Environmental Changes Turn On The Sinorhizobium Melitloti Exor-Exos/Chvi (Rsi) Host Invasion Switch, Shari N. Walcott Feb 2017

Environmental Changes Turn On The Sinorhizobium Melitloti Exor-Exos/Chvi (Rsi) Host Invasion Switch, Shari N. Walcott

Dissertations, Theses, and Capstone Projects

The free-living Gram-negative soil bacterium, Sinorhizobium meliloti, must switch into its host-invading form in order to infect the root hairs of the host plant, alfalfa (Medicago sativa), and establish a nitrogen-fixing symbiosis. The activation of the switch is believed to occur inside the infection chamber that is formed by curling of the root hairs. It is not fully understood what signals in the environment of the root hairs trigger and infection chamber S. meliloti to switch into a host-invading form since these signals were not extensively examined until now. This switch can be observed directly, due to …

The Hetz Gene Regulates Heterocyst Formation In Anabaena Sp. Strain Pcc 7120, Patrick Videau, Vaille Swenson, Michael Gaylor, S M. O'Hanlon, L M. Cozy

Research & Publications

To form a complex multicellular organism, stem cells must differentiate into each cell/tissue type along proper spatiotemporal scales. The study of differentiation and organismal development has historically been conducted in prokaryotes due to their genetic and morphological simplicity. Anabaena sp. strain PCC 7120 is a multicellular filamentous cyanobacterium that differentiates a morphologically distinct secondary cell type, the heterocyst, in response to a lack of combined environmental nitrogen. Heterocysts are regularly spaced along filaments and fix atmospheric dinitrogen to maintain organismal viability in its absence. Previous work suggested that the hetZ gene is involved in heterocyst differentiation, but the insertional mutants …

Development Of Cellular Assays To Monitor Enzymatic And Biological Activity Of Cd73: A Key Modulator Of Anti-Tumor Immune Response, Alexandra Fanuka Jan 2017

Development Of Cellular Assays To Monitor Enzymatic And Biological Activity Of Cd73: A Key Modulator Of Anti-Tumor Immune Response, Alexandra Fanuka

Graduate School of Biomedical Sciences Theses and Dissertations

Ecto-5’-nucleotidase, known as CD73, is an extracellular enzyme that converts adenosine monophosphate (AMP) to adenosine and has recently been identified as a potential drug target for cancer immunotherapy. Its immunosuppressive effects, mediated by the activity of adenosine, are associated with higher rates of tumor invasion and metastasis, as well as poorer prognoses overall in many cancer types. CD73 is often co-expressed with ectonucleoside triphosphate diphosphohydrolase-1 (CD39), which catalyzes the conversion of adenosine triphosphate (ATP) to adenosine diphosphate (ADP), and ADP to AMP on the surface of tumor cells. Dual expression further propagates immunosuppressive effects of adenosine in the tumor microenvironment. …

Knowledge of the characteristics that promote group success is key to understanding the adaptive advantages of sociality for many group-oriented species. In this study, nine Gray Wolves Canis lupis were each observed twelve times over a period of six months to determine if the amount of time spent in the wild/captivity had any effect on aggressive behaviors. The number of aggressive behaviors were recorded for each wolf during each observation. The nine wolves were separated into three groups depending on their upbringing; born and raised in captivity, born in the wild and raised in captivity, and born and raised in …

Low Cost, Multi-Purpose Genotyping Panels For Dairy And Beef Cattle, Michelle Judge Jan 2017

Low Cost, Multi-Purpose Genotyping Panels For Dairy And Beef Cattle, Michelle Judge

Theses

rhe selection of animals based on DNA has revolutionised animal breeding, but the associated high cost of obtaining genotypes has limited its uptake. The objective of this thesis was to develop a low-cost, low-density, multi-purpose genotyping panel for the procurement of reliable genotype information, and to quantify the long-term consequences of using such low-density genotype panels in breeding programs. The objectives were achieved through a combination of real-life cattle genotype data and simulations to mimic a cattle population. The in-silico development of genotype panels was based on actual genotypes from up to 58,705 beef cattle. Alternative novel strategies were used …

A Comparison Of Benthic Macroinvertebrate Assemblages Between Perennial And Intermittent Headwater Streams Of The Mattole River In Northern California, Usa, Mason S. London

Cal Poly Humboldt theses and projects

Intermittent streams are common throughout the world and comprise 60% or more of total river lengths in the conterminous United States. Despite their prevalence, intermittent streams are understudied, particularly first-order headwater streams, which are vital for maintaining the function, health and biotic diversity of river networks. In June 2016, I sampled five intermittent and five perennial headwater streams in the Mattole River watershed in northwestern coastal California, USA, to compare benthic macroinvertebrate (BMI) assemblages between intermittent and perennial streams. BMI samples were collected using a 500µm mesh D-net at eight randomly located riffles along a 150-m reach, and then composited, …

Ionic Regulation Of Critical Cellular Processes In Non-Excitable Cells, Brandon M. Franklin Jan 2017

Ionic Regulation Of Critical Cellular Processes In Non-Excitable Cells, Brandon M. Franklin

Theses and Dissertations--Biology

There are long-standing hypotheses that endogenous ion currents act to control cell dynamics in development, wound healing and regeneration. However, the mechanisms employed by cells to detect the electric field (EF) and translate it into a discernable message to drive specific cell behaviors, such as migration, proliferation and differentiation, are not well understood. A better understanding of how cells are able to sense EFs and react to them is vital to understanding physiological mechanisms are involved in regeneration. Ion channel signaling provides a reasonable suspect for mediating these effects based on their documented involvement in proliferation, migration and differentiation.

Three treatment groups were used; immediate catch-and-release, three-minute, and a seven- minute …

Effect Of Abiotic Factors On Bluegill Size Structure In Nine Central Minnesota Lakes, Ryan Henry Jan 2017

Effect Of Abiotic Factors On Bluegill Size Structure In Nine Central Minnesota Lakes, Ryan Henry

Journal of Earth and Life Science

Bluegill are the most targeted species of fish in Minnesota. In recent years, the abundance of slow growing, small Bluegill has increased within the state. Angler harvest of Bluegill can reduce the average length of Bluegill up to four times the original length. Abiotic factors such as maximum depth, average water clarity, littoral area, and percent littoral area can influence the dynamics of a lake system. There has been limited research on whether these abiotic factors can influence the size structure of a Bluegill population. The objective of this study is to determine if there is a direct relationship between …

Chronic stress causes deleterious changes in physiological function in systems ranging from neural cells in culture to laboratory rodents, sub-human primates and humans. It is notable, however, that the vast majority of research in this area has been conducted in males. In this review, we provide information about chronic stress effects on cognition in female rodents and contrast it with responses in male rodents. In general, females show cognitive resilience to chronic stressors which impair male cognitive function using spatial tasks including the radial arm maze, radial arm water maze, Morris water maze, Y-maze and object placement. Moreover, stress often …
