Background:

The homeodomain leucine zipper (HD-Zip) transcription factor family is one of the largest plant specific superfamilies, and includes genes with roles in modulation of plant growth and response to environmental stresses. Many HD-Zip genes are characterized in Arabidopsis (Arabidopsis thaliana), and members of the family are being investigated for abiotic stress responses in rice (Oryza sativa), maize (Zea mays), poplar (Populus trichocarpa) and cucumber (Cucmis sativus). Findings in these species suggest HD-Zip genes as high priority candidates for crop improvement.

Both rainfed and irrigated soybean production are important in Nebraska (western US Corn Belt), accounting for a respective 48 and 52% of the state’s soybean production of 7 Mt on a respective 55 and 45% share of the state soybean area of 1.9 Mha. To date, no assessment of factors that may account for regional and inter-annual variation in yield and irrigation amount has been performed. To accomplish that objective, we evaluated a database containing on-farm field yields and total irrigation amount used in those fields. Soybean seeds contain high levels of oil and protein, and are the important sources of vegetable oil and plant protein for human consumption and livestock feed. Increased seed yield, oil and protein contents are the main objectives of soybean breeding. Resistance to SCN is complex, polygenic, race and cultivar specific, and it is controlled by several quantitative trait loci (QTL). In soybean, nitrogen fixation is more sensitive to drought than other physiological processes like photosynthesis. The sensitivity of nitrogen fixation to drought has been associated with high shoot concentrations of ureide and nitrogen under well-watered conditions. Previous research by Hwang et al. (2013) detected quantitative trait loci (QTLs) in a KS4895 by Jackson population associated with shoot ureide and nitrogen concentrations. The present research evaluated the use of these QTLs in selecting genotypes with drought tolerant nitrogen fixation. Salinity is a common abiotic stress causing soybean yield loss worldwide. Use of tolerant cultivars is an effective and economic approach to coping with this stress. Toward this, research is needed to identify salt-tolerant germplasm and better understand the genetic and molecular basis of salt tolerance in soybean. The objectives of this study were to identify salt-tolerant genotypes, to search for SNPs and QTLs associated with salt tolerance, and to identify candidate salt tolerance genes. Growers are constantly seeking ways to improve yield in soybean [Glycine max (L.) Merr.]. There has been much interest in the use of selected alternative practices to maximize soybean yield. The soybean genome duplicated ~14 and 45 million years ago and has many paralogous genes, including those in urease activation (emplacement of Ni and CO2 in the active site). Activation requires the UreD and UreF proteins, each encoded by two paralogues. UreG, a third essential activation protein, is encoded by the single-copy Eu3, and eu3 mutants lack activity of both urease isozymes. eu2 has the same urease-negative phenotype, consistent with Eu2 being a single-copy gene, possibly encoding a Ni carrier. Increasing atmospheric CO2 concentration could increase crop productivity and alter crop nutrient dynamics. This study was conducted (3 yrs) with two crops ([Glycine max (L.) Merr.] and grain sorghum [Sorghum bicolor (L.) Moench.]) grown under two CO2 levels (ambient and twice ambient) using open top field chambers on a Blanton loamy sand under no-tillage. Macronutrient and micronutrient concentrations and contents were determined for grain, stover, and roots. Although elevated CO2 tended to reduce nutrient concentrations, high CO2 consistently increased nutrient content especially in grain tissue; this response pattern was more notable with macronutrients. Stem borers are the most important maize pests in the humid forest zone of Cameroon. Field trials were conducted in the long and short rainy seasons of 2002 and 2003 to assess the level of damage and yield reductions caused by stem borers in monocropped maize and in maize intercropped with non-host plants such as cassava, cowpea and soybean. The intercrops were planted in two spatial arrangements, i.e. alternating hills or alternating rows. A greenhouse study was conducted to determine the effects of irrigation waters differing in salt composition on growth characteristics, salt ion and selenium (Se) accumulation, and distribution in plant components of the soybean (Glycine max L. Merr.) cultivar “Manokin.” Plants were grown in sand cultures and irrigated with isoosmotic solutions containing (1) Cl as the dominant anion, or (2) a mixture of salts containing equal molar amounts of SO2-4  and Cl. The conversion of AFLP bands into polymorphic sequence-tagged-site (STS) markers is necessary for high-throughput genotype scoring. Technical hurdles that must be overcome arise from genome complexity (particularly sequence duplication), from the low-molecular- weight nature of the AFLP bands and from the location of the polymorphism within the AFLP band. We generated six STS markers from ten AFLP bands (four AFLPs were from co-dominant pairs of bands) in soybean (Glycine max). Large DNA insert libraries in binary T-DNA vectors can assist in the isolation of the gene(s) under-lying a quantitative trait locus (QTL). Binary vectors facilitate the transfer of large-insert DNA fragments containing a QTL from E. coli to Agrobacterium sp. and then to plants. We constructed two soybean large-insert libraries from cv. Forrest in the pCLD04541 (V41) binary vector after partial digestion of genomic high-molecular-weight DNA with BamHI or HindIII. The libraries contain 76,800 clones with an average insert size of 125 kb, and therefore represent 9.5-fold haploid genome equivalents.
