Zinc And Manganese Effect Seed Quality And Germination In Common Bean (Phaseolus Vulgaris L.), Mehrdad Yarnia, Masoud Soltani̇ Bouljak, Parisa Bolouri, İnci̇ Tolay

Turkish Journal of Agriculture and Forestry

Zinc (Zn) and manganese (Mn) are important micronutrients in plant growth and development. In this study, the effect of Zn and Mn sulfate treatments at different growth stages of mother plants in common bean on seed production and germination potential was carried out. Results showed that zinc sulfate (ZnSo4) treatments significantly affected yield and pod number per plant compared to manganese sulfate tetrahydrate (MnSO4) treatments. Treatment in any form or at any stage of growth increased yield and its components in common bean. The common bean (Phaseolus vulgaris L.) is the most consumed food legume in the world and is a major source of dietary protein, carbohydrates, and valuable micronutrients, especially in developing countries. Diversity Arrays Technology (DArTseq), based on genome reduction with restriction enzymes, provides a rapid, high-throughput, and cost-effective tool capable of generating thousands of genotyped single nucleotide polymorphisms (SNPs) for a genome-wide analysis of genetic diversity. In this study, we aimed to characterize common bean accessions using SNPs detected by a DArTseq approach. This study evaluated seedling traits (shoot/root length, fresh and dry weights of shoots/roots, number of leaves, shoot diameter), phosphorus (P) content, mycorrhizal colonization (MC), and relative mycorrhizal dependency (RMD) of 21 common bean genotypes inoculated by 4 different arbuscular mycorrhizal fungi (AMF) [Glomus intraradices (Gi), Glomus mosseae (Gm), Gigaspora margarita (Gim), and commercial AMF (cAMF)] in a growth chamber under controlled growing conditions. The common bean (Phaseolus vulgaris L.) is an important crop throughout the world and is highly recommended for many developing countries due to its nutritional components. A retrotransposon-based DNA marker system, interprimer binding sites (iPBSs), developed from reverse transcriptase primer binding sites, was used to determine genetic diversity in common bean accessions. The genetic polymorphism and effectiveness of the marker system were evaluated. A total of 180 polymorphic bands were detected using 47 iPBS primers among 67 common bean accessions, with an average of 4 polymorphic fragments per primer. In this study, screening of 15 Phaseolus vulgaris cultivars and genotypes (Zülbiye, Kınalı, Perla, Kwintus, Özayşe, Tokat Sırık, Musica, Şelale, Nadide, Gina, Serra, Karabağ, Funda, and Lepus) and 1 Pisum sativum cultivar (Araka) for host suitability to 4 root-lesion nematodes, Pratylenchus thornei, P. crenatus, P. neglectus, and P. penetrans, was carried out in 2010 and 2011 under controlled conditions. Alternate planting combinations of maize (Zea mays L.) with common bean (Phaseolus vulgaris L.) or cowpea (Vigna sinensis L.) were compared with the solitary planting of each crop during 2003 and 2004 under the East Mediterranean conditions in Turkey. The objectives of this study were to determine the changes to some agronomic properties that significantly affect seed yield, and to conserve landraces of the common bean of Artvin province, especially in areas that will be flooded by dam projects, by collecting them before they are lost. Landraces of common bean were collected from 279 locations in 74 villages in 7 districts of the province.
