Nematode Biodiversity In Lincoln, Nebraska's Tallgrass Prairie Corridor, Abigail Borgmeier Aug 2021

Nematode Biodiversity In Lincoln, Nebraska's Tallgrass Prairie Corridor, Abigail Borgmeier

Department of Plant Pathology: Dissertations, Theses, and Student Research

The tallgrass prairie was once one of the most diverse grasslands on the planet, however over 95% has been converted to agriculture in the past 150 years. The remaining prairie is still host to a wide variety of plants, insects, mammals, and soil-dwelling microorganisms, creating an incredibly complex ecosystem. The first aim of this study is to compare three approaches for the measurement of nematode diversity within a 10-mile-long protected prairie habitat corridor near Lincoln, Nebraska. The methods are 1) a traditional morphological analysis of 150 nematodes per study site, 2) a metabarcoding analysis using the 18S genetic marker of …

Harold W. Manter Laboratory of Parasitology: Faculty and Staff Publications

Sporadic and geographically widespread reports of parasites affecting the Belted Kingfisher (Megaceryle alcyon) have been published but few have described details of the pathology. A female, adult kingfisher was found dead in a heavily wooded area of a zoo in Maryland, USA. At necropsy, numerous sexually dimorphic, 4.4–40.5-cm adult Monopetalonema alcedinis nematodes were found tightly wound within the coelomic cavity between organs and completely filling the caudal thoracic and abdominal air sacs. Abundant, 30–60-mm diameter, larvated, thick-walled ova were found in the bronchi and parabronchi, within the mesentery, and in the serosa of multiple coelomic organs. Monopetalonema alcedinis …

Monophyly of the superfamily Dioctophymatoidea was assessed based on analyses of DNA sequence variation among 3 of 4 constituent genera (5 species). Represented is the first molecular phylogenetic evaluation of the Dioctophymatoidea using maximum parsimony, maximum likelihood, and Bayesian inference of 18S nuclear DNA (786 base-pair [bp] segment) and mitochondrial cytochrome oxidase 1 (293 bp) genes. Dioctophymatoidea is monophyletic and includes a clade with Dioctophyme renale and Eustrongylides ignotus (Dioctophymatidae) as the sister of Soboliphyme baturini, Soboliphyme jamesoni, and Soboliphyme abei (Soboliphymatidae). Within Soboliphymatidae, S. baturini is the sister of S. jamesoni and S. abei.

Fifty-eight individuals belonging to 10 species of bats, insectivores, and pikas were examined for helminths from 4 collection sites in Mongolia in 1999. Two species of bats (Vespertilio murinus and Eptesicus gobiensis) were infected with a single species of trematode (Plagiorchis vespertilionis), which represents a new record for the country. One individual of E. gobiensis also harbored 1 unidentified filaroid nematode. The acanthocephalan Moniliformis moniliformis was found in the hedgehog Hemiechinus auritus. Cestodes in the genus Catenotaenia and the herein described Schizorchis mongoliensis n. sp. were recovered from pikas belonging to the species Ochotona alpina …

Harold W. Manter Laboratory of Parasitology: Faculty and Staff Publications

One nine-banded armadillo (Dasypus novemcinctus) and 1 yellow armadillo (Euphractus sexcinctus) were necropsied in the field during an expedition to collect parasites of mammals in Bolivia. A total of 205 Aspidodera binansata Railliet and Henry, 1913 (Heterakoidea: Aspidoderidae), and 40 specimens of Lauroia bolivari n. sp. (Heterakoidea: Aspidoderidae) were recovered from the cecum and large intestines of D. novemcinctus and E. sexcinctus. Aspidodera esperanzae Fujita et al., 1995, is proposed as a junior synonym of A. binansata based on the structure of the cordons on the hood. Lauroia bolivari n. sp. has an undercut cephalic …

Identification And Characterisation Of A Cdna Sequence Encoding A Glutamic Acid-Rich Protein Specifically Transcribed In Trichinella Spiralis Newborn Larvae And Recognised By Infected Swine Serum, Dante S. Zarlenga, Patricia Boyd, J. Ralph Lichtenfels, Dolores Hill, H. Ray Gamble

Harold W. Manter Laboratory of Parasitology: Faculty and Staff Publications

Presently, little is known of the mechanism by which Trichinella penetrates and modulates reprogramming of muscle cells. In light of evidence demonstrating strong protective characteristics of antigens derived from this stage, understanding this process may shed light on potential targets for effective abatement of infection. To this end, a PCR-derived cDNA expression library was constructed using 0.5 mg of total RNA from Trichinella spiralis newborn larvae. The library consisted of >125,000 insert-containing clones. Approximately 40-50 × 103 clones were screened immunologically using sera from pigs experimentally infected with 7,000 Trichinella L1. Multiple clones reacting positively with the swine infection …

Sterol Composition Of The Corn Root Lesion Nematode, Pratylenchus Agilis, And Corn Root Cultures, David J. Chitwood, William R. Lusby

Harold W. Manter Laboratory of Parasitology: Faculty and Staff Publications

Sterols from mixed stages of the com root lesion nematode, Pratylenchus agilis, and uninfected corn root cultures were analyzed by gas chromatography-mass spectrometry. Twenty-eight sterols were identified in P. agilis, including nine not previously detected in nematodes. The major sterols were 24-ethylcholest-22-enol, 24-ethylcholesta-5,22-dienol, 24-methy1cholestanol, 24-ethylcholestanol, isofucostanol, and 24-ethylcholesterol. The principal corn root sterols were 24-ethylcholesta-5,22-dienol, 24-methylcholesterol, 24-ethylcholesterol, isofucosterol, and cycloartenol. Therefore, the major metabolic transformation of sterols by P. agilis was saturation of the sterol nucleus. In addition, very small amounts of 4α-methylsterols were biosynthesized by P. agilis. The 4-methylation pathway is unique to nematodes and was …
