspecific hybridisation that is sterile (without seeds) as a strategy to mitigate its weed risk.
Methods: A conventional breeding strategy was used to create triploid hybrids from hand-crosses between tetraploid and diploid species of leucaena. Molecular markers were developed to confirm successful crosses and flow cytometry was used to confirm the ploidy level. The plants are being …

The Philippine Agricultural Scientist
Stevia is a perennial shrub commercially cultivated for the sweet-tasting compounds known as steviol glycosides (SVglys). SVglys are extracted from the green leaves, thus maintenance of vegetative growth is critical in stevia production. This study was conducted to describe the flowering habit and response of potted stevia plants to different photoperiods under Philippine conditions. Photoperiodic treatments (10, 11, 12, 13, and 14 h) were provided using light chambers. Stevia has both terminal and axillary flowering habits. It has five white florets enclosed in a capitulum and exhibits an opposite branching pattern. Results showed that stevia is a facultative short-day plant …

Micronutrient Concentration Effects On Lettuce Growth And Susceptibility To Pythium, Kalyn M. Helms
Micronutrient Concentration Effects On Lettuce Growth And Susceptibility To Pythium, Kalyn M. Helms
Graduate Theses and Dissertations
In hydroponic production waterborne pathogens such as Pythium are ubiquitous and continually threaten a wide range of Controlled Environment Agriculture (CEA) crops in hydroponic production, including but not limited to: lettuce, spinach, basil, arugula, cucumber, tomato, sweet pepper, roses, chrysanthemums, and cannabis (Sutton et al., 2006; Gull, 2002; McGehee and Raudales, 2021; Gillespie, 2020). Despite extensive sanitation measures, disease control in hydroponics is fallible and requires constant surveillance and management to minimize outbreaks (Sutton et al., 2006). A potential disease suppression strategy is to increase micronutrient concentrations within hydroponic systems to naturally strengthen plant defenses against pathogens such as Pythium. …

Identification Of Quantitative Trait Loci (Qtls) For Resistance To Bacterial Leaf Streak: Xanthomonas Translucens Using Qtl And Association Mapping In Three Populations Of Soft Red Winter Wheat, Benjamin Tyler Meritt
LSU Doctoral Dissertations
Bacterial leaf streak (BLS) and black chaff, caused by Xanthomonas translucens pv. undulosa (Xtu), can be a very destructive disease of wheat, especially in the warmer, wetter areas of the Southeastern U.S. Yield losses of up to 40 percent have been recorded in some cases in southern wheat growing regions. With no effective agronomic or chemical method of disease control, identification of genetic resistance is seen as a promising solution. Three soft red winter wheat populations (GAWN, ARK-SNP, and AGS 2060- AGS 2035 DH) representative of soft red winter wheat germplasm in the southeastern U.S. developed by …

Genomics And Transcriptomics To Protect Rice (Oryza Sativa L.) From Abiotic Stressors: -Pathways To Achieving Zero Hunger, Mushtaq Ahmad
Center for Plant Science Innovation: Faculty Publications
More over half of the world’s population depends on rice as a major food crop. Rice (Oryza sativa L.) is vulnerable to abiotic challenges including drought, cold, and salinity since it grown in semi-aquatic, tropical, or subtropical settings. Abiotic stress resistance has bred into rice plants since the earliest rice cultivation techniques. Prior to the discovery of the genome, abiotic stressrelated genes were identified using forward genetic methods, and abiotic stress-tolerant lines have developed using traditional breeding methods. Dynamic transcriptome expression represents the degree of gene expression in a specific cell, tissue, or organ of an individual organism at …

Molecular Pathways Of Wrky Genes In Regulating Plant Salinity Tolerance, Lewis Price, Yong Han, Tefera Angessa, Chengdao Li
Grain and Other Field Crops Research Articles
Salinity is a natural and anthropogenic process that plants overcome using various responses. Salinity imposes a two-phase effect, simplified into the initial osmotic challenges and subsequent salinity-specific ion toxicities from continual exposure to sodium and chloride ions. Plant responses to salinity encompass a complex gene network involving osmotic balance, ion transport, antioxidant response, and hormone signalling pathways typically mediated by transcription factors. One particular transcription factor mega family, WRKY, is a principal regulator of salinity responses. Here, we categorize a collection of known salinity-responding WRKYs and summarize their molecular pathways. WRKYs collectively play a part in regulating osmotic balance, ion …

Background: Access to biologically available nitrogen is a key constraint on plant growth in both natural and agricultural settings. Variation in tolerance to nitrogen deficit stress and productivity in nitrogen limited conditions exists both within and between plant species. However, our understanding of changes in different phenotypes under long term low nitrogen stress and their impact on important agronomic traits, such as yield, is still limited.
Results: Here we quantified variation in the metabolic, physiological, and morphological responses of a sorghum association panel assembled to represent global genetic diversity to long term, nitrogen deficit stress and the relationship …

Arkansas Agricultural Experiment Station Research Series
While the basic growth and development of the cotton plant have not changed significantly in recent history, the business of cotton production is ever-changing. The last two years have seen us plant a crop just about as late as we thought possible, yet extended favorable conditions at season's end have been our salvation, helping to lead us to record yields. The economic environment over the last few years has been such that farmers need to produce record or near-record yields to advance. Unfortunately, production levels at the state yield average barely cover out-of-pocket expenses.
Great uncertainties exist for the upcoming …

Quinoa is being evaluated in cropping systems in many countries outside of its natural range of South America. Very few attempts have been made by farmers or researchers to grow or evaluate quinoa under Australian environments. Given the growing popularity of quinoa with consumers, new commercial opportunities for farmers and international interest in the crop, it was timely to undertake a comprehensive evaluation of the potential of quinoa in Australia. Two advanced selections and nine germplasm lines (six of Chilean and three of Bolivian origin) identified in an earlier project were tested in 23 field trials at 14 locations on …

Divergent selection of populations in contrasting environments leads to functional genomic divergence. However, the genomic architecture underlying heterogeneous genomic differentiation remains poorly understood. Here, we de novo assembled two high-quality wild barley (Hordeum spontaneum K. Koch) genomes and examined genomic differentiation and gene expression patterns under abiotic stress in two populations. These two populations had a shared ancestry and originated in close geographic proximity but experienced different selective pressures due to their contrasting micro-environments. We identified structural variants that may have played significant roles in affecting genes potentially associated with well-differentiated phenotypes such as flowering time and drought response between …

Production of hybrid Phalaenopsis seedlings is generally applied by in vitro culture techniques. The final stage of in vitro culture is acclimatization. The acclimatization stage is crucial because the plantlets must adapt to the ex-vitro environment. Shade supports plantlet growth during the early stages of acclimatization. This study aims to determine the effect of shade level on the physiological and anatomical characteristics of the Phalaenopsis hybrid orchid at the acclimatization stage. The research used Phalaenopsis plantlet hybrid and shading level. Plantlets were shaded at 40%, 55%, and 70%. Parameters measured included: photosynthetic pigment content, number and area of leaves, number …

Stevia (Stevia rebaudiana Bertoni) is a low-calorie alternative sweetener. The superior varieties of Stevia in Indonesia are limited. Availability of seeds plays an important role in the Stevia plant breeding program for both the selection stage and the production of high-yielding varieties. Tissue culture is the best method of Stevia propagation, but the low adaptability of plantlets causes death in the acclimatization phase. The growth and root morphology of 12 crossbreed Stevia were evaluated. A Completely Randomized Design was used with factorial consisting of two factors were Stevia genotypes (STG1,7,8,10, SBG3,4,5,7,10, SGB2,3, and SBT11) and the plant growth regulator (PGR) …

Crosscutting Understandings Of Fusarium Resistance In Sorghum And Wheat For Development Of Breeding Strategies And Novel Opportunities In Germplasm Improvement, Arlyn Ackerman
All Dissertations
Fusarium causes widespread disease in crops resulting in yield and quality loss across a range of host taxa that results in global incidence of carcinogenic mycotoxins contaminating the world’s food systems. This research sought to crosscut frameworks developed across the Fusarium host crop species of wheat (Triticum aesitivum) and sorghum (Sorghum bicolor [L] Moench) to further understand mycotoxin suppression and resistance. A deeper understanding of the biochemical resistances available in sorghum grain mold (SGM) was built by following research of wheat based upon deoxynivalenol resistance to Fusarium head blight (FHB). Through this research, phenotyping methods for Fusarium damaged kernels …

LSU Doctoral Dissertations
Salinity is emerging as a major threat to rice production worldwide due to climate change and anthropogenic sources. We performed three different population development approaches using exotic salt tolerant donors in US rice backgrounds to identify the salt tolerant QTLs/genes and tolerant breeding lines. First approach involved development of a biparental introgression lines (ILs) population by utilizing a salinity tolerant donor ‘TCCP’ in an elite rice cultivar ‘Cheniere’ background and mapping of QTLs at germination, seedling, and reproductive stages A total of seven QTLs for five germination traits, 16 QTLs for seven seedling stage traits, and 30 QTLs for ten …

As a vegetable oil, consisting principally of triacylglycerols, is the major storage form of photosynthetically-fixed carbon in oilseeds which are of significant agricultural and industrial value. Photosynthesis in chlorophyll-containing green seeds, along with photosynthesis in leaves and other green organs, generates ATP and reductant (NADPH and NADH) needed for seed fatty acid production. However, contribution of seed photosynthesis to fatty acid accumulation in seeds have not been well-defined. Here, we report the contribution of seed-photosynthesis to fatty acid production by probing segregating green (photosynthetically-competent) and non-green or yellow (photosynthetically-non-competent) seeds in siliques of an Arabidopsis chlorophyll synthase mutant. Using this …

Pheromones have become an environmentally friendly alternative to conventional insecticides for pest control. Most current pheromone-based pest control products target lepidopteran pests of high-value crops, as today’s manufacturing processes cannot yet produce pheromones at low enough costs to enable their use for lower-value crops, especially commodity crops. Camelina sativa seeds genetically modified to express (Z)-11-hexadecenoic acid, a sex pheromone precursor of several moth species, provided the oil from which the precursor was isolated, purified and transformed into the final pheromone. Trap lures containing this pheromone were then assessed for their capacity to manage moth pests in the field. Plant-derived pheromone …

Modern soybean [Glycine max (L.) Merr] cultivars have low overall genetic variation due to repeated bottleneck events that arose during domestication and from selection strategies typical of many soybean breeding programs. In both public and private soybean breeding programs, the introgression of wild soybean (Glycine soja Siebold and Zucc.) alleles is a viable option to increase genetic diversity and identify new sources for traits of value. The objectives of our study were to examine the genetic architecture responsible for seed protein and oil using a recombinant inbred line (RIL) population derived from hybridizing a G. max …

Tedera is a forage legume that can provide out-of-season green feed in Mediterranean climates. To date, growers have had no comprehensive soil nutrition guidelines to optimise tedera production. We undertook field and glasshouse studies to understand tedera’s macronutrient requirements. Three field experiments were sown with tedera cv. Lanza® at Cunderdin, Dandaragan and Three Springs in Western Australia. These experiments evaluated seven levels of phosphorus (P) (0–30 kg ha−1) and potassium (K) (0–80 kg ha−1) and two combined treatments with P and K. Glasshouse pot experiments were conducted using tedera cultivars Lanza® and Palma and lucerne cultivar SARDI Grazer. Ten concentrations …

Salinity causes a detrimental impact on plant growth, particularly when the stress occurs during germination and early development stages. Barley is one of the most salt-tolerant crops; previously we mapped two quantitative trait loci (QTL) for salinity tolerance during germination on the short arm of chromosome 2H using a CM72/Gairdner doubled haploid (DH) population. Here, we narrowed down the major QTL to a region of 0.341 or 0.439 Mb containing 9 or 24 candidate genes belonging to 6 or 20 functional gene families according to barley reference genomes v1 and v3 respectively, using two DH populations of CM72/Gairdner and Skiff/CM72, …
