In species who consume folivorous diets, immature individuals must contend with the challenges of extracting nutrients from fibrous foods before dietary adaptations and strategies are fully developed. Additionally, immatures have distinct nutritional needs to support their stage-specific metabolic and biophysiological requirements. To meet these stage-specific needs, while constrained by underdeveloped feeding strategies and digestive capacities, immatures may adopt distinct diets better suited to their specific developmental context. However, where dietary modification is constrained by low dietary diversity or landscape homogeneity, it is unclear how immature individuals compensate through alternative strategies. The modern broiler is growing at a rapid rate generating tremendous amounts of heat. A sensitive Net Energy (NE) system is needed to measure body heat production (HP) generated primarily by daily maintenance and synthesis and degradation of myofibrillar and sarcoplasmic protein. The first two chapters present evaluation of the classic way to calculate NE versus a new methodology; the Arkansas NE (Ark NE) system, with birds from two genetic lines fed diets with different AA content or different ME content in two different environmental temperatures.

I tested the hypothesis that recombinant leptin protein can be introduced to zebrafish in vivo through non-invasive soaking in a solution containing the protein. One way to study various molecules’ effects in vivo is through intraperitoneal or intracerebroventricular injections during the embryonic or larval stage, which is invasive, difficult to administer, and can have a high mortality rate. 48 hours post fertilization (hpf) zebrafish were soaked in a His-tagged recombinant leptin protein solution at 10 nM and 100 nM concentrations (produced by Genscript). After soaking, zebrafish larvae were washed extensively to remove all recombinant protein on their exterior before homogenization. …

The prevalence of non-alcoholic fatty liver disease (NAFLD) in children has increased over the past decades, creating a need for animal models that recapitulate the features of the pediatric disease. Iberian pigs have a leptin-resistant phenotype characterized by hyperleptinemia, hyperphagia, and extreme adipogenesis. We hypothesized that neonatal Iberian pigs fed a high fat high-fructose (HFF) diet will develop a pattern of liver injury resembling pediatric NAFLD. Consumption of endophyte-infected tall fescue results in a syndrome of negatively altered physiological systems, collectively known as fescue toxicosis. Another challenge to endophyte-infected tall fescue -based beef cattle operations is that the soils often are selenium (Se) poor, necessitating the need to provide supplemental Se. Fifty pregnant Katahdin ewes grazed pastures with high (HE) and moderate (MI) levels of Neotyphodium coenophialum-infected tall fescue between February 1 and May 24 to determine the impacts of tall fescue toxins on body weight (BW), body condition scores, FAMACHA scores, fecal egg counts (FEC), and serum Cu, Zn, prolactin, and non-esterified fatty acids (NEFA). Within those 50 ewes, 15 were selected randomly for fecal and rumen sampling to assess the impacts of HE and MI on the microbiome community. Two trials were performed using one-day-old male Cobb x Cobb 500 broilers to determine how dietary phytate and phytase levels as well as phytase phase feeding impacted bird performance parameters, tibia characteristics, and malonaldehyde (MDA) content of the liver, breast and thigh tissues. The first experiment consisted of 1,008 birds randomly placed in 48 floor pens within two commercial broiler houses at the Applied Broiler Research Farm (ABRF; 21 birds per pen; 0.76 ft2 per bird). 4,096 broiler chicks were randomly allocated to 128 floor pens (32 birds/pen). 2,048 day-old male broilers were placed in the east end of a barn, and the following week 2,048 day-old male broilers were placed in the west end. At each placement day, half of the chicks were Cobb 500 and half were Ross 708, and each pen contained only one breed source. East end birds received coccidiostat in the feed, west end received coccidial vaccine, and each end was under separate environmental control. It is well established that l,25(OH)2D3 is a major regulator of calcium homeostasis. The steroid exerts its effects on its target organs by two mechanisms, a slow mechanism mediated by nuclear vitamin D receptors (nVDR), and a rapid mechanism mediated by the l,25(OH)2D3 membrane-associated rapid response steroid binding protein (l,25D3-MARRS bp).
