Neural mechanisms for grip relaxation are relatively unknown and understudied, as compared to mechanisms for grip initiation. Yet, termination of motor activity is as important as initiation in daily function. This knowledge gap presents incomplete understanding of neural control of hand function and its impairment with aging and neurologic disorders. The purpose of this dissertation was to identify and examine neural mechanisms of grip relaxation in healthy young adults, with aging, and in chronic stroke survivors. Stroke is the most common cause of long-term severe disability and the motor system that is most commonly affected in stroke. One of the mechanisms that underlies recovery of motor deficits is reorganization or remapping of functional representations around the motor cortex. This mechanism has been shown in monkeys, but results in human subjects have been variable. Acquired Brain Injury (ABI) includes any damage to the brain resulting from traumatic (e.g. motor vehicle accident) or non-traumatic (e.g. stroke) incidence, that occurs after birth and is not resulting from genetic or congenital factors. Individuals with ABI report that prospective memory (PM) deficits are the most detrimental cognitive impairment following injury, persistently and negatively impacting their ability to function properly in everyday life. PM refers to the ability to remember to carry out intended tasks in the future, including the recall of both time and event regulated intentions. Ischemic stroke is a major world-wide health problem, resulting in death and disability especially in the older population. A sex difference exists in functional recovery post-stroke, with post-menopausal women having worse functional outcome as compared to age-matched men. Although the mechanisms underlying this sex difference are not entirely clear, it is recommended that any potential therapy for stroke recovery be tested in pre-clinical models including both male and females in order to determine the efficacy of the proposed treatment on the entire population.
