Exploring The Developmental Effects Of Antidepressants In Caenorhabditis Elegans, Alexandra Van Stone

Annual Research Symposium

It is so important to explore the potential side effects of antidepressants especially if these side effects could lead to a future detriment to an individual’s financial and social well-being. A better understanding of these side effects in regards to human developmental issues is imperative considering issues related to mental health and depression affect a large percentage of the population. In this study, we tested if Caenorhabditis (C.) elegans could be a potential model organism to predict possible side effects associated with select antidepressants. Their developmental problems potentially encountered upon drug treatment may be encountered in humans.

Sulforaphane Pre-Treatment Improves Cytoprotection Against Opportunistic Pathogens, Robert Darby, Nathan Clark Feb 2024

Sulforaphane Pre-Treatment Improves Cytoprotection Against Opportunistic Pathogens, Robert Darby, Nathan Clark

Annual Research Symposium

Alcohol is associated with increased mortality and morbidity globally. Pulmonary infections with opportunistic pathogens can occur in healthy humans; however, binge alcohol intoxication (≥ 0.08% BAC) is a major risk factor. We have previously shown that a single dose of alcohol comparable to binge alcohol intoxication increases infection by reducing alveolar macrophage function in vivo. The aim of this study was to 1) test the therapeutic potential of the phytonutrient sulforaphane (SFN) given as a pre-treatment, and 2) test the alcohol-induced effects on phagocytic function in murine and human macrophages in vitro.

Investigating The Impact Of Ozempic Treatment On Alcohol Consumption In Individuals With Class 1 And Class 2 Obesity, Yvannia Gray, Selin Karadag Feb 2024

Investigating The Impact Of Ozempic Treatment On Alcohol Consumption In Individuals With Class 1 And Class 2 Obesity, Yvannia Gray, Selin Karadag

Annual Research Symposium

The purpose of this experiment is to see if there is a correlation between Ozempic (semaglutide) treatment for weight loss and alcohol consumption. Ozempic is a glucagon-like peptide-1 (GLP-1) agonist that lowers blood sugar and appetite as it mimics the effects of GLP-1. In previous studies involving rodents, it has been shown that GLP-1 agonists can also be used to reduce addictive substance intake. We hypothesize that subjects who receive Ozempic treatment will exhibit reduced consumption of alcohol over the span of a month compared to subjects who do not receive Ozempic treatment.

Ksp1 Is An Autophagic Receptor Protein For The Snx4-Assisted Autophagy Of Ssn2/Med13, Sara E Hanley, Stephen D Willis, Steven J Doyle, Randy Strich, Katrina F Cooper Feb 2024

Ksp1 Is An Autophagic Receptor Protein For The Snx4-Assisted Autophagy Of Ssn2/Med13, Sara E Hanley, Stephen D Willis, Steven J Doyle, Randy Strich, Katrina F Cooper

Rowan-Virtua School of Osteopathic Medicine Departmental Research

Ksp1 is a casein II-like kinase whose activity prevents aberrant macroautophagy/autophagy induction in nutrient-rich conditions in yeast. Here, we describe a kinase-independent role of Ksp1 as a novel autophagic receptor protein for Ssn2/Med13, a known cargo of Snx4-assisted autophagy of transcription factors. In this pathway, a subset of conserved transcriptional regulators, Ssn2/Med13, Rim15, and Msn2, are selectively targeted for vacuolar proteolysis following nitrogen starvation, assisted by the sorting nexin heterodimer Snx4-Atg20. Here we show that phagophores also engulf Ksp1 alongside its cargo for vacuolar proteolysis. Ksp1 directly associates with Atg8 following nitrogen starvation at the interface of an Atg8-family interacting …

Methamphetamine-Induced Dna Double-Stranded Breaks: The Impact Of The Dopamine Transporter And Insights Into The Mechanisms Of Dna Damage In Mouse Neuro 2a Cells, Lizette Couto Feb 2024

Methamphetamine-Induced Dna Double-Stranded Breaks: The Impact Of The Dopamine Transporter And Insights Into The Mechanisms Of Dna Damage In Mouse Neuro 2a Cells, Lizette Couto

Dissertations, Theses, and Capstone Projects

Methamphetamine (METH) abuse remains a global health concern, with emerging evidence highlighting its genotoxic potential. In the central nervous system METH enters dopaminergic cells primarily through the dopamine transporter (DAT), which controls the dynamics of dopamine (DA) neurotransmission by driving the reuptake of extracellular DA into the presynaptic neuronal cell. Additional effects of METH on the storage of DA in synaptic vesicles lead to the dysregulated cytosolic accumulation of DA. Previous studies have shown that after METH disrupts intracellular vesicular stores of DA, the excess DA in the cytosol is rapidly oxidized. This generates an abundance of reactive oxygen species …

Vascular And Metabolic Consequences Of T Cell Aging, David J. Buckley Jan 2024

Vascular And Metabolic Consequences Of T Cell Aging, David J. Buckley

Kinesiology Dissertations - Archive

Aging is the major non-modifiable risk factor for cardiovascular disease (CVD), which is the leading cause of death in the United States. Many common CVD are diseases of the arteries. Two major hallmarks of arterial aging are 1) increased large artery stiffness and 2) endothelial dysfunction which is characterized by impaired endothelium dependent vasodilation and blunted nitric oxide (NO) bioavailability. Importantly, these adverse alterations in the arteries occur prior to, and are predictors of the onset of overt CVD. Advancing age is also a non-modifiable risk factor for impaired glucose homeostasis and is directly or indirectly associated with almost every …

Revolutionizing Our Understanding Of Specific Platelet-Pathogen Interactions, Hammodah Rawhi Hammodah Alfar Jan 2024

Revolutionizing Our Understanding Of Specific Platelet-Pathogen Interactions, Hammodah Rawhi Hammodah Alfar

Theses and Dissertations--Molecular and Cellular Biochemistry

Aside from their classical role in maintaining hemostasis, platelets are considered first-line vascular guardians that influence both the innate and adaptive immune systems. This is mediated by either direct interactions with immune cells or through the release of pre-stored cytokines and chemokines upon interacting with pathogens such as viruses. While this establishes platelets as potential players in the response to pathogens, it remains unclear what platelets can do with pathogens and how important that is to immune responses, especially against viruses. Most systemic viremias, such as HIV-1, present with mild thrombocytopenia, which is mediated through several mechanisms such as a …

Fillet Weight Comparison For Bluegill, Black Crappie, And Yellow Perch, Mikenna A. Pattrin Jan 2024

Fillet Weight Comparison For Bluegill, Black Crappie, And Yellow Perch, Mikenna A. Pattrin

Journal of Earth and Life Science

Panfish, including bluegill Lepomis macrochirus, black crappie Pomoxis nigromaculatus, and yellow perch Perca flavescens are three sought-after species. Anglers often perceive a change in regulations as an attempt at limiting the amount of harvest. However, the goal of many regulation proposals is to increase the size structure of panfish, allowing anglers to achieve their ideal harvest with less fish. The primary objective of this study was to assess the relationship between panfish fillet weight and total length to estimate how many fillets would be needed to reach a harvest weight of 0.5 lbs. The secondary objective was to …

Plk1 Overexpression Drives Tumorigenesis In A Prostate Cancer Mouse Model, Emile Kachouh

Theses and Dissertations

Prostate cancer is the leading cause of cancer-related deaths among men worldwide, and the development of resistance to existing treatments necessitates the exploration of new therapeutic strategies, including combination therapies. This study investigates the role of Polo-like kinase 1 (PLK1), located on chromosome 16p12.2, as an oncogene in prostate cancer. We generated a transgenic mouse model with prostate-specific overexpression of PLK1 to closely mimic human prostate cancer progression. Quantitative PCR (qPCR) analysis was employed to characterize the molecular profile of PLK1, revealing its implication in multiple pathways. To further elucidate the functional consequences of PLK1 overexpression, we confirmed phenotypic progression …

The Splice Index As A Prognostic Biomarker Of Strength And Function In Myotonic Dystrophy Type 1, Marina Provenzano Jan 2024

The Splice Index As A Prognostic Biomarker Of Strength And Function In Myotonic Dystrophy Type 1, Marina Provenzano

Theses and Dissertations

Myotonic dystrophy type 1 (DM1) is a slowly progressive, multisystem disorder caused by a CTG repeat expansion in the DMPK 3’UTR that leads to global dysregulation of alternative splicing. The resulting decline in physical function is slow, and no reliable biomarkers exist for predicting disease progression; however, an RNA mis-splicing biomarker associated with weakness may have utility in predicting functional outcomes. Here we validate the Splice Index (SI) as a potential biomarker of DM1-associated strength and function. Muscle biopsies of the tibialis anterior were collected from DM1-affected individuals at baseline (n = 46) and 3-months (n = 34), along with …

The purpose of this research is to improve understanding of the neurodevelopmental effects of embryonic exposure to elevated inflammation and oxidative stress induced by the antipyretic drug acetaminophen (APAP). Our study was the first to examine the interactive effects of APAP and inflammation in zebrafish embryos and how the treatments affect brain development and larval behavior. Experimental groups of zebrafish larvae were exposed to lipopolysaccharide (LPS) to induce inflammation, APAP, or LPS + APAP and larval behavior was analyzed using Ethovision automated behavioral tracking software. We also measured changes in whole-brain Glycogen Synthase Kinase 3 Beta (GSK3B) and GSK3B phosphorylation, …

Effects Of Unilateral Lower Limb Suspension On Quadriceps’ Structure And Contractile Function, Eve N. Elmore Jan 2024

Effects Of Unilateral Lower Limb Suspension On Quadriceps’ Structure And Contractile Function, Eve N. Elmore

Theses and Dissertations--Kinesiology and Health Promotion

BACKGROUND: Muscle unloading such as bed rest or space flight decreases strength and cross-sectional area concomitant with a slow-to-fast fiber type shift, of anti-gravitational muscles such as the quadriceps. Diffusion tensor imaging (DTI) quantifies tissue anisotropic properties such as Fractional Anisotropy, Mean Diffusivity, Axial Diffusivity, and Radial Diffusivity, as well as muscle volume, fascicle length, and physiological cross-sectional area, and has recently been adapted to assess skeletal muscle structure. In skeletal muscle, anisotropic properties correlate with muscle fiber type and contractile function and may be responsive to unloading-induced atrophy.

PURPOSE: To assess the changes in quadriceps’ size, anisotropy, and contractile …

Inhibiting Pi3k/Akt To Enhance Brain Uptake Of Anticancer Agents In Glioblastoma, Louis Rodgers Jan 2024

Inhibiting Pi3k/Akt To Enhance Brain Uptake Of Anticancer Agents In Glioblastoma, Louis Rodgers

Theses and Dissertations--Pharmacy

Glioblastoma (GBM) is the deadliest and most common malignant CNS tumor. Despite advances in understanding its biology and the development of targeted therapies, effective treatment options remain limited. As a result, patient survival rates are dismal, with a 5-year survival rate of only 6.8% and an overall survival of less than two years. This poor outcome is partly due to our inability to treat GBM cells that infiltrate the whole brain. Focal treatments like surgery and radiotherapy alone are insufficient due to these invasive tumor cells. Therefore, chemotherapy following resection and irradiation of the primary tumor is essential to eliminate …

The Delineation Of Platelet Pathology In Cardiometabolic Disease, Chi Peng Jan 2024

The Delineation Of Platelet Pathology In Cardiometabolic Disease, Chi Peng

Theses and Dissertations--Pharmacology and Nutritional Sciences

Obesity is a multifactorial disease with many comorbidities leading to multiorgan dysfunction. Current studies point towards dysregulation of platelet signaling as a driver for increased risk of cardiovascular disease and stroke. The exact mechanisms remain elusive due to the complex interplay of factors involved, warranting the need to further understand how platelets are affected. Our preliminary data evaluating changes in platelet and coagulation parameters from bariatric patients showed that obesity did not affect platelet count or clotting time as demonstrated in INR values. However, female patients had increased bleeding events before surgery that were decreased after surgery. Due to limitations …

Long Term Trends In Mortality Rates Of A Smallmouth Bass Population, Caleb D. Anderson Jan 2024

Long Term Trends In Mortality Rates Of A Smallmouth Bass Population, Caleb D. Anderson

Journal of Earth and Life Science

Smallmouth bass Micropterus dolomieu are known as a popular sport fish for anglers. When a known smallmouth bass fishery appears to have a dwindling population, it raises questions about the population health. A common indicator of the health of the population is the mortality rate. By comparing mortality, conclusions can be made on whether smallmouth bass are dying at a different rate in the system over time. Therefore, the objective of this study was to see if there had been a significant change in smallmouth bass mortality over a span of thirty years in Round Lake (DOW:010204), Aitkin County, Minnesota. …

Relationship Between Northern Pike And Walleye Cpue In Minnesota Lakes, Simon Oslund Jan 2024

Relationship Between Northern Pike And Walleye Cpue In Minnesota Lakes, Simon Oslund

Journal of Earth and Life Science

Northern pike Esox lucius and walleye Sander vitreus are both top predators in freshwater systems. Due to their predatorial nature, high populations of one species can prohibit populations of the other species from thriving, specifically northern pike foraging on walleye. This information is tracked by the Minnesota Department of Natural Resources through catch per unit effort (CPUE). CPUE data was gathered for all lakes in Beltrami County, Minnesota, and a regression analysis was done to test for a relationship between pike and walleye. It was found that the relationship between northern pike and walleye CPUE was significant (P = 0.02). …

Journal of Earth and Life Science

Burbot Lota lota in north-central Minnesota lakes have the potential to reach large sizes and consume large volumes of prey. This caveat may make burbot susceptible to higher rates of biocontamination, bioaccumulation and biomagnification. The objective of this study was to determine how changes in age, length, gender, weight, and lake affect total mercury concentrations in burbot. In this experiment 28 burbot were angled from three lakes: Cass (n = 17), Winnibigoshish (n = 4), and Bad Medicine (n = 7). Then tissue samples were taken from each fish and were lyophilized and homogenized. Homogenized tissue samples were analyzed by …

Relationship Between Fishing Hours And Number Of Fish Harvested From Upper Red Lake, Dylan Ohlenkamp Jan 2024

Relationship Between Fishing Hours And Number Of Fish Harvested From Upper Red Lake, Dylan Ohlenkamp

Journal of Earth and Life Science

Total hours fishermen spend fishing and total amount of fish harvested per year are some of the most important pieces of knowledge gained from creel surveys. They provide crucial information that assesses fisheries mortality and pressure. MN DNR uses their results from creel surveys to improve or impose regulations, and to maintain healthy fisheries. Based on results from similar studies, there is expected to be a positive relationship between hours fished and fish harvested. To determine the relationship between these two variables on Upper Red Lake, data from two years were selected when the variables were at their highest and …

A Guide To Otoliths Of Minnesota Fishes, Baylor J. Short Jan 2024

A Guide To Otoliths Of Minnesota Fishes, Baylor J. Short

Journal of Earth and Life Science

Investigations into the diet patterns of piscivores can provide crucial information on predator-prey relationships, population dynamics, and responses to changing ecosystems. However, digestive processes often remove or alter physical characteristics that are traditionally used to identify consumed fish. This problem has been addressed to some degree with advances in molecular technologies, although these methods can be costly and require specific training and equipment to do so. In contrast, bony structures such as otoliths, vertebrae, cleithra, and others frequently have morphologies that are unique among families, genera, and species. Because these structures are resistant to digestion, they can be used to …
