The ability of uropathogenic Escherichia coli (UPEC) to successfully establish an infection within the urinary tract relies on the pathogen to be able to navigate the complex interactions between pathogen and host. During a urinary tract infection (UTI), UPEC must ascend, adhere, and form bacterial communities in various infection niches, from the bladder (cystitis) to the kidneys (pyelonephritis). Each of these steps is critical for bacterial survival, and host and pathogen factors required at each stage can vary depending on host sex and anatomic niche. Urinary tract infections (UTIs) are among the most common bacterial infections which plague humans. Community-onset UTI is widely viewed as a disease only of women; indeed, its occurrence between 2 and 60 years of age is almost exclusive to females. However, the disease also affects substantial male patient populations, namely infants and the very elderly. While female cases of complicated and upper-tract UTI (pyelonephritis) outnumber male cases overall, men carry an increased risk of mortality from these infections. Urinary tract infections (UTI) are one of the most common bacterial infections, of which, the majority are caused by uropathogenic Escherichia coli (UPEC). Many bacterial pathogens including UPEC synthesize and secrete chemically diverse metabolites called siderophores, which are classically defined by their ability of bind and deliver iron(III), an essential nutrient, to pathogens during infections. UPEC isolates can express multiple siderophore systems, of which, the virulence-associated siderophore yersiniabactin (Ybt) binds to both iron(III) and copper(II) during urinary tract infections. In this thesis we show that Ybt interacts with multiple physiologic transition metals and acts as a metallophore system to deliver …
