Anti-Nlrp3 Inflammasome Natural Compounds: An Update, Baolong Liu, Jiujiu Yu
Anti-Nlrp3 Inflammasome Natural Compounds: An Update, Baolong Liu, Jiujiu Yu
Department of Nutrition and Health Sciences: Faculty Publications
The nucleotide-binding domain and leucine-rich repeat related (NLR) family, pyrin domain containing 3 (NLRP3) inflammasome is a multimeric protein complex that recognizes various danger or stress signals from pathogens, the host, and the environment, leading to activation of caspase-1 and inducing inflammatory responses. This pro-inflammatory protein complex plays critical roles in pathogenesis of a wide range of diseases including neurodegenerative diseases, autoinflammatory diseases, and metabolic disorders. Therefore, intensive efforts have been devoted to understanding its activation mechanisms and to searching for its specific inhibitors. Approximately forty natural compounds with anti-NLRP3 inflammasome properties have been identified. Here, we provide an update …

Honey has been used as a nutrient, an ointment, and a medicine worldwide for many centuries. Modern research has demonstrated that honey has many medicinal properties, reflected in its anti-microbial, anti-oxidant, and anti-inflammatory bioactivities. Honey is composed of sugars, water and a myriad of minor components, including minerals, vitamins, proteins and polyphenols. Here, we report a new bioactive component‒vesicle-like nanoparticles‒in honey (H-VLNs). These HVLNs are membrane-bound nano-scale particles that contain lipids, proteins and small-sized RNAs. The presence of plant-originated plasma transmembrane proteins and plasma membrane-associated proteins suggests the potential vesicle-like nature of these particles. H-VLNs impede the formation and activation …

Zika virus (ZIKV) infection to a pregnant woman can be vertically transmitted to the fetus via the placenta leading to Congenital Zika syndrome. This is characterized by microcephaly, retinal defects, and intrauterine growth retardation. ZIKV induces placental trophoblast apoptosis leading to severe abnormalities in the growth and development of the fetus. However, the molecular mechanism behind ZIKV-induced apoptosis in placental trophoblasts remains unclear. We hypothesize that ZIKV infection induces endoplasmic reticulum (ER) stress in the trophoblasts, and sustained ER stress results in apoptosis. HTR-8 (HTR-8/SVneo), a human normal immortalized trophoblast cell and human choriocarcinoma-derived cell lines (JEG-3 and JAR) were …

Although exercise modulates appetite regulation and food intake, it remains poorly understood how exercise impacts decision-making about food. The purpose of the present study was to assess the impact of an acute exercise bout on hypothetical choices related to the amount and timing of food intake. Forty-one healthy participants (22.0 ± 2.6 years; 23.7 ± 2.5 kg/m2 , 56% female) completed 45 min of aerobic exercise and a resting control condition in randomized order. Food amount preferences and intertemporal food preferences (preference for immediate vs. delayed consumption) were assessed using electronic questionnaires with visual food cues. Compared to rest, …

Applications Of The Critical Power Model To Dynamic Constant External Resistance Exercise: A Brief Review Of The Critical Load Test, Haley C. Bergstrom, Taylor K. Dinyer, Pasquale J. Succi, Caleb C. Voskuil, Terry J. Housh
Department of Nutrition and Health Sciences: Faculty Publications
The study and application of the critical power (CP) concept has spanned many decades. The CP test provides estimates of two distinct parameters, CP and W0 , that describe aerobic and anaerobic metabolic capacities, respectively. Various mathematical models have been used to estimate the CP and W0 parameters across exercise modalities. Recently, the CP model has been applied to dynamic constant external resistance (DCER) exercises. The same hyperbolic relationship that has been established across various continuous, whole-body, dynamic movements has also been demonstrated for upper-, lower-, and whole-body DCER exercises. The asymptote of the load versus repetition relationship is defined …

Department of Nutrition and Health Sciences: Faculty Publications
Whole red raspberry polyphenols (RRW), including ellagic acid, and their gut-derived metabolite, urolithin A (UroA), attenuate inflammation and confer health benefits. Although results from recent studies indicate that polyphenols and UroA also provide neuroprotective effects, these compounds differ in their bioavailability and may, therefore, have unique effects on limiting neuroinflammation. Accordingly, we aimed to compare the neuroprotective effects of RRW and UroA on BV-2 microglia under both 3 h and 12 and 24 h inflammatory conditions. In inflammation induced by lipopolysaccharide (LPS) and ATP stimulation after 3 h, RRW and UroA suppressed pro- inflammatory cytokine gene expression and regulated the …

Covid-19 Contagion Among Communities With Limited English Proficiency: Lesson From Volunteerism, Falah N. Rashoka M.S
Department of Nutrition and Health Sciences: Faculty Publications
The COVID-19 affects all communities in the United States, but its impact differs from one community to another. Undoubtedly, the vulnerable populations are more impacted than others, especially refugees and immigrants. This study seeks to understand the impact of COVID-19 on marginalized communities and the need to communicate health-related information equally. A lesson learned from observation during an ongoing pandemic, including what I learned through my volunteer work between mid-March and August 2020. The study finds that neighbors with limited English Proficiency (LEP) and limited education could quickly transfer the COVID-19 virus to another neighbor with high income and well-educated. …

Iron is an essential biometal, but is toxic if it exists in excess. Therefore, iron content is tightly regulated at cellular and systemic levels to meet metabolic demands but to avoid toxicity. We have recently reported that adaptive thermogenesis, a critical metabolic pathway to maintain whole-body energy homeostasis, is an irondemanding process for rapid biogenesis of mitochondria. However, little information is available on iron mobilization from storage sites to thermogenic fat. This study aimed to determine the iron-regulatory network that underlies beige adipogenesis. We hypothesized that thermogenic stimulus initiates the signaling interplay between adipocyte iron demands and systemic iron liberation, …

Department of Nutrition and Health Sciences: Faculty Publications
There is a critical need for research to examine factors that hinder or facilitate access to healthcare in the context of COVID-19. The coronavirus pandemic has exacerbated barriers to healthcare for marginalized communities globally, leaving many without access to needed health services. Resettled refugees, in particular, must grapple with added challenges to locating and obtaining culturally appropriate healthcare services.
Recent focus groups shed light on the experiences of Yazidi refugees in the U.S. Midwest during COVID-19. Yazidis are a historically persecuted religious and ethnic minority group from northern Iraq (Kizilhan, 2017). Many Yazidis migrated to the U.S. on Special Immigrant …

Cancer cachexia is a metabolic syndrome with alterations in gene regulatory networks that consequently lead to skeletal muscle wasting. Integrating microRNAs mRNAs omics profiles offers an opportunity to understand transcriptional and post-transcriptional regulatory networks underlying muscle wasting. Here, we used RNA sequencing to simultaneously integrate and explore microRNAs and mRNAs expression profiles in the tibialis anterior (TA) muscles of the Lewis Lung Carcinoma (LLC) model of cancer cachexia. We found 1,008 mRNAs and 18 microRNAs differentially expressed in cachectic mice compared with controls. Although our transcriptomic analysis demonstrated a high heterogeneity in mRNA profiles of cachectic mice, we identified a …

Purpose Weight loss can result in the loss of muscle mass and bone mineral density. Resistance exercise is commonly prescribed to attenuate these effects. However, the anabolic endocrine response to resistance exercise during caloric restriction has not been characterized. Methods Participants underwent 3-day conditions of caloric restriction (15 kcal kg FFM−1) with post-exercise carbohydrate (CRC) and with post-exercise protein (CRP), and an energy balance control (40 kcal kg FFM−1) with post-exercise carbohydrate (CON). Serial blood draws were taken following five sets of five repetitions of the barbell back squat exercise on day 3 of each condition. …

Fulminant hepatic failure (FHF) is a rare, life-threatening liver disease with a poor prognosis. Administration of D-galactosamine (GalN) and lipopolysaccharide (LPS) triggers acute liver injury in mice, simulating many clinical features of FHF in humans; therefore, this disease model is often used to investigate potential therapeutic interventions to treat FHF. Recently, suppression of the nucleotide-binding domain and leucine-rich repeat related (NLR) family, pyrin domain containing 3 (NLRP3) inflammasome, was shown to alleviate the severity of GalN/LPS-induced liver damage in mice. Therefore, the goal of this study was to find dietary exosome-like nanoparticles (ELNs) with therapeutic potential in curbing FHF by …

Background: Nut butter-based Ready to Use Supplemental Foods (RUSF) are an effective way to add nutrients and calories to diets of malnourished and food insecure populations. The RUSF formulations have been further modified to add micronutrients including iron and folic acid needed during pregnancy and lactation. Because docosahexaenoic acid (DHA, C22:6 n-3) enhances fetal development and birth outcomes, it has been suggested that perhaps RUSF formulations for pregnancy should also include this Omega 3 fatty acid. The goal of the present study was to gain an understanding of Zambian women’s knowledge of nutritional needs in pregnancy through structured focus group …

Supplementation with n-3 long-chain (LC) polyunsaturated fatty acids (PUFA) is known to promote thermogenesis via the activation of brown adipose tissue (BAT). Agricultural products that are biofortified with α-linolenic acid (ALA), the precursor of n-3 LC PUFA, have been launched to the market, but their impact on BAT function is unknown. This study aimed to evaluate the effects of ALA-biofortified butter on lipid metabolism and thermogenic functions in the BAT. C57BL/6 mice were fed a high-fat diet containing ALA-biofortified butter (n3Bu, 45% calorie from fat) for ten weeks in comparison with the isocaloric high-fat diets prepared from conventional butter or …

Emerging evidence suggests intestinal microbiota as a central contributing factor to the pathogenesis of Relapsing-Remitting-Multiple-Sclerosis (RRMS). This novel RRMS study evaluated the impact of fecal-microbiota-transplantation (FMT) on a broad array of physiological/clinical outcomes using deep metagenome sequencing of fecal microbiome. FMT interventions were associated with increased abundances of putative beneficial stool bacteria and short-chain-fatty-acid metabolites, which were associated with increased/improved serum brain-derived-neurotrophic-factor levels and gait/walking metrics. This proof-of-concept single-subject longitudinal study provides evidence of potential importance of intestinal microbiota in the pathogenesis of MS, and scientific rationale to help design future randomized controlled trials assessing FMT in RRMS patients.

R-Α-Lipoic Acid And 4-Phenylbutyric Acid Have Distinct Hypolipidemic Mechanisms In Hepatic Cells, Bo He, Régis Moreau
Department of Nutrition and Health Sciences: Faculty Publications
The constitutive activation of the mechanistic target of rapamycin complex 1 (mTORC1) leads to the overproduction of apoB-containing triacylglycerol-rich lipoproteins in HepG2 cells. R-α-lipoic acid (LA) and 4-phenylbutyric acid (PBA) have hypolipidemic function but their mechanisms of action are not well understood. Here, we reported that LA and PBA regulate hepatocellular lipid metabolism via distinct mechanisms. The use of SQ22536, an inhibitor of adenylyl cyclase, revealed cAMP’s involvement in the upregulation of CPT1A expression by LA but not by PBA. LA decreased the secretion of proprotein convertase subtilisin/kexin type 9 (PCSK9) in the culture media of hepatic cells and increased …

Urban Food Sovereignty: Urgent Need For Agroecology And Systems Thinking In A Post-Covid-19 Future, Ali Loker, Charles A. Francis
Department of Agronomy and Horticulture: Faculty Publications
The current COVID-19 pandemic has brought attention to challenges associated with our dominant industrial food system in the U.S. The general public now has more appreciation for farm workers and meatpacking employees, as well as those in grocery stores and in food transportation who are suddenly recognized as essential frontline workers. It apparently takes a crisis for us to focus clearly on the fragility of this system and the lives of people on whom we depend. In this commentary we discuss the definition of food sovereignty, how it manifests in urban areas, and how the COVID-19 pandemic can trigger viable …

Purpose: The purposes of the present study were: (1) to determine whether the physical working capacity at the fatigue threshold (PWCFT) model that has been used for estimating the onset of neuromuscular fatigue in the vastus lateralis (VL) during incremental treadmill running could also be applied to the vastus medialis (VM), biceps femoris (BF), and semitendinosus (ST) muscles; and (2) if applicable, to compare the running velocities associated with the PWCFT among these muscles.
Methods: Eleven subjects (age 21.7 ± 1.8 years) performed an incremental treadmill test to exhaustion with electromyographic signals recorded from the …

Background: Few studies have compared performance fatigability (PF) for bilateral versus unilateral isokinetic tasks. Objectives: The purpose of this study was to examine: Mode- specific testing responses to isokinetic fatigue, differences in PF between bilateral and unilateral leg extensions, and the effects of fatiguing, unilateral, dynamic leg extensions on contralateral isokinetic peak torque (PT) and maximal voluntary isometric contraction (MVIC). Methods: Eight men (mean ± SD: age= 22.5 ± 2.5 yr.) completed pre- and post-testing for PT and MVIC following 50 bilateral, unilateral right or left leg maximal, isokinetic leg extensions at 180°·s-1, on three separate days. Fatigue-induced decreases in …

Background/Objectives—γ-Tocopherol has unique properties that protect against nitrogen oxide-mediated cellular damage. To elucidate the potential role of γ-tocopherol in the aging process, we examined the associations of serum γ-tocopherol levels with all-cause and cause-specific mortality.
Subjects/Methods—Among participants in the biorepository subcohort of the Multiethnic Cohort Study, pre-cancer diagnostic serum γ-tocopherol levels were measured in a subset of 3904 men and 4461 women. Of these, 22.7% of men and 13.5% of women died during a mean follow- up time of 9.6±2.6 years. Hazard ratios (HRs) and 95% confidence intervals (95%CIs) for mortality associated with γ-tocopherol were estimated by Cox proportional hazards …
Dietary Depletion Of Milk Exosomes And Their Microrna Cargos Elicits A Depletion Of Mir-200a-3p And Elevated Intestinal Inflammation And Chemokine (C-X-C Motif) Ligand 9 Expression In Mdr1a−/− Mice, Di Wu, Hatem Kittana, Jiang Shu, Stephen D. Kachman, Juan Cui, Amanda E. Ramer-Tait, Janos Zempleni

Background: Exosomes transfer regulatory microRNAs (miRs) from donor cells to recipient cells. Exosomes and miRs originate from both endogenous synthesis and dietary sources such as milk. miR-200a-3p is a negative regulator of the proinflammatory chemokine (C-X-C motif) ligand 9 (CXCL9). Male Mdr1a−/− mice spontaneously develop clinical signs of inflammatory bowel disease (IBD).
Objectives: We assessed whether dietary depletion of exosomes and miRs alters the severity of IBD in Mdr1a−/− mice owing to aberrant regulation of proinflammatory cytokines.
Methods: Starting at 5 wk of age, 16 male Mdr1a−/− mice were fed either milk exosome– and RNA-sufficient (ERS) or milk exosome– and …
