Early-Life Stress (ELS) is associated with vulnerability to mood disorder, but it’s not well understood how ELS contributes to deficits in cognitive function. Atypical PKMzeta is critical for LTP maintenance and memory. The current study aims to characterize the ELS phenotype with respect to this key marker of hippocampal LTP.

Childhood maltreatment is strongly associated with depression, which is characterized by reduced reactivity to reward. Identifying factors that mitigate risk for depression in maltreated children is important for understanding etiological links between maltreatment and depression as well as improving early intervention and prevention. We examine whether high reward reactivity at behavioral and neurobiological levels is a marker of resilience to depressive symptomology in adolescence following childhood maltreatment. A sample of 59 adolescents (21 with a history of maltreatment; Mean Age = 16.95 years, SD = 1.44) completed an fMRI task involving passive viewing of emotional stimuli. BOLD signal changes to …

Social stress, including bullying during adolescence, is a risk factor for common psychopathologies such as depression. To investigate the neural mechanisms associated with juvenile social stress-induced mood-related endophenotypes, we examined the behavioral, morphological, and biochemical effects of the social defeat stress model of depression on hippocampal dendritic spines within the CA1 stratum radiatum. Adolescent (postnatal day 35) male C57BL/6 mice were subjected to defeat episodes for 10 consecutive days. Twenty-four h later, separate groups of mice were tested on the social interaction and tail suspension tests. From 2000-2009, suicide mortality rates increased by 15% (Rockett, et al., 2012) and 98% of individuals who complete suicide have diagnosable psychopathology (Bertolote, Fleischmann, De Leo, & Wasserman, 2003). Individuals at risk for suicide exhibit cognitive impairments in decision-making and problem-solving abilities (Pollock & Williams, 2004; Jollant, et al., 2005). These cognitive deficits may be related to deficits in performance monitoring. Performance monitoring is the ability to continually assess behavior in order to make adjustments and improve performance. Separation and Its Effect on the Forced Swim Test In the Guinea Pig Early-life stress such as parental neglect, absence, or abandonment, has been hypothesized to increase the susceptibility for developing depression later in life via sensitization of stress-responsive physiological systems (e.g., pro-inflammatory cytokines, hypothalamic-pituitary-adrenal axis). Guinea pigs offer a potential model, but study has been limited to behavioral observations obtained during maternal separation tests. This thesis examined the generalization of this response by asking whether it would cross-sensitize to behavior in another depressive-related paradigm, the forced swim test. Depression is a serious, costly, and heterogeneous disorder for which no one genetic determinant has been identified. Research has shown that stress, and subsequent hypothalamic-pituitary-adrenal (HPA) axis dysregulation, is a significant predictor of depression, and one particular stressor that has been linked to vulnerability to depression and HPA axis dysregulation is early life trauma. Due to the heterogeneity and complexity of depression, it is likely that specific gene-environment interactions play a role in the development of depression. Interaction between catechol-O-methyltransferase (COMT) Val158Met variants with specific environmental factors can potentially increase vulnerability to depression. The present proposed experiment …
