Accurate estimation of the hip joint center (HJC) is critical for biomechanical modeling and clinical assessment. Common methods include predictive methods (e.g., regression equations) and functional methods such as sphere-fitting. In infants, femoral head-based methods are often limited due to incomplete ossification of the anatomy and poor visualization in computed tomography (CT) images. This study evaluates an alternative approach using MRI-based sphere fitting of the ossified acetabular rim, comparing its accuracy to traditional femoral head-based methods. Cancer remains a predominant health challenge that is responsible for a significant portion of global morbidity and mortality. Epirubicin (EPI), a chemotherapeutic anti-cancer drug, has shown remarkable efficacy in combating various malignancies. However, it is known to have undesirable side effects on the heart, collectively referred to as cardiotoxicity. This research investigates the adverse effects of chemotherapy on cardiac contractility through an in vitro examination of the mechanics of healthy and infarcted animal heart tissues. Patellofemoral Pain Syndrome (PFPS) is the most prevalent overuse injury in cycling, often linked to altered neuromuscular activation patterns of the quadriceps and hamstrings. While sensory feedback has successfully modified cyclists posture, there is a critical lack of evidence-based interventions targeting the underlying muscle activation imbalances associated with chronic knee pain. This study investigates how type Ia feedback from muscle spindles can be organized into groups representing agonistic muscle pairs through Spike Timing Dependent Plasticity (STDP). A single degree of freedom joint is actuated with four biologically modeled muscles forming two agonistic pairs. In order to emulate the sensory dynamics of biological muscle spindles, sensors in the model record the active length and velocity states of each muscle, the two primary factors eliciting type Ia afferent responses. This thesis evaluates whether accessible and affordable technologies can provide clinically significant, quantifiable gait analysis comparable to traditional marker-based motion capture systems. Modern high-end systems offer excellent accuracy but remain financially and logistically inaccessible to many clinicians and researchers. To address this gap, a framework was developed using markerless pose estimation using the model, MediaPipe, inertial measurement units (IMUs) processed through an Extended Kalman Filter (EKF), and surface electromyography (sEMG) to measure joint kinematics and neuromuscular activity during gait. This thesis investigates the biomechanical behavior of the human knee under varying conditions of motion, load, and fatigue, with a focus on clinical angle analysis and gait stability. Four studies were conducted using motion capture data and a modified Grood and Suntay Joint Coordinate System (JCS) to evaluate knee joint kinematics—specifically flexion-extension, abduction-adduction, and internal-external rotation. Spinal ligaments play a crucial role in maintaining the mechanical stability of the lumbar spine. These dense, collagenous tissues not only resist excessive motion but also distribute loads between spinal components to protect neural structures. Traditionally, the mechanical response of ligaments has been modeled using linear elastic assumptions, which treat the tissue as having a constant stiffness regardless of loading history. While this simplifies analysis, it fails to capture the time-dependent behaviors, such as creep, stress relaxation, and hysteresis, that are characteristic of biological tissues.

Gait impairments arise from systemic diseases, age-related degeneration, musculoskeletal dysfunctions, or neurological conditions. While traditional rehabilitation can be effective, they often face challenges such as high costs, inaccessibility, and low patient engagement. To address these challenges, my work introduces a virtual reality-based rehabilitation (VRBR) system, integrating real-time motion and electromyographic (EMG) muscle activation feedback with a gamified virtual environment for enhanced adaptability and engagement. The system includes a custom-designed hip-exoskeleton that provides adaptive spring-like assistance or resistance, supporting both mobility-impaired users and strength training. Chronic low back pain (LBP) has widespread negative impacts on quality of life. Current treatments for chronic LBP fail to provide long-term efficacy and motivate the need to understand key drivers of poor patient outcomes. Mounting evidence indicates that sleep disturbances and chronic LBP have a strong reciprocal relationship. Women may be particularly vulnerable to the negative impacts of the sleep-pain axis, exhibiting greater prevalence in both conditions. This dissertation addresses four specific aims that collectively attempted to advance the experimental and computational analysis of total knee arthroplasty (TKA) components. The first study focused on the development of a novel whole knee joint simulator capable of simultaneous tibiofemoral and patellofemoral knee loading. This simulator employs custom fixturing to facilitate dynamic, unconstrained, muscle-driven PF articulation alongside controlled TF contact mechanics. Validation against experimental measurements showed strong agreement, demonstrating the simulator's potential as a valuable tool for future TKA design and surgical technique investigations.

Individuals with hip-related pain exhibit altered movement patterns, and worse fear of movement (kinesiophobia), compared to asymptomatic controls. Exacerbation of hip pain during movement and the role of hip symptom duration may help further our understanding of specific movement patterns that are associated with hip-related pain. Contemporary total knee arthroplasty (TKA) has not fully restored natural patellofemoral (P-F) mechanics across the patient population. Previous experimental simulations have been limited in their ability to create dynamic, unconstrained, muscle-driven P-F articulation while simultaneously controlling tibiofemoral (T-F) contact mechanics. The purpose of this study was to develop a novel experimental simulation and validate a corresponding finite element model to evaluate T-F and P-F mechanics. Osteoarthritis (OA) is a form of arthritis that develops in the joint due to overuse and aging causing pain, discomfort, and disability. Total Knee Arthroplasty (TKA) is a surgical procedure performed when OA symptoms are severe with an estimated 600,000 patients in the United States currently receiving TKA. Studies have reported dissatisfaction of the knee for 14-39% of patients. The glenohumeral joint (GH) is commonly conceptualized as a ball-and-socket joint [1], and its center of rotation (COR) is presumed to coincide with the geometric center of the medial-superior region of the humeral head [2]. Recent research has endorsed improvements in COR estimation through invasive and noninvasive techniques, including cadaver studies, stereophotogrammetry, and motion capture (MOCAP) systems. Competitive sports require rapid and intense movements, such as jump landings, making athletes susceptible to injuries due to altered neuromuscular control and joint mechanics. Biomechanical features during landings are associated with injury risk, emphasizing proper movement and postural stability. Computer vision techniques offer a time-efficient, noninvasive, and unbiased method to assess jump-landings and identify injury risks. An Institutional Review Board (IRB)-approved study was conducted to investigate the effects of the Tsunami Bar® (TB), a flexible barbell, on ground reaction force (GRF) production and muscle activity in the quadricep, hamstring, and gluteal muscle groups during phases of the squat exercise and compare the effects to the effects to using a rigid barbell (RB). A two-by-two repeated measures Analysis of Variance (ANOVA) test was used to compare the results. Brain–body interactions (BBIs) have been the focus of intense scrutiny since the inception of the scientific method, playing a foundational role in the earliest debates over the philosophy of science. Contemporary investigations of BBIs to elucidate the neural principles of motor control have benefited from advances in neuroimaging, device engineering, and signal processing. While marker-based motion capture remains the gold standard in measuring human movement, accuracy is influenced by soft-tissue artifacts, particularly for subjects with high body mass index (BMI) where markers are not placed close to the underlying bone. Obesity influences joint loads and motion patterns, and BMI may not be sufficient to capture the distribution of a subject’s weight or to differentiate differences between subjects. The use of mathematical and computational models to understand complex biological systems, such as the human birth process, is a rapidly growing field in medicine. These models can be used to optimize and personalize medical treatments for individual patients, enhance training, and aid in educational efforts. This dissertation explores how multimodal walking impacts quantitative and qualitative aspects of gait for persons with multiple sclerosis (pwMS) experiencing foot-drop. Foot-drop can dramatically impede mobility and clinicians routinely prescribe ankle-foot-orthosis (AFO) and functional electrical stimulation (FES) devices to alleviate its impacts on daily life. However, little is known about how these devices affect pwMS while traversing environments with real-world complexity. To explore this topic, an interventional, parallel assigned study was conducted. A realistic nature pathway containing changes in floor pitch, audiovisual stimulation, and during-trial tasks (for dual-tasking) was generated in an immersive virtual reality system called CAREN: Computer Assisted …
