The genus Magnolia includes over 250 species that range in ploidy level from diploid to hexaploid. Although there is basic information on ploidy levels of various species, sampling is limited with specific cultivars and hybrids. The objective of this research was to determine relative genome sizes and relationships to ploidy levels among a diverse collection of species, hybrids, and cultivars using flow cytometry. Nuclei were extracted, stained with 4’, 6–diamidino–2–phenylindole (DAPI), and analyzed using a flow cytometer. Relative genome sizes were determined using Pisum sativum as the reference genome. Rain gardens are increasingly being used as small scale stormwater best management practices (BMPs) to reduce stormwater runoff through infiltration and to remove pollutants through filtration. In 2007, as part of a comprehensive water quality restoration process of Holmes Lake in Lincoln, Nebraska, 20 pilot rain gardens were installed in residential and school properties in the watershed. Currently, assessment and monitoring has been limited to participant surveys and cannot be used to determine if hydraulic or vegetative problems exist within the garden area. The 2010 Rice Research Verification Program ( RRVP) was conducted on twenty-two commercial rice fields across the state. Counties rarticirating in the program included Arkansas, Ashley, Chicot, Clark, Clay, Cross, Desha, Drew, Greene, Jackson, Jefferson, Lafayette, Lawrence, Lonoke, Mississippi, Phillips, Poinsett (2 fields), Prairie, Randolph, St. Francis, and White for a total of 1456 acres. Grain yield in the 2010 RRVP averaged 167 bu/acre ranging from 113 to 215 bu/acre. The 2010 RRVP average yield was 25 bu/acre greater than the estimated Arkansas state average of 142 bu/acre. Plants propagated from vegetative cuttings have become an increasingly important market in the United States. Significant economic losses occur annually due to poor performance, damage or death of cuttings. This occurs in large part due to the industry's lack of knowledge of the proper postharvest environments to provide to cuttings. This project was conducted to improve the understanding of the postharvest physiology of unrooted cuttings in order to optimize performance and longevity. Identification Of Salt-Induced Changes In Leaf And Root Proteomes Of The Wild Tomato, Solanum Chilense, Suping Zhou, Roger J. Sauvé, Zong Liu, Sasikiran Reddy, Sarabjit Bhatti, Simon D. Hucko, Tara Fish, Theodore W. Thannhauser

This article reports salt-induced changes in leaf and root proteomes after wild tomato (Solanum chilense) plants were treated with 200 mmNaCl. In leaf tissues, a total of 176 protein spots showed significant changes (P < 0.05), of which 104 spots were induced and 72 spots suppressed. Salt-induced proteins are associated with the following pathways: photosynthesis, carbohydrate metabolism, glyoxylate shunt, glycine cleavage system, branched-chain amino acid biosynthesis, protein folding, defense and cellular protection, signal transduction, ion transport, and antioxidant activities. Suppressed proteins belong to the following categories: oxidative phosphorylation pathway, photorespiration and protein translational machinery, oxidative stress, and ATPases. In root tissues, 106 protein spots changed significantly (P< 0.05) after the salt treatment, 63 spots were induced, and 43 suppressed by salt treatment. Salt-induced proteins are associated with the following functional pathways: regeneration of S-adenosyl methionine, protein folding, selective ion transport, antioxidants and defense mechanism, signal transduction and gene expression regulation, and branched-chain amino acid synthesis. Salt-suppressed proteins are receptor kinase proteins, peroxidases and germin-like proteins, malate dehydrogenase, and glycine dehydrogenase. The False Root-Knot nematode Nacobbus aberrans is a plant parasitic nematode that causes yield losses in several crops and plant protection agencies have established regulations to prevent infestations from spreading to new agricultural areas. The taxonomic status of the nematode has been the subject of controversy due to wide degree of variation exhibited by the species. This variability has led to the suggestion that N. aberrans is actually a species complex rather than a single species. The booklet was designed to be helpful to both high school students and high school instructors. All students enrolled in agriculture programs have projects known as SAE’s. This booklet includes a variety of topics that are relevant to having an SAE project. All information was based on industry statistics and information gained from courses. Three tomato (Solanum lycopersicum) cultivars [Walter LA3465 (heat-tolerant), Edkawi LA 2711 (unknown heat tolerance, salt-tolerant), and LA1310 (cherry tomato)] were compared for changes in leaf proteomes after heat treatment. Seedlings with four fully expanded leaves were subjected to heat treatment of 39/25 °C at a 16:8 h light–dark cycle for 7 days. Leaves were collected at 1200 hr, 4 h after the light cycle started. For ‘Walter’ LA3465, heat-suppressed proteins were geranylgeranyl reductase, ferredoxin-NADP (+) reductase, Rubisco activase, transketolase, phosphoglycerate kinase precursor, fructose–bisphosphate aldolase, glyoxisomal malate dehydrogenase, catalase, S-adenosyl-L-homocysteine hydrolase, and methionine synthase. Native plants are playing an increasing role in sustainable landscapes that use fewer resources such as water and fertilizer. Unfortunately, many native plants are not available in the nursery trade, or if they are available they may only be found as seedling plants grown for the reclamation industry. Such plants are high quality and have a valuable role in the reclamation of disturbed sites such as fire-damaged areas. Fuel-efficient poinsettia production has become an important topic in recent years due to increased fuel costs while wholesale prices have remained relatively unchanged. Fuel-efficient production results in energy saving, reduces costs associated with production and improves profitability of the poinsettia crop. Experiments were carried out to determine the effects of Fascination (a plant growth regulator), production temperature, light, and cold pre-harvest temperature on bract expansion, cyathia development, and retention for fuel-efficient poinsettia production.

Murashige and Skoog 1962 performed one-factor-at-a-time (OFAT) experimentation in order to regenerate tobacco callus on semi-solid agar medium. This work became an established medium for tissue culture micropropagation and experimentation. Micropropagation is done in niche markets with herbaceous perennials, among other crops, and the optimization of various inputs to produce maximal responses is a necessary step towards process development. This current study of macronutrient factors simultaneously altered media volume, amount of tissue (plants per vessel), sucrose, nitrogen (as NO3- and NH4+ ions), and K+ in a d-optimal design space with only 55 experimental units (including 5 true replicates). Background:

It is difficult to accurately interpret chromosomal correspondences such as true orthology and paralogy due to significant divergence of genomes from a common ancestor. Analyses are particularly problematic among lineages that have repeatedly experienced whole genome duplication (WGD) events. To compare multiple “subgenomes” derived from genome duplications, we need to relax the traditional requirements of “one-to-one” syntenic matchings of genomic regions in order to reflect “one-to-many” or more generally “many-to-many” matchings. However this relaxation may result in the identification of synteny blocks that are derived from ancient shared WGDs that are not of interest. For many downstream analyses, we …

The nursery and greenhouse industry, distributed throughout Kentucky, is diverse and contributes significantly to the local and state economy. This industry represents the largest segment of Kentucky’s sales of horticultural products and is comprised almost completely of farm families. Nursery and greenhouse crops are produced and marketed in containers or grown in the field and dug and sold as balled and burlapped or bare root plants. Most of Kentucky’s nursery and greenhouse crops are marketed in Kentucky and surrounding states.

This research has revealed significant chemopreventive potential belonging to the planet’s most renewable and abundant plant source: amenity grasses.  The characterization results from Chapter 1 demonstrated the potential chemoprevention attributes of supina bluegrass and bermudagrass crude extracts linked to the extensive phenolic acid profiles of these amenity grasses.  In contrast to cereals, amenity grass phenolic extracts (AGPE) offer appreciable amounts of unbound/free ferulic acid that would be available for rapid in vivo absorption.  The literature has shown ferulic acid to be a highly available and health benefitting phenolic compound.  Chapter 1 in vitro studies demonstrated AGPE-induced antiproliferation and apoptotic induction …

Knowledge of genetic diversity, population structure, and degree of linkage disequilibrium (LD) in target association mapping populations is of great importance and is a prerequisite for LD-based mapping. In the present study, 96 genotypes comprising 92 accessions of the US peanut minicore collection, a component line of the tetraploid variety Florunner, diploid progenitors A. duranensis (AA) and A. ipaënsis (BB), and synthetic amphidiploid accession TxAG-6 were investigated with 392 simple sequence repeat (SSR) marker bands amplified using 32 highly-polymorphic SSR primer pairs. Not all genes are created equal. Despite being supported by sequence conservation and expression data, knockout homozygotes of many genes show no visible effects, at least under laboratory conditions. We have identified a set of maize (Zea mays L.) genes which have been the subject of a disproportionate share of publications recorded at MaizeGDB. We manually anchored these ‘‘classical’’ maize genes to gene models in the B73 reference genome, and identified syntenic orthologs in other grass genomes. Whole genome duplications, or tetraploidies, are an important source of increased gene content. Following whole genome duplication, duplicate copies of many genes are lost from the genome. This loss of genes is biased both in the classes of genes deleted and the subgenome from which they are lost. Many or all classes are genes preferentially retained as duplicate copies are engaged in dose sensitive protein–protein interactions, such that deletion of any one duplicate upsets the status quo of subunit concentrations, and presumably lowers fitness as a result. Transcription factors are also preferentially retained following every whole genome duplications studied. This …

It is intended to increase the resources available to the public to incorporate water efficient principles and plants into the urban landscape.
