Automated Conserved Non-Coding Sequence (Cns) Discovery Reveals Differences In Gene Content And Promoter Evolution Among Grasses, Gina Marie Turco, James C. Schnable, Brent S. Pedersen, Michael Freeling

Conserved non-coding sequences (CNS) are islands of non-coding sequence that, like protein coding exons, show less divergence in sequence between related species than functionless DNA. Several CNSs have been demonstrated experimentally to function as cis-regulatory regions. However, the specific functions of most CNSs remain unknown. Previous searches for CNS in plants have either anchored on exons and only identified nearby sequences or required years of painstaking manual annotation. Here we present an open source tool that can accurately identify CNSs between any two related species with sequenced genomes, including both those immediately adjacent to exons and distal sequences separated by …

Cultivated sorghum [Sorghum bicolor (L.) Moench subsp. bicolor] can interbreed with a feral weedy relative shattercane [S. bicolor nothosubsp. drummondii (Steud.) de Wet ex Davidse]. Traits introduced from cultivated sorghum could contribute to the invasiveness of a shattercane population. An experiment was conducted to determine the potential for pollenmediated gene flow from grain sorghum to shattercane. Shattercane with juicy midrib (dd) was planted in soybean [Glycine max (L.) Merr.] fields during 2 yr in concentric arcs at varying distances from a 0.39 ha sorghum pollen source with dry midrib (DD). The arcs …

Second generation feedstocks for bioethanol will likely include a sizable proportion of perennial C4 grasses, principally in the Panicoideae clade. The Panicoideae contain agronomically important annual grasses including Zea mays L. (maize), Sorghum bicolor (L.) Moench (sorghum), and Saccharum officinarum L. (sugar cane) as well as promising second generation perennial feedstocks including Miscanthus × giganteus and Panicum virgatum L. (switchgrass). The underlying complexity of these polyploid grass genomes is a major limitation for their direct manipulation and thus driving a need for rapidly cycling comparative model. Japanese beetle, Popillia japonica Newman (Coleoptera: Scarabaeidae), is an important quarantine pest of nurseries. Nursery plant movement from P. japonica-infested regions is regulated by the U.S. Domestic Japanese Beetle Harmonization Plan (DJHP), which classifies states by risk categories. Treatments for category 2 states include preharvest soil surface treatment of nursery plants grown in field soil using Discus SC, Marathon (1G or 60 WP), or Flagship (0.22G or 25 WG). Comparing Effects Of Insecticides On Two Green Lacewings Species, Chrysoperla Johnsoni And Chrysoperla Carnea (Neuroptera: Chrysopidae), K. G. Amarasekare, P. W. Shearer

This study compared lethal and sublethal effects of five insecticides, chlorantraniliprole, cyantraniliprole, spinetoram, novaluron, and lambda-cyhalothrin, on adult and second instars of two green lacewing species, Chrysoperla carnea (Stephens) and Chrysoperla johnsoni Henry, Wells and Pupedis (Neuroptera: Chrysopidae) in the laboratory. Formulated pesticides were tested using concentrations equivalent to the high label rate dissolved in 378.5 liters of water. Novaluron and lambda-cyhalothrin were toxic to larvae and no treated larvae survived to the adult stage. Larva to adult survival was reduced in chlorantraniliprole, cyantraniliprole, and spinetoram treatments. Larva to adult developmental time and sex ratio were not different among the …

Golf course superintendents are managing creeping bentgrass (Agrostis stolonifera L.) putting greens throughout the transition zone where temperatures can exceed optimum levels for consecutive days in the summer. The stress of creeping bentgrass associated with these supraoptimal temperatures has been well documented, but the management practices implemented on putting greens to increase green speeds may exacerbate these environmental stresses. To date, the physiological effects of these management practices in combination have not been evaluated for putting green turf. In March 2006, four groundcover management systems (GMS) and two nutrient sources (NS) were implemented for their ability to alter the soil physical condition of a newly established, organically managed apple (Malus x domestica Borkh.) orchard. Annual applications of municipal green compost (GC), shredded office paper (SP), wood chips (WC), and mow-blow (MB) grass mulch were utilized as GMS, and NS supplied to trees were from composted poultry litter (PL), a commercial organic fertilizer (CF), or an untreated control (NF) in a 4x3 factorial study. Suitable orchard land in regions of the Intermountain West is becoming more limited due to urban sprawl. With the loss of suitable farmland, increasing production costs, and the lack of sound fertility information for these regions, fruit growers face challenges to produce high quality fruit for market demand. Current standard management practices are not sufficient to optimize yield and fruit quality in the marginal farm land that is currently be used for fruit production. Fertility management of orchard trees is vital to tree health, yield, and fruit quality.

Dicamba-resistant soybeans are being developed to provide an additional herbicide mechanism-of-action for postemergence weed control in soybean. Numerous broadleaf species, including horseweed, have evolved resistance to glyphosate. It is anticipated that dicamba will be used by farmers as a primary tool to manage these weeds. Studying and understanding variability in horseweed response to dicamba will aid in developing appropriate risk management strategies to extend the utility of the dicamba-resistance technology. Horseweed plants from ten Nebraska populations were treated with one of nine doses of dicamba in greenhouse experiments. At 28 days after treatment (DAT) visual injury estimations were made and …

Laboratory Bioassays To Estimate The Lethal And Sublethal Effects Of Various Insecticides And Fungicides On Deraeocoris Brevis (Hemiptera: Miridae), K. G. Amarasekare, P. W. Shearer

This laboratory bioassay focused on lethal and sublethal effects of five insecticides (chlorantraniliprole, cyantraniliprole, spinetoram, novaluron, and lambda-cyhalothrin) and two fungicide treatments (sulfur and a mixture of copper hydroxide and mancozeb) on the predatory mired bug, Deraeocoris brevis (Uhler) (Hemiptera: Miridae) using second instars and adult males and females. Formulated pesticides were tested using concentrations that were equivalent to the high label rate (1x) (high rate) and 1/10th of that amount (0.1x) (low rate) dissolved in 378.5 liters of water. Lambda-cyhalothrin was highly toxic to D. brevis nymphs and adults at both rates, whereas both rates of novaluron were highly …

This Senior Thesis in Environmental Analysis seeks to explore the ways in which certain food-oriented programs for incarcerated women and women on parole critically resist the Prison Industrial Complex and the Industrial Food System by securing social and ecological equity through the acquisition of food justice. It focuses on three case studies: the Crossroads’ Meatless Mondays program, Fallen Fruit from Rising Women: A Crossroads Social Enterprise, and Cultivating Dreams Prison Garden Project: An Organic Garden for Women in Prison.
