During spaceflight, the immune system of astronauts becomes impaired due to microgravity, resulting in increased susceptibility to infections caused by pathogens such as Candida albicans. One of the most common clinical treatments for aggressive candidiasis is amphotericin B, a polyene antimycotic that targeted ergosterol in the fungal cell membrane and compromised the stability of the membrane. However, due to the chemical similarities between ergosterol and cholesterol, this treatment also affects the somatic cells of patients it is administered to. Entamoeba histolytica is a food-and-water-borne human protozoan pathogen that relies on a two-stage life cycle for successful proliferation outside of the host. E. histolytica exists in its dormant cyst form while outside of its host and converts to the motile trophozoite (amoeba) form in the small intestine. The trophozoites colonize the large intestine to cause symptomatic disease in ~10% of infections, resulting in ~100 million cases of amoebic dysentery each year. Trophozoites can convert back to the cyst form, which is shed in feces to continue the infection cycle.

Long-read single-cell RNA sequencing provides an opportunity to understand human health and disease at isoform resolution, revealing cellular diversity and disease mechanisms difficult to resolve with bulk or short-read methodologies.

Using a modified PIPseq workflow and computational pipeline adapted for Oxford Nanopore (ONT) sequencing, we profiled isoform usage across immune cells, integrating marker expression and isoform discovery, generating the largest long-read single-cell dataset of human immune cells from a single individual to date. We identified non-canonical protein-coding variants of GZMB and CD3G enriched in unexpected cell types. Apicomplexa constitute a large phylum of single-celled, obligate intracellular protozoan parasites. Notably, Plasmodium spp. and Babesia spp. are apicomplexan parasites that infect red blood cells. Plasmodium species are the causative agent of malaria and are transmitted by Anopheles mosquitoes, affecting large human populations, whereas Babesia spp., transmitted through the bite of Ixodes ticks cause babesiosis.

In this dissertation, we investigate the essential genome of these parasites using high-throughput transposon mutagenesis. African Sleeping Sickness is a Neglected Tropical Disease (NTD) which poses a significant health risk to impoverished populations in 36 Sub-Saharan African countries. It is a devastating disease caused by the protozoan parasite Trypanosoma brucei, which is transmitted to mammals via the bite of infected tsetse flies. The disease manifests in two stages: the hemolymphatic stage and the meningoencephalitic stage. In the latter stage, it invades the Central Nervous System (CNS) and can be fatal if left untreated. Equine Herpesvirus-1 (EHV 1) is a worldwide significant pathogen that causes respiratory illness, abortion, and neurological disorders in equine. Current vaccines, including live attenuated, inactivated, and subunit platforms, do not prevent viral latency, mucosal shedding, or cross-strain immunity, requiring alternative approaches. To address these gaps, this project explores the design, synthesis, and in vitro testing of an mRNA vaccine targeting immunogenic EHV 1 glycoprotein (gB, gC, gD, gG, and gM). Epitopes were computationally predicted using the Immune Epitope Database (IEDB), codon-optimized for Bos taurus, and cloned into pUCIDT vectors using SP6/T7 promoters. การคลอดก่อนกำหนด (ก่อน 37 สัปดาห์) เป็นปัญหาสาธารณสุขสำคัญทั่วโลก และในไทย ซึ่งเป็นสาเหตุหลักของการเสียชีวิตและภาวะแทรกซ้อนในทารกแรกเกิด การคลอดก่อนกำหนดส่วนใหญ่เกิดขึ้นเองโดย ไม่ทราบสาเหตุ จากหลักฐานก่อนหน้านี้พบว่า anti-HLA antibody อาจมีส่วนเกี่ยวข้องกับการคลอดก่อน กำหนดในประชากรชาวชิลี วัตถุประสงค์ของงานวิจัยจึงต้องการศึกษาความถี่และความสัมพันธ์ของแอนติบอดีต่อ Human Leukocyte Antigen class I ในหญิงตั้งครรภ์ชาวไทยที่คลอดก่อนกำหนด และสร้างโมเดลทำนายภาวะ คลอดก่อนกำหนดโดยใช้ปัจจัยเสี่ยง และผล anti-HLA class I กลุ่มตัวอย่างประกอบด้วยหญิงตั้งครรภ์ชาวไทยที่ คลอดที่โรงพยาบาลเจริญกรุงประชารักษ์จำนวน 299 คน แบ่งเป็นกลุ่มคลอดก่อนกำหนด 135 คน และกลุ่ม คลอดตามกำหนด 164 คน ได้รับการตรวจ anti-HLA class I ด้วยเทคนิค Multiplex Fluorescent Immunoassay และรวบรวมข้อมูลทางคลินิก ผลการศึกษาพบว่าความชุกของการตรวจพบ anti-HLA class I antibody โดยรวมอยู่ที่ 33.11% และสูงกว่าอย่างมีนัยสำคัญทางสถิติในกลุ่มที่คลอดก่อนกำหนด (42.96%) เมื่อ เทียบกับกลุ่มที่คลอดตามกำหนด (25.00%) การวิเคราะห์ความสัมพันธ์ด้วยสถิติ Chi-square test และ Logistic Regression พบว่าการตรวจพบ anti-HLA class I มีความสัมพันธ์อย่างมีนัยสำคัญกับการคลอดก่อนกำหนด (P < 0.001) โดยเพิ่มโอกาสในการคลอดก่อนกำหนดสูงขึ้น 2.260 เท่า นอกจากนี้งานวิจัยได้สร้างแบบจำลองการ ทำนายการคลอดก่อนกำหนดโดยใช้อัลกอริทึม Machine Learning หลายประเภท และใช้วิธีการคัดเลือกตัวแปร ที่แตกต่างกัน พบว่าแบบจำลอง LightGBM ที่ใช้การคัดเลือกตัวแปรแบบ Backward มีประสิทธิภาพสูงสุดในแง่ ของความแม่นยำ (Accuracy = 0.6018 ± 0.0482) และความสามารถในการตรวจจับการคลอดก่อนกำหนด (Sensitivity = 0.6444 ± 0.0726) ซึ่งแสดงให้เห็นว่า anti-HLA class I antibody และปัจจัยเสี่ยงบางประการ สามารถนำมาใช้เป็นตัวบ่งชี้เพื่อทำนายความเสี่ยงของการคลอดก่อนกำหนดได้ ผลการวิจัยนี้เป็นองค์ความรู้ พื้นฐานที่สำคัญในการพัฒนา biomarker เพื่อช่วยแพทย์ในการวางแผนการดูแลรักษาหญิงตั้งครรภ์ในอนาคต

โรคฟันผุเป็นโรคติดเชื้อเรื้อรังที่เกิดจากความไม่สมดุลของจุลชีพในช่องปาก โดยเฉพาะ Streptococcus mutans (SM) ซึ่งสามารถผลิตกรด สร้างไบโอฟิล์ม และทนต่อสภาวะเป็นกรดได้ การใช้โพรไบโอติกถูกเสนอเป็นแนวทางใหม่ในการควบคุมเชื้อก่อโรค แต่อาจมีประสิทธิภาพไม่เพียงพอ จึงเกิดความสนใจในการใช้พรีไบโอติกร่วมกัน งานวิจัยนี้มุ่งศึกษาผลของสารสกัดว่านหางจระเข้ (Aloe vera; AV) ในฐานะพรีไบโอติก ร่วมกับ Lacticaseibacillus rhamnosus GG (LGG) และ/หรือ Lactiplantibacillus plantarum 299V (LP) ในการยับยั้งคุณสมบัติการก่อโรคของ SM UA159 ในหลอดทดลอง ผลการทดลองพบว่า AV ที่ความเข้มข้นต่ำสุด 0.25% ซึ่งอุดมด้วยพอลิแซ็กคาไรด์ แสดงคุณสมบัติของการเป็นพรีไบโอติก โดยสามารถยับยั้งการเจริญของ SM ได้อย่างมีนัยสำคัญ (p < 0.05) และส่งเสริมการเจริญของโพรไบโอติกทั้งสองสายพันธุ์ (p < 0.05) การทำงานร่วมกันของ AV และโพรไบโอติกช่วยเพิ่มค่า pH ลดความเป็นกรด (p < 0.05) ลดการสร้างไบโอฟิล์ม (p < 0.05) และลดการแสดงออกของยีนสำคัญที่เกี่ยวข้องกับปัจจัยการก่อโรค ได้แก่ ยีนที่เกี่ยวข้องกับเมแทบอลิซึมและการสร้างสภาวะความเป็นกรด (eno, ldh, atpD) และยีนที่ควบคุมการสร้างไบโอฟิล์ม (gtfB, gtfC, dexA, และ dexB) ขณะเดียวกันเพิ่มการแสดงออกของ aguD (p < 0.05) ซึ่งเกี่ยวข้องกับระบบสร้างความเป็นด่าง ส่งผลให้เกิดสภาวะที่ไม่เอื้อต่อการเจริญของ SM นอกจากนี้ AV ยังเพิ่มการอยู่รอดของโพรไบโอติกผ่านการเพิ่มระดับยีน rpoB และ 16S rRNA (p < 0.05) ผลการทดลองสรุปได้ว่า AV ร่วมกับ LGG และ LP มีฤทธิ์เสริมกันในการลดความรุนแรงของ SM ผ่านหลายกลไกเชิงโมเลกุล พร้อมทั้งสนับสนุนการเจริญของจุลินทรีย์ที่เป็นประโยชน์ ซึ่งสอดคล้องกับแนวคิดการป้องกันโรคฟันผุในศตวรรษที่ 21 ที่มุ่ง “การปรับสมดุลของชุมชนจุลินทรีย์ในช่องปาก” ตาม Ecological Plaque Hypothesis และชี้ให้เห็นถึงศักยภาพของซินไบโอติกจากพืชสมุนไพรในการพัฒนาเป็นแนวทางใหม่เพื่อการป้องกันโรคฟันผุอย่างยั่งยืน

วัณโรค (TB) ซึ่งมีสาเหตุเกิดจากเชื้อ Mycobacterium tuberculosis (MTB) และโรคติดเชื้อกลุ่ม Non-tuberculous mycobacteria (NTM) เป็นโรคติดเชื้อที่มีความสำคัญซึ่งเกิดจากเชื้อกลุ่ม Mycobacteria ส่งผลกระทบต่อปอดและอวัยวะอื่นๆ ของร่างกาย แม้ว่าสาเหตุของการก่อโรคต่างกัน แต่อย่างไรก็ตามอาการแสดงมีความคล้ายคลึงกัน เช่น ไอ มีไข้ น้ำหนักลด และอ่อนเพลีย แนวทางในการรักษาวัณโรคและโรคติดเชื้อกลุ่ม NTM นั้นเป็นการรักษาโดยการใช้ยาต้านเชื้อ Mycobacteria หลายชนิดผสมผสานกัน แต่อย่างไรก็ตามแนวทางในการรักษาและระยะเวลาที่ใช้ในการรักษาแตกต่างกันระหว่างโรคดังกล่าว ดังนั้นวิธีการวินิจฉัยที่มีความรวดเร็วและจำเพาะในการจำแนกการติดเชื้อระหว่างสองกลุ่มจึงมีความจำเป็น เพื่อที่ผู้ป่วยจะได้รับการรักษาและการจัดการกับผู้ป่วยที่เหมาะสมต่อไป การศึกษาในครั้งนี้มีวัตถุประสงค์ในการพัฒนา Primer ในการจำแนกเชื้อกลุ่ม NTM จาก MTB และพัฒนาชุดตรวจแถบตรวจสารพันธุกรรมต้นแบบที่สามารถอ่านผลการทดสอบได้ด้วยตาเปล่าจากการเพิ่มประมาณสารพันธุกรรมที่อุณหภูมิเดียวซึ่งจำเพาะต่อยีน IS1081 และ ku ของเชื้อ MTB และ NTM ตามลำดับ แถบตรวจที่ผลิตพัฒนาขึ้นซึ่งผลิตจาก Nitrocellulose MTB/NTM strip สามารถตรวจจับผลผลิต Multiplex-recombinase polymerase amplification (M-RPA) ได้ โดยผลการทดสอบเบื้องต้นพบว่า Primer ที่ออกแบบขึ้นมานั้นสามารถเพิ่มปริมาณสารพันธุกรรมและจำแนก DNA ที่สกัดได้จากโคโลนีของเชื้อ MTB และ NTM ได้อย่างถูกต้อง โดยปฏิกิริยาบ่มที่อุณหภูมิ 37 ˚C เป็นระยะเวลา 30 นาที และเวลาในการอ่านผลของแถบตรวจ MTB/NTM strip คือ 15 นาที โดยตรวจสอบความเข้มข้นของเชื้อที่น้อยที่สุดที่ตรวจวัดได้ด้วยเทคนิค M-RPA ร่วมกับแถบตรวจ MTB/NTM strip สำหรับโคโลนีของเชื้อ MTB และ NTM คือ 1.97 x 104 CFU/mL และไม่พบปฏิกิริยาข้ามกลุ่มสำหรับเชื้อแบคทีเรียก่อโรคทั่วไปสายพันธุ์อื่น รวมถึงเชื้อกลุ่ม Higher bacteria เทคนิค M-RPA ร่วมกับแถบตรวจ MTB/NTM strip ได้นำมาประเมินกับ DNA …

Defense of mucosal tissues from microbial infection and allergy is reliant on continual production of antibodies. The aryl hydrocarbon receptor (AhR) is known to regulate B cell development and is associated with suppression of systemic humoral immunity. Recent attention has been paid to the role of the AhR in altering expression of cell adhesion molecules (CAMs). B cells express CAMs and chemokine receptors to migrate around the body for localized secretion of antibodies. Since the evolution of complex organisms, there have been communities of symbionts inhabiting them. Thanks to the development of parallel sequencing techniques, DNA of entire microbial communities can be sequenced from a single sample. Some microbes can be parasitic in nature, such as Haemosporida, an order of eukaryotes causing malaria. While most symbionts form a more commensal host relationship in wild systems, host-symbiont dynamics can vary in pathogenicity based on biotic and abiotic factors. Coral disease is one of the biggest challenges facing coral reefs that actively changes biodiversity resulting in coral decline. With the rising threat of diseases, corals require biomarkers that reflect the immune systems and differences between common coral tissue loss diseases to best assist in coral restoration efforts. Non-small cell lung cancers (NSCLC) constitute 80-85% of lung cancers and are the leading cause of cancer-related deaths globally. The most common cause is prolonged smoking. Current treatment options for NSCLC include surgery, radiation, chemotherapy, targeted drug therapy, and immunotherapy. ภาวะโรคไม่ติดต่อเรื้อรัง หรือ Non-Communicable Disease (NCDs) เป็นภาวะโรคที่ไม่ได้ติดต่อจากคนสู่คนได้รวมถึงไม่ได้เกิดจากการติดเชื้อโรค โดยสาเหตุหนึ่งของภาวะกลุ่มโรคนี้คือการเกิดภาวะ oxidative stress ที่ทำให้ร่างกายมีปริมาณของสารอนุมูลอิสระไม่สมดุล โดยที่ผ่านมาจำนวนผู้ป่วยในโรคนี้มีจำนวนเพิ่มขึ้นอย่างต่อเนื่อง ซึ่งแนวทางการรักษาส่วนใหญ่มักจะมาจากการรักษาตามอาการ ดังนั้นการแก้ปัญหาโรคนี้ให้ได้ผลมากที่สุดคือการป้องกันการเกิดโรคมากกว่าการรักษา ผู้วิจัยจึงสนใจที่จะนำมะกรูดที่มีสรรพคุณต้านอนุมูลอิสระมาทดสอบ โดยมีวัตถุประสงค์เพื่อศึกษาฤทธิ์ของสารสกัดจากใบมะกรูดและผิวมะกรูด ในการช่วยลดการเกิดภาวะ oxidative stress สำหรับการช่วยยืดอายุของ C. elegans ที่ถูกชักนำให้เกิดภาวะ oxidative stress โดยแบ่งการศึกษาออกเป็น 4 ขั้นตอน ได้แก่ ตอนที่ 1 ศึกษาฤทธิ์ของสารสกัดจากมะกรูดต่อ E.coli OP50 ที่เป็นอาหารของ C. elegans พบว่าสารสกัดจากใบมะกรูดและผิวมะกรูดไม่มีผลในการยับยั้งการเจริญของ E.coli OP50 เมื่อเทียบกับการยับยั้งการเจริญเติบโตของยา Penicillin-Streptomycin ที่ใช้เป็นกลุ่มควบคุม จึงสามารถใช้สารสกัดจากใบมะกรูดและผิวมะกรูดในการทดสอบได้ โดยที่ไม่มีผลต่อ E.coli OP50 ที่ใช้เป็นอาหารของ C. elegans ตอนที่ 2 การศึกษาฤทธิ์ของสารสกัดจากใบมะกรูดและผิวมะกรูดที่มีผลต่อการรอดชีวิตของ C. elegans ที่ถูกชักนำให้เกิดภาวะ oxidative stress พบว่าสารสกัดจากใบมะกรูดและผิวมะกรูดที่ความเข้มข้น 50 , 100 และ 200 ug/ml มีอัตราการรอดชีวิตของ C. elegans เมื่อเทียบกับ C. elegans ที่ไม่ได้ให้สารสกัดจากใบมะกรูดและผิวมะกรูด แตกต่างกันอย่างมีนัยสำคัญทางสถิติ 0.05 และตอนที่ 3 การศึกษาฤทธิ์ของสารสกัดจากใบมะกรูดและผิวมะกรูดต่อการยับยั้งปริมาณ ROS ของ C. elegans ที่ถูกชักนำให้เกิดภาวะ oxidative stress โดยพบว่าใบมะกรูดและผิวมะกรูดที่ความเข้มข้น 50 , 100 และ 200 ug/ml มีค่าปริมาณสาร ROS ของ C. elegans ที่ให้สารสกัดจากใบมะกรูดและผิวมะกรูด เมื่อเทียบกับ C. elegans ที่ไม่ให้สารสกัดจากใบมะกรูดและผิวมะกรูด …

Severe Acute Respiratory Syndrome Coronavirus -2 (SARS-CoV-2) has caused unprecedented morbidity and mortality worldwide. There are two pathophysiological hallmarks associated with severe patient outcomes after SARS-CoV-2 acute infection, namely delayed/weakened interferon production and overactive inflammatory responses. Our previous work has shown that these altered immune responses are due, at least in part, to significant alterations in 3D genome organization and epigenetic landscape following SARS-CoV-2 infection that are distinct from and more severe than changes caused by other viral infections or immune stimulants. Glioblastoma is the most commonly diagnosed central nervous system primary malignancy and it is considered a terminal diagnosis with few treatment options available. Glioblastoma tumors frequently develop treatment resistance due in part to their highly heterogenic nature. The heterogeneity of glioblastoma is partially attributed to the presence of glioma stem-like cells (GSC), which are highly invasive and resistant to chemotherapy and irradiation treatments. Salicylic acid (SA) is a central plant hormone mediating immunity, growth, and development. Recently, studies have highlighted the sensitivity of the SA pathway to changing climatic factors and the plant microbiome. Here we summarize organizing principles and themes in the regulation of SA biosynthesis, signaling, and metabolism by changing abiotic/biotic environments, focusing on molecular nodes governing SA pathway vulnerability or resilience. การดื้อยา colistin มีแนวโน้มเพิ่มสูงขึ้นในเชื้อแบคทีเรียกลุ่ม Enterobacterales เนื่องมาจากการนำยา colistin กลับมาใช้ในการรักษาเชื้อดื้อยาหลายขนาน โดยเฉพาะอย่างยิ่งเชื้อแบคทีเรียแกรมลบที่ดื้อยา carbapenem ประกอบกับการถ่ายทอดยีน mobile colistin-resistant (mcr) ผ่านทาง plasmid ซึ่งส่งผลให้การแพร่ระบาดของเชื้อดื้อยา colistin เกิดขึ้นในวงกว้างอย่างรวดเร็ว สถานการณ์เชื้อกลุ่ม Enterobacterales ที่ดื้อยา colistin ในประเทศไทยกำลังเป็นที่สนใจ เนื่องมาจากเริ่มพบอัตราเชื้อดื้อยาที่สูงขึ้นอย่างต่อเนื่องทั้งในภาคการปศุสัตว์และในโรงพยาบาล การศึกษาครั้งนี้ได้ศึกษาลักษณะจีโนมของเชื้อ E. coli และ K. pneumoniae ที่ดื้อยา colistin ที่เพาะแยกได้จากผู้ป่วยของโรงพยาบาลเลิดสิน กรุงเทพมหานคร ในช่วงระหว่างเดือนตุลาคม พ.ศ. 2564 ถึงเดือนกันยายน พ.ศ. 2565 และศึกษาความสัมพันธ์และการกระจายตัวของเชื้อในแผนกต่างๆ ของโรงพยาบาล เชื้อที่เก็บได้ทั้งหมดจำนวน 54 ตัวอย่าง ประกอบด้วยเชื้อ E. coli และ K. pneumoniae จำนวน 14 และ 40 ตัวอย่าง ตามลำดับ ซึ่งให้ผลดื้อยา colistin เมื่อทดสอบด้วยเครื่องอัตโนมัติ Sensititre™ Gram Negative MIC Plate เชื้อส่วนใหญ่เพาะแยกจากปัสสาวะ (46.30%, p-value=0.38) และเสมหะ (29.63%, p-value=0.63) โดยเพาะแยกเชื้อได้จากผู้ป่วยใน (81.48%) มากกว่าผู้ป่วยนอก (18.52%) อย่างมีนัยสำคัญ (p-value=0.000) ภายหลังนำเชื้อทั้งหมดไปวิเคราะห์ลำดับเบสด้วยเทคนิค Illumina whole genome sequencing (WGS) สามารถจำแนกสายพันธุ์ของเชื้อได้อย่างมีประสิทธิภาพ โดยเชื้อ E. coli และ K. pneumoniae มีรูปแบบยีนก่อโรคที่แตกต่างกันอย่างเห็นได้ชัด และพบยีน blaCTX-M, blaTEM, blaOXA ,blaSHV ,ยีน aac, ยีน qnr …

โรคหลอดเลือดแดงโคโรนารี (Coronary Artery Disease, CAD) เป็นโรคไม่ติดต่อชนิดเรื้อรังที่เกิดจากหลอดเลือดแดงที่กล้ามเนื้อหัวใจตีบหรือตัน สาเหตุหลักเกิดจากไขมันและเนื้อเยื่อสะสม มีผลทำให้เยื่อบุผนังหลอดเลือดชั้นในหนาตัวขึ้นและเกิดการอุดตัน จากการศึกษาที่ผ่านมาพบว่า ox-LDL เป็นจุดเริ่มต้นของโรคดังกล่าว เนื่องจากมีคุณสมบัติต่างจาก normal LDL โดยไม่สามารถจับได้กับ LDL receptor ได้ แต่สามารถจับได้กับ scavenger receptor ที่ผิวเซลล์แมคโคฟาจจึงทำให้ ox-LDL สามารถเข้าเซลล์ได้อย่างไม่มีสิ้นสุด เกิดเป็นโฟมเซลล์ (foam cells) สะสมอยู่ในผนังหลอดเลือดเกิดเป็นรอยขีดไขมัน (fatty streak) ประกอบกับการกระตุ้นเกล็ดเลือดและเซลล์อิมมูน ทำให้เกิดเป็นลิ่มเลือดอุดตันทางเดินของเส้นเลือด แม้ว่า ox-LDL จะเป็นสาเหตุสำคัญของการเกิดโรคแต่การตรวจวัดระดับไขมัน ox-LDL ยังไม่แพร่หลายในห้องปฏิบัติการเทคนิคการแพทย์ เนื่องจากชุดตรวจมีราคาแพง ใช้เวลาในการวิเคราะห์นาน และยังต้องใช้เครื่องมือที่มีความจำเพาะ ดังนั้นการศึกษาในครั้งนี้มีวัตถุประสงค์เพื่อพัฒนาเทคนิคการตรวจวัดระดับ ox-LDL ด้วยเทคนิคการตรวจวัดบนฐานกระดาษร่วมกับ immunogold silver enhancement โดยออกแบบกระดาษกรองให้มีรูปแบบเป็น microtiter plate อาศัยเทคนิค wax printing ในการสร้างขอบเขต หาสภาวะที่เหมาะสมสำหรับออกซิไดซ์กระดาษ เพื่อให้มีคุณสมบัติเหมาะสมกับการตรึงแอนติบอดี โดยเปลี่ยนเซลลูโลสเป็นหมู่แอลดีไฮด์ งานวิจัยนี้ได้นำสารละลายลิเธียมคลอไรด์มาช่วยเพิ่มประสิทธิภาพการออกซิไดซ์ร่วมกับสารละลายโซเดียมเพอร์ริออเดท พบว่ากระดาษที่ออกซิไดซ์แล้วสามารถคงสภาพนาน 72 ชั่วโมง และแอนติบอดีที่ตรึงไว้คงสภาพนานที่สุด 4 ชั่วโมง ทดลองวิเคราะห์วัดปริมาณ std. ox-LDL และ ox-LDL ที่เตรียมเองในห้องปฏิบัติการด้วยสภาวะเทคนิคดังกล่าว พบว่าสีปฏิกิริยาที่เกิดขึ้นยังไม่ชัดเจน อันเนื่องมากจากข้อจำกัดของความจำเพาะของแอนติบอดี การทำให้เกิดสีด้วย silver enhancement ซึ่งยังคงต้องมีการพัฒนาต่อไปเพื่อให้ผลการวิเคราะห์มีความชัดเจน นอกจากนี้ได้พัฒนากล่องถ่ายภาพเพื่อใช้เก็บผลการวิเคราะห์บนฐานกระดาษ ที่สามารถถ่ายภาพด้วยแสงและระยะที่คงที่ สามารถเคลื่อนย้ายได้ง่ายในภาคสนาม อย่างไรก็ตามแม้การพัฒนาวิธีการวิเคราะห์ ox-LDL ยังไม่ประสบผลสำเร็จ แต่ยังพอได้แนวทางและต้นแบบในการศึกษาต่อไปในอนาคต

Aspergillosis เป็นโรคติดเชื้ออันตรายถึงแก่ชีวิต มีสาเหตุมาจากเชื้อราในจีนัส Aspergillus การวินิจฉัยโรคในปัจจุบัน การเพาะแยกเชื้อและจัดกลุ่มตามลักษณะทางสัณฐานวิทยายังคงเป็นวิธีมาตรฐาน การตรวจหา galactomannan มักใช้ตรวจวินิจฉัยโรคในผู้ป่วยกลุ่มเสี่ยงเท่านั้น ในขณะที่เทคนิคทางอณูชีววิทยาอาศัยข้อมูลสารพันธุกรรมของ “its region” ในการจำแนกกลุ่มเชื้อระดับสปีชีส์ แต่อย่างไรก็ตามข้อมูลดังกล่าวถูกพิสูจน์ว่าสามารถจำแนกกลุ่มเชื้อราก่อโรคในคนในระดับสปีชีส์ได้ต่ำ ในด้านการรักษาสปีชีส์ของเชื้อ Aspergillus มักไม่ถูกนำมาพิจารณาร่วมเพื่อการเลือกกลุ่มยาแม้ว่ายังคงมีผู้ป่วยบางรายที่ตอบสนองต่อการรักษาไม่ดีเนื่องจากเชื้อสาเหตุดื้อต่อยาต้านเชื้อรา ด้วยเหตุดังกล่าวเพื่อศึกษาความสัมพันธ์ระหว่างสปีชีส์และรูปแบบความไวรับต่อยา การค้นหายีนที่สามารถจำแนกเชื้อ Aspergillus ในระดับสปีชีส์ได้อย่างแม่นยำจึงเป็นสิ่งจำเป็นยีน “tef-1α” เป็นสารพันธุกรรมที่ได้รับการพิสูจน์แล้วว่ามีคุณสมบัติสามารถจำแนกเชื้อราเส้นสายในระดับสปีชีส์ได้สูง ในงานวิจัยนี้ ผู้วิจัยได้เปรียบเทียบผลการจำแนกกลุ่มเชื้อ Aspergillus ที่แยกได้จากผู้ป่วยจำนวน 227 ตัวอย่าง โดยใช้ข้อมูลสารพันธุกรรม “its region” และ“tef-1α” จากผลการศึกษาพบว่าสารพันธุกรรมจากยีน“tef-1α” (D=0.8461) มีความสามารถในการจำแนกเชื้อในระดับสปีส์สูงกว่าบริเวณ “its” (D=0.796) โดยเฉพาะอย่างยิ่งใน section Fumigati ข้อมูลสารพันธุกรรมจาก “its region” สามารถแบ่งเชื้อกลุ่มดังกล่าวออกได้เพียง 1 สปีชีส์ คือ A. fumigatus ในขณะที่ยีน“tef-1α” สามารถแบ่งออกเป็น A. fumigatus และอีก 2 สปีชีส์ คือ A. lentulus และ A. udagawa ซึ่งเป็นเชื้อที่ดื้อต่อยา amphotericin B โดยกำเนิด โดยสรุป ผลจากงานวิจัยนี้ชี้ให้เห็นว่าการจำแนกสปีชีส์ของเชื้อ Aspergillus โดยใช้ยีนทุติยภูมิ “tef-1α” สามารถบ่งชี้กลุ่มยาที่เหมาะสมต่อการรักษาได้

RNA interference (RNAi) is a pervasive gene regulatory mechanism in eukaryotes based on the action of multiple classes of small RNA (sRNA). Exploiting RNAi pathways in non-model systems have great potential for creating potent RNAi technologies. Here, we accessed RNAi-mediated control of gene expression in the two-spotted spider mite, Tetranychus urticae (T. urticae) using engineered dsRNA designed to modulate the host RNAi pathway and increase RNAi efficacy. Analysis of Dicer (Dcr) generated fragments revealed how exogenous RNAs access the host RNAi pathway in this animal, opening avenues for designing RNAi technology for their control. Salmonella is a relatively abundant, virulent species of bacteria that is most known for spreading gastrointestinal diseases through food. These illnesses result in approximately 1.35 million infections, including over 25,000 hospitalizations each year, in the U.S. alone (CDC.gov). As antibiotic resistance becomes an increasingly urgent public health problem, the importance of developing alternative treatment methods is only becoming more crucial. One of the genes responsible for this virulence is known as hilA. HilA is the main transcriptional regulator of Salmonella Pathogenicity Island-1 gene (UniProt).
