Chemical genetics is increasingly being employed to decode traits in plants that may be recalcitrant to traditional genetics due to gene redundancy or lethality. However, the probability of a synthetic small molecule being bioactive is low; therefore, thousands of molecules must be tested in order to find those of interest. Liquid handling robotics systems are designed to handle large numbers of samples, increasing the speed with which a chemical library can be screened in addition to minimizing/standardizing error. Seeds employ sensory systems that assess various environmental cues over time to maximize the successful transition from embryo to seedling. Here we show that the Arabidopsis F-BOX protein COLD TEMPERATURE-GERMINATING (CTG)-10, identified by activation tagging, is a positive regulator of this process. When overexpressed (OE), CTG10 hastens aspects of seed germination. CTG10 is expressed predominantly in the hypocotyl, and the protein is localized to the nucleus. CTG10 interacts with PHYTOCHROME-INTERACTING FACTOR 1 (PIF1) and helps regulate its abundance in planta. CTG10-OE accelerates the loss of PIF1 in light, increasing germination efficiency, while PIF1-OE lines fail to complete germination in …

This research was conducted to determine the effect of concentration of Ethyl Methyl Sulfonate (EMS) to performance of three types of Phalaenopsis Planlets and to know the optimum concentration of EMS on any type of Phalaenopsis orchids. Background
De novo heterozygous assembly is an ongoing challenge requiring improved assembly approaches. In this study, three strategies were used to develop de novo Vitis vinifera ‘Sultanina’ genome assemblies for comparison with the inbred V. vinifera (PN40024 12X.v2) reference genome and a published Sultanina ALLPATHS-LG assembly (AP). The strategies were: 1) a default PLATANUS assembly (PLAT_d) for direct comparison with AP assembly, 2) an iterative merging strategy using METASSEMBLER to combine PLAT_d and AP assemblies (MERGE) and 3) PLATANUS parameter modifications plus GapCloser (PLAT*_GC).
Results
The three new assemblies were greater in size than the AP assembly. PLAT*_GC had the …

Several crops were evaluated in high tunnels or hoophouses. Most trials evaluated different cultivars or varieties. One report addressed specific growing practices: plant spacing for sweet corn.

Pumpkins are grown on 5,000 acres in Indiana with an average yield of 6.75 tons per acre. Successful pumpkin production requires the use of varieties that yield well and produce pumpkins of the size, shape, color, and quality demanded by the market. Genetic resistance to the fungal disease powdery mildew is present in some varieties. This trial was designed to evaluate performance of pumpkin varieties in northern Indiana with and without treatment for powdery mildew. The trial included fourteen orange jack-o-lantern size pumpkins, one small or 'pie' pumpkin and one specialty yellow pumpkin.

Obtaining corn hybrid seeds (Zea mays L.) with high vigour depends on the parental lines and the direction of the cross, and this relates to seed desiccation tolerance and composition. This research studied reciprocal crosses between pairs of proprietary, elite parent lines (L1 and L5; L2 and L4) producing hybrid seeds with different qualities attempting to correlate vigour with seed composition, focusing on storage proteins, starch and soluble sugar amounts. In 2013, Castro et al., produced the first linkage map for primocane-fruiting blackberries using the cross ‘Prime-Jim®’ x ‘Arapaho.’ This mapping population has been maintained since the original experiment with the hopes of conducting future studies. Further research was determined to be needed on the population to better characterize additional phenotypic traits, and since nine years had passed since the seedlings were established, there was a need to assess the continued genetic purity of the population using molecular methods. Cultivated strawberry, Fragaria x ananassa Duchesne, is host for many pathogens. One of the most destructive diseases of strawberry is anthracnose, whose symptoms include fruit rot, leaf and petiole lesions, crown rot, wilt, and death. Three species of Colletotrichum are considered causative agents of anthracnose diseases of strawberry. Colletotrichum acutatum causes anthracnose fruit rot, has a broad host range, and occurs in most areas of the world where strawberries are grown. Colletotrichum fragariae, the primary causal fungus of anthracnose crown rot, may infect all aboveground parts of the strawberry plant, and has restricted host and geographic ranges. Plants have an intriguing tripartite genetic system: Nuclear genome 9 Mitochondria 9 Plastids and their interactions may impact germplasm breeding. In grapevine, the study of cytoplasmic genomes has been limited, and their role with respect to grapevine germplasm diversity has yet to be elucidated. In the present study, the results of an analysis of the cytoplasmic diversity among 6073 individuals (comprising cultivars, interspecific hybrids and segregating progenies) are presented. Genotyping by sequencing (GBS) was used to elucidate plastid and mitochondrial DNA sequences, and results were analyzed using multivariate techniques. Midwest Vegetable Trial Reports
This is a compilation of 19 research trial reports from six land-grant universities in the midwestern United States. Crops include bok choy, cantaloupe, cucumber, pepper, pumpkin, sweet corn, squash, tomato, and watermelon. Several crops were evaluated in high tunnels or hoophouses. Most trials evaluated different cultivars or varieties, including one investigating cantaloupe variety influence on cucumber beetle presence and incidence of bacterial wilt. Trials on mini sweet peppers and other specialty sweet peppers in the Midwest have not recently been reported. We undertook this project to compare yield and fruit characteristics of hybrid and heirloom peppers grown in high tunnels using organic and conventional production methods.
