Beyond The Fat: Protein Metabolism And Muscle Function As Part Of The Migratory Syndrome Of Songbirds, Cory R. Elowe

Migration allows animals to seasonally exploit favorable habitats that are geographically disparate, and migratory animals have a suite of adaptations to complete their extraordinary journeys. This “migratory syndrome” of traits is comprised of seasonally-responsive adaptations that together result in changes to metabolism, body composition, and reproductive status, all of which act to increase the probability of a successful migration. The focus of my dissertation is to investigate the suite of changes in metabolism and muscle function that enable birds to undertake long-duration flights. Aging in skeletal muscle is characterized by a loss in muscular performance. This is in part related to the direct loss of muscle mass due to senescence, known as sarcopenia. With age, skeletal muscles lose force production, contractile speed, and power production. The force velocity relationship of muscle is a product of force production and contraction speed, both of which decline with age; however, the mechanisms and trajectory of this decline are not well understood. Salt inducible kinase 1 (SIK1) has been considered a stress-inducible kinase since it was first cloned in 1999. Continued efforts since this time have been dedicated to characterizing the structure and function of SIK1. Such research has laid the ground work for our understanding of SIK1 action and regulation in tissue and stimuli dependent manners. The fundamental findings of this dissertation continue in this tradition and include investigations of SIK1 regulatory mechanisms in skeletal muscle cells, the cellular and physiological effects of SIK1 loss of function in vitro and in vivo, and intracellular metabolic and mitochondrial regulation by this …

Myogenin Modulates Exercise Endurance By Altering Skeletal Muscle Metabolism, James Eric Meadows

The function of myogenic regulatory factors (MRFs) during adult life is not well understood. The requirement of one of these MRFs, myogenin (Myog), during embryonic muscle development suggests an equally important role in adult muscle. In this study, we have determined the function of myogenin during adult life using a conditional allele of Myog. In contrast to embryonic development, myogenin is not required for adult viability, and Myog-deleted mice exhibited no remarkable phenotypic changes during sedentary life. Remarkably, sedentary Myog-deleted mice demonstrated enhanced exercise endurance during involuntary treadmill running. A Motor And A Brake: Two Leg Extensor Muscles Acting At The Same Joint Manage Energy Differently In A Running Insect, Anna N. Ahn, Robert J. Full

The individual muscles of a multiple muscle group at a given joint are often assumed to function synergistically to share the load during locomotion. We examined two leg extensors of a running cockroach to test the hypothesis that leg muscles within an anatomical muscle group necessarily manage (i.e. produce, store, transmit or absorb) energy similarly during running. Using electromyographic and video motion-analysis techniques, we determined that muscles 177c and 179 are both active during the first half of the stance period during muscle shortening. Using the in vivo strain and stimulation patterns determined during running, we measured muscle power output. …
