Consumer behavior toward food/beverages is influenced by multisensory attribute perceptions as well as emotional experiences. Traditional methods of sensory testing lack the ability to capture emotional responses and as a result, measuring food/beverage-evoked emotions remains a research challenge. There were three objectives of this dissertation study. Firstly, this study aimed to develop prediction models of acceptance of and preference for basic taste solutions using sensory attribute intensities and emotional responses. Sharks are harvested globally and sold in a variety of commercial products. However, they are particularly vulnerable to overfishing and many species are considered protected or endangered. The objective of this study was to identify species in various commercial shark products and to assess the effectiveness of three different DNA barcoding primer sets. Thirty-five products were collected for this study, including fillets, jerky, soup, and cartilage pills. DNA barcoding of these products was undertaken using two full-length primer sets and one mini-barcode primer set within the cytochrome c oxidase subunit (COI) gene. The antioxidant capacity of dried Agaricus bisporus mushrooms (DAB) in beef has previously been assessed. However, interactions between lipid oxidation products, mushroom polyphenols, and bovine proteins present in beef to explain the mushroom's antioxidative effect, has not been determined. Oven-dried or lyophilized DAB with and without 15 g NaCl/kg beef (1.5%) or 20 g NaCl/kg beef (2%) were added to sarcoplasmic protein homogenates from top round beef. Malondialdehyde and volatile aldehyde binding to sarcoplasmic protein (SP) were monitored. Oven dried had 64% higher total phenolic compared to lyophilized DAB, leading to ∼50% lower malondialdehyde content in beef with oven …

Candida albicans inhabits the gastrointestinal tract dormant commensal member but can become an opportunistic pathogen when the host microflora or immune system is compromised. Adhesion to a biological or synthetic surface, followed by a morphological change from the yeast to hyphae phenotype. Biofilm formation is becoming a common occurrence on types of medical devices. The primary objective of this study was to compare the anti-inflammatory effects of phenolic and volatile compounds extracted from cranberries. The Griess Reagent System assay was used to measure the in vitro anti-inflammatory capabilities of cranberry phenolic and volatile extracts on RAW 264.7 mouse macrophage cells. This study tested the anti-inflammatory capabilities of the cranberry phenolic and volatile extracts before, as a preventative treatment, and after, as a means of treating pre-existing inflammation, inducing inflammation with lipopolysaccharide (LPS). Inflammation is one of defense reactions of our body tissues against harmful stimuli such as infection and tissue injury. However, it has been reported that chronic inflammation can lead to chronic inflammatory diseases including diabetes, cardiovascular diseases and cancer. This transition from acute to chronic inflammation state is usually induced by pro-inflammatory mediators and cytokines including nitric oxide (NO), prostaglandin E2 (PGE2), interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α), which accelerate the development of other chronic diseases. Coffee continues to be one of the most widely consumed beverages worldwide. How an individual perceives a cup of coffee is impacted by a plethora of factors including origin, growing climate, roasting level, and consumption habits. This thesis utilized both trained descriptive panelists and untrained consumer panelists to analyze how serving/consumption temperature modulates sensory perception of brewed coffee in regards to appearance, aroma, flavor, taste, and mouthfeel. The study discussed herein looks both to continue and to expand upon previous work conducted here at Seton Hall University, which investigated the effect of cold plasma processing on sweet basil and their essential oils. It was found that the application of cold plasma treatment increases plant growth and eugenol concentration, which is an essential oil component and one of the potential natural antioxidants used in food preservation. Among plant based agricultural products, Nebraska ranks first nationwide in the production of Great Northern beans (GNB), second in proso millet production, and eighth in production of winter wheat. The present research was focused on the effect of processing on nutritive components and in vitroprotein digestibility of these crops with the aim of promoting their human consumption. Proso millet based extrudates had physical properties similar to commonly extruded rice but had lower expansion than corn. The desire for performance enhancement is a common goal amongst athletes. Recently a number of studies have reported various performance enhancing effects of dietary nitrate (NO3-) via beetroot juice (BRJ) supplementation. Research has indicated that the various performance enhancing effects of BRJ can be attributed to a decrease resting systolic blood pressure (SBP), diastolic blood pressure (DBP), and submaximal volume of oxygen consumption (VO2), while increasing vasodilation and mitochondrial efficiency. Epidemiological studies have shown an association between high carrot consumption and low prevalence of cancer. This observation has been thought to be attributed to carrot carotenoids. Despite this, various intervention trials have displayed no changes in incidence or increased incidence of cancer with carotenoid supplementation. Quality control is crucial for any brewery and especially important in satellite breweries that need to imitate the standard set by the parent brewery. Zinc, magnesium, iron, sodium manganese, and calcium are trace elements found in wort and are essential for enzymes that catalyze fermentation reactions. Ulcerative colitis (UC) is a disabling inflammatory bowel disease. UC is characterized by chronic, relapsing inflammation of the colon and rectum. Current treatments such as monoclonal antibodies against TNF-α, IL-6, IL-12/p40, adhesion molecules, Janus kinases (JAK) inhibitors have side effects or lose their effects over time. Alternative approaches with fewer side effects for patients are needed. Montmorency tart cherries (Prunus cerasus) are a good source of anti-inflammatory flavonoids. Brown seaweeds contain a number of bioactive compounds. The xanthophyll, fucoxanthin, has in vivo efficacy against disorders such as type 2 diabetes, obesity and cancer. Organic solvents are traditionally employed to extract fucoxanthin, but carry a toxic chemical and environmental burden. The aim of this study was to optimise a fucoxanthin extraction method using enzymes, water, low-temperature dehydration and mechanical blending, to produce yields comparable to those achieved with an organic solvent (acetone). Ergot alkaloids, in their active isomeric form, affect animal health and performance, and adsorbents are used to mitigate toxicities by reducing bioavailability. Adsorbents with high specificity (molecularly imprinted polymers: MIP) adsorb ergot alkaloids in vitro, but require evaluation for biological implications. Using ex vivo myography, synthetic polymers were evaluated for effects on the bioactivity of ergotamine tartrate (ETA). Polymers were first evaluated using isotherms. Lateral saphenous veins were collected from 17 steers for four independent studies: dose response of ETA, adsorbent dose response, validation of pre-myograph incubation conditions and MIP/ non-molecularly imprinted polymer (NIP) comparison. Smoking is a global epidemic that creates serious health and economic burden. It is the primary, preventative factor for the majority of causes of death worldwide. Analysis of publicly available data revealed that smoking prevalence rates among the youth in developing countries, especially in Bulgaria, are alarmingly high. The Bulgarian population has one of the highest percentages of smokers and the second highest rate among teenage girls. Lowering of greening formed from oxidized chlorogenic acid (CGA) and amino groups, and favoured at alkaline pH, was investigated using acidic ingredients (sour cream, buttermilk, yoghurt, and honey) in sunflower butter cookies. Cookies with maple syrup added were used as a positive control. Changes in greening intensity, greening reactants (total phenols, CGA, protein), antioxidant capacity, tryptophan and Schiff base fluorescence were measured. Percentage greening, pH and aw of cookies followed the same order: maple syrup> sour cream ≥ buttermilk> yoghurt> honey. pH was positively correlated with greening intensity (r=0.77) and negatively correlated with CGA (r=-0.96). Effects of curcumin, a biologically active ingredient of turmeric, were tested on the Ca2+transients induced by the activation of α7 subunit of the human nicotinic acetylcholine (α7nACh) receptor expressed in SH-EP1 cells. Curcumin caused a significant potentiation of choline (1 mM)-induced Ca2+ transients with an EC50 value of 133 nM. The potentiating effect of curcumin was not observed in Ca2+ transients induced by high K+ (60 mM) containing solutions or activation of α4β2 nACh receptors and the extent of curcumin potentiation was not altered in the presence of …

In this study, a novel galactomannoglucan named as TJ2 was isolated from Agaricus brasiliensis with microwave extraction, macroporous resin, ion exchange resin and high resolution gel chromatography. TJ2 is composed of glucose, mannose and galactose in the ratio 99.2:0.2:0.6. Infrared spectra (IR), methylation analysis and nuclear magnetic resonance spectra indicated that TJ2 mainly contained a b-(1?3) – linked glucopyranosyl backbone. Interestingly, TJ2 significantly promoted RAW264.7 cell proliferation, and was able to activate the cells to engulf E. coli. Blackberries are grown worldwide for commercial fresh markets. Three Arkansas-grown fresh-market blackberry genotypes (‘Natchez’, ‘Ouachita’, and A-2418) were evaluated for consumer sensory and compositional attributes at the University of Arkansas Food Science Department, Fayetteville. The compositional attributes of the blackberries were within an acceptable range for commercial markets (soluble solids=8.20-11.90%, pH=2.79-3.18, titratable acidity=1.09-1.32%). In terms of soluble solids to titratable acidity ratio, ‘Ouachita’ (10.92) had the highest ratio, followed by ‘Natchez’ (8.93) and A-2418 (6.25). Cricket (Acheta domesticus), silkworm pupae (Bombyx mori), and locust (Locusta migratoria) dry flours were obtained to examine the biochemical properties and composition of the flours. This study aimed to characterize the protein and lipid components of three insect species utilizing proximate composition analyses, amino acid composition analysis, protein solubility, SDS-PAGE, fatty acid composition analysis, and thin layer chromatography to determine lipid classes, and lipid extraction efficiency. Kjeldahl determined the cricket, locust, and silkworm flours contained 72.0%, 53.1%, and 71.2% protein, respectively. All proximate composition analyses were significantly (p< 0.05) different between species. Amino acid composition analysis revealed that the flours contained 3.6-3.9% and 0.90-1.5% of lysine and methionine, respectively. Many countries have cereal grains and legumes as a staple in their diet that contain these limiting amino acids. Essential amino acids were 22% of total amino acids. Protein solubility revealed that the three species studied were most soluble in alkaline environments with highest protein solubility occurring at pH 13 at 66% solubility in silkworm. Lowest solubility occurred in more acidic conditions between pH 4-5. SDS-PAGE revealed five major protein fractions with estimated molecular weights of 27 (cuticle proteins), 41 (arginine kinase), which has been identified as an allergen in shrimp, 42 (actin), 71 (hemocyanin), and 220 myosin) kDa. Soxhlet extraction determined cricket, silkworm, and locust flours contained 15.4%, 33.3%, and 11.4% lipid, respectively. The only omega-3 fatty acid found in the flours was α-linolenic acid. Silkworm flour contained the most α-linolenic acid at 33.3% of total fatty acids, followed by locust (13.7%), and cricket (0.6%). TLC resolved lipids for typical lipid classes. The four major lipid classes noted were triacylglycerol, free fatty acid, cholesterol, and phospholipid. Lipid extraction efficiency found that the organic solvents chloroform and methanol had the highest lipid extraction yield in both cricket (69.3%) and locust (93.0%). Methyl-tertiary-butyl ether (MTBE) extracted lipids most efficiently in the silkworm. Proteins and lipids found in insects need to be isolated to further or commercial use.

The effects of dietary ingredients on the proteome profile of postmortem beef longissimus lumborum (LL) muscle were evaluated. In the first experiment, the influence of dietary ractopamine on the whole-muscle proteome of beef LL was examined. Five proteins were differentially abundant between ractopamine-fed (RAC) and non-ractopamine fed (CON) groups. The differentially abundant proteins were over-abundant in RAC and were related to muscle structure development (F-actin-capping protein subunit beta-2 and PDZ and LIM domain protein-3), chaperone (heat shock protein beta-1), oxygen transportation (myoglobin), and glycolysis (L-lactate dehydrogenase A chain). These findings indicated that ractopamine influences the abundance of proteins associated with …
