Paralympic athletes (PA) appear to be more prone to chronic overuse injuries from daily wheelchair or crutch use. Over half of these injuries are shoulder related which can deleteriously impact quality of life. Adaptive powerlifters (AP) are a subdivision of Paralympic athletes and are at a higher risk for catastrophic injuries as compared to their counterparts, due to the compound of fatigue and lifting of maximal weights. For this reason, it is vital to have well-designed training plans for these athletes in order to preserve quality of life and maximize performance in competition. Skeletal muscle oxidative capacity plays a critical role in human health and disease. Although current models of oxidative phosphorylation sufficiently describe skeletal muscle energetics during moderate-intensity contractions, much is still unknown about the mechanisms that control and limit oxidative phosphorylation during high-intensity contractions. In particular, the oxygen cost of force generation is augmented during exercise at workloads above the lactate threshold. Presently, it is unclear whether this augmentation in muscle oxygen consumption is driven by increased rates of oxidative ATP synthesis (ATPOX) or by decreases in the efficiency of ATPOX due to mitochondrial uncoupling. Intracellular acidosis is a putative agent of skeletal muscle fatigue, in part, because acidosis depresses the calcium (Ca2+) sensitivity and force production of muscle (18, 50). However, the molecular mechanisms behind this depression in Ca2+ sensitivity and force production are unknown. This gap in knowledge poses a significant challenge in generating a complete understanding of the fatigue process. To close this gap, the ability of myosin to bind to a single actin filament was measured under acidic conditions, in a laser trap assay, with and without regulatory proteins. Load-velocity relationships may vary between people of different strength levels and across different loads. The purpose of this dissertation was to investigate how external loads influence the velocity characteristics of the back squat exercise, and the influence of strength on these variables. Healthy male students with a history of resistance training completed repetitions at specified intensities of their estimated one-repetition maximum (1RM) until they reached 1RM. Purpose: The purpose of these studies was to determine if arterial angulations (through sitting or standing), shear stress (through local heating), or a combination of these factors affected vascular function. Moreover, this studied estimated the magnitude of the effect that biological sex has on these vascular responses. Methods: Twenty-six healthy, young (18-40 years old) males (n=13) and females (n=13) completed two experimental trials (2-h sitting and 2-h standing). In a randomized fashion, one leg was passively heated in order to increase shear rate. Following a 48-h washout, participants returned to the laboratory to complete the other trial. The purpose of the study was to measure the effects of an acute pre-competition high-carbohydrate (HCHO) meal versus a mixed-macronutrient (MM) meal on physiology, perceptual responses, and performance in 15 division I female collegiate soccer players. Being there are conjectural advantages to pre-exercise meals higher in fiber, fat, and protein, this study was conducted to evaluate the physiological and perceptual effects of pre-competition MM meals. This study compared the effects of a 4 week, 30 s weight training program (30SEC), to a low-repetition weight training program (LoRep), on rowing performance and aerobic capacity in preconditioned, provincial and national collegiate female rowers. 30SEC (n=5) performed 5 sets of dumbbell deadlifts (DL) and dumbbell bench pulls (BP), executing a maximum of 18 repetitions in 30 s, three times per week. LoRep (n=5) performed 4-5 sets of 2-6 repetitions of weight training exercises twice per week. The current investigation was an examination of the kinetic and kinematic characteristics of the back squat using accentuated eccentric loading (AEL) and cluster set programming strategies. Trained male subjects (age = 26.1 ± 4.1 years, height = 183.5 ± 4.3 cm, body mass = 92.5 ± 10.5 kg, back squat to body mass ratio = 1.8 ± 0.3) volunteered to complete four different load condition sessions involving traditionally loaded straight sets (TL), traditionally loaded cluster sets (TLC), AEL cluster sets (AEC), and AEL straight sets where only the first repetition of each set used eccentric overload (AEL1). The purpose was to evaluate muscle deoxygenation (Δ[HHb]), oxygen uptake (VO2), ventilation (VE), and arterialized blood lactate [La-] throughout exhaustive incremental (RAMP) and severe-intensity exercise (SI) during follicular (FOL) and luteal (LUT) menstrual phases. During RAMP, Δ[HHb]/VO2 was 65% lower in LUT below gas exchange threshold (p0.05). During SI,Δ[HHb]/VO2 was 18% lower in LUT during the kinetic phase (p0.05). No FOL/LUT differences in RAMP-performance (FOL: 218 ± 35 W; LUT: 221 ± 29 W) or SI-endurance (FOL: 99 ± 20 s; LUT: 96 ± 15 s) (p>0.05) were found. The purpose of this study was to determine the efficacy of implementing two Postural Restoration exercises and relating principles on deadlifts using a hexagonal barbell. Specifically, we aimed to examine the effects of the intervention on muscle activation and the location of the ground reaction force (GRF) center of pressure. Sixteen subjects (age 18-35 yrs.; male (n=14); female (n=2)), with at least 1 year of deadlifting experience, were randomly assigned into a control and intervention group (both n=8). Evidence suggests high-intensity exercise training increases incidence of upper respiratory infection in young-adult athletes (Spence et al. 2007). Collegiate athletes experience chronic stress, which has been shown to result in increased proportions of late-differentiated CD8+ T-cells of a dysfunctional phenotype (Bosch et al. 2009). However, many studies fail to consider possible moderators such as psychological stress and cytomegalovirus (CMV) infection. The pathophysiology of atherosclerotic cardiovascular disease (CVD) is highlighted by vascular dysfunction and low-grade vascular inflammation. Furthermore, the site-specific distribution of atherosclerosis throughout the arterial vasculature is primarily determined by local hemodynamic force. Therefore, this dissertation outlines three experiments designed to investigate the role of acute mental and physical (i.e., aerobic exercise), and vascular wall shear stress (SS) on the inflammatory aspects of atherosclerosis. Chapter 2 examines the effect of acute laboratory-induced mental stress on intracellular pro-inflammatory signaling pathways in peripheral blood mononuclear cells.
