Muscle size does not fully explain variations in muscle strength. Fat content has been implicated in muscle weakness, though this relationship remains unclear. The relationship between fat and strength may vary between scales (e.g., cellular, organ, and organism). The goal of this dissertation was to clarify the role of fat in the structure and function of muscle using in vitro and in vivo techniques across multiple scales in adults 21-45 years old. Study 1 tested the agreement of intramyocellular lipid (IMCL) content between oil red o (ORO) and magnetic resonance spectroscopy (MRS) techniques. These measures of IMCL were also compared …

Previous studies examining the effects of exercise training on metabolism have primarily used males as participants. It has been shown that as little as one hour of aerobic exercise in males leads to the alteration of several metabolic enzymes, including phosphoenolpyruvate carboxykinase (PEPCK) and glucose-6-phosphate (G6Pase). Changes in glycogen content and differences in the usage of glycogen for fuel following exercise training have also been shown in male rats. However, it is unclear whether females demonstrate the same training related metabolic adaptations as males.

What remains unclear is how characteristics of muscle energetics affect whole-body energetics during daily living, and what effects this may have on mobility. Cachexia is a multisystemic and multifactorial syndrome prevalent in cancer patients. It is clinically defined by involuntary loss of >5% weight in a six-month window, despite nutritional interventions. A negative energy balance characterizes cancer cachexia (CC), it is associated with weakness and fatigue in skeletal muscle. Impaired muscle function is associated with lower quality of life in cancer patients. Defects in mitochondrial function are strongly associated with muscle wasting. Overhead sports such as baseball, tennis, volleyball, softball and swimming are known to cause rotator cuff imbalances and injuries. These injuries are typically caused by the repeated use of “throwing motion,” where the humerus is externally rotated and shoulder abducted and elevated. Previous research has shown that the function of the upper trapezius muscle in overhead athletes is likely to have improper activation (timing, magnitude) during various shoulder motions and/or activities of daily living. Skeletal muscle oxidative capacity plays a critical role in human health and disease. Although current models of oxidative phosphorylation sufficiently describe skeletal muscle energetics during moderate-intensity contractions, much is still unknown about the mechanisms that control and limit oxidative phosphorylation during high-intensity contractions. In particular, the oxygen cost of force generation is augmented during exercise at workloads above the lactate threshold. Presently, it is unclear whether this augmentation in muscle oxygen consumption is driven by increased rates of oxidative ATP synthesis (ATPOX) or by decreases in the efficiency of ATPOX due to mitochondrial uncoupling. Eccentric resistance training has been shown to be beneficial for improving multiple performance and health metrics. However, recommendations on eccentric training frequency have not been established. PURPOSE: To investigate the effects of volume-matched resistance training frequency comparing one versus three training days per week of isokinetic multiple-joint eccentric training on strength and lower body function adaptations during a 4-week training period. The hallmark of this injury is incomplete muscle regeneration, excessive fibrosis, and chronic inflammatory signaling resulting in permanent functional loss. Cachexia is the unintentional loss of body weight secondary to chronic disease and is prevalent is roughly 50% of cancer patients. The loss of body weight and skeletal muscle mass is reduced functional capacity associated with reduced life quality. Chronic kidney disease (CKD) is a progressive condition that may negatively affect musculoskeletal health. These comorbidities may include malnutrition, osteoporosis, and decreased lean body mass. Secondary sarcopenia due to CKD may be associated with mobility limitations and elevated fall risk. Physical therapists are well-positioned among the health care team to screen for secondary sarcopenia in those with CKD and for the treatment of musculoskeletal comorbid conditions that may affect functional performance. Cancer-related cachexia is a complex metabolic syndrome that involves the unintentional loss of skeletal muscle mass that cannot be reversed by standard nutritional intervention. Skeletal muscle mass depletion directly impacts patient survival and life quality, and therefore treatments that preserve muscle mass and function may have significant implications for cancer patient treatment and survival. Intro: Female athletes experience non-contact ACL injuries at 5 times the rate of male athletes. These injuries occur more frequently at the end of halves and may be associated with exercise-induced fatigue causing knee instability. The purpose of this pilot study was to determine the effect of strenuous exercise on lateral knee movement during the landing phase of a jump. Contractile and cytoskeletal protein concentrations have been shown to differ on the basis of fiber type in whole muscle homogenates. The purpose of this study was to compare the content of the intermediate filament protein, desmin, between type I and type IIa single muscle fibers from a mixed muscle in human subjects. Biopsies were taken from the vastus lateralis of six recreationally active males. Approximately 150 single muscle fibers were dissected from each sample and analyzed using SDS-PAGE to determine myosin heavy chain (MHC) composition.
