An Extractive Nanoelectrospray Ionization-Mass Spectrometry Method For Chinese Herbal Medicine Authentication, Chia-Hsin Lee, Hui-Chi Huang, Mei-Chun Tseng, Chao-Jung Chen

Herein, we describe a rapid, sensitive, and nondestructive method—extractive nanoelectrospray ionization-mass spectrometry (EnESI-MS)—for traditional Chinese medicine (TCM) authentication. The mass-spectral fingerprints of volatile compounds released from various TCMs can be rapidly acquired using EnESI-MS without sample pretreatment. EnESI-MS was applied to successfully differentiate between two commonly used medicinal herbs, Schisandra Chinensis and Schisandra sphenanthera, which are morphologically similar but exhibit different therapeutic effects. Specific volatile compounds of each herb in a ten-component Chinese herbal product, Jia Wei Xiao Yao San, were also identified, and the method was applied to discriminate between the commercial product and a substandard version.

L-theanine involves a great number of health benefits and dietary supplements containing this molecule are becoming increasingly popular. There is, therefore, a growing need to find ways to discriminate between natural L-theanine extracted from tea leaves and the cheaper, synthetic one obtained using specific bacterial enzymes. A first attempt of stable isotope ratio analysis characterization of the possible synthetic adulterant L-theanine (δ13C of -14.3±1.5‰), obtained from vegetable substrates with C4 photosynthetic cycle, and of the more expensive natural L-theanine (δ13C of -24.4±1.3‰), extracted from Camellia sinensis plants with C3 photosynthetic cycle, is reported here .

In this work, the stable isotope ratios of carbon, nitrogen, hydrogen, oxygen, and mineral elements and their stoichiometric methods were examined as possible factors that could certify Chinese tea based on its production years. A total of 43 multi-element stable isotope ratios of Xiangzhujing Pu'er tea in five production years were determined through inductively coupled plasma mass spectrometry (ICP-MS) and elemental analyzer-isotope ratio mass spectrometry (EA-IRMS) methods.
