Pathogenic bacteria are still a threat to human beings worldwide as their presence is often unknown even if they are already present in fresh produces. This in turn resulted in several outbreaks and recalls across the United States. This dissertation investigated the survival of pathogenic bacteria and their potential non-pathogenic surrogates after antimicrobial interventions in different modes of applications on the surface of cantaloupes, butternut squash, and fresh cilantro. Edible insects are emerging as sustainable and nutritious alternatives to conventional protein sources. However, concerns about food safety, particularly the presence of pathogenic bacteria such as Escherichia coli and Listeria monocytogenes, remain a barrier to consumer acceptance and industrial use. Yellow mealworm (Tenebrio molitor) larvae have gained attention as a sustainable and nutritionally balanced food source, yet their high natural bacterial load raises food safety concerns. This study investigates the impact of pre-processing, processing, and storage conditions on microbial load and diversity in mealworms, comparing thermal (hot) and non-thermal (cold) treatments. These treatments include freeze-drying, wet heat treatments, dry heat treatments, and the combination of wet and dry heat treatments against untreated controls. Foodborne illnesses are a global public health challenge. Foodborne illnesses, like norovirus, often have a large disease and societal burden. Norovirus is a single-stranded, positive-sense RNA virus, with 10 identified genogroups (GI-GX). Genogroups GI and GII are responsible for nearly all human cases of acute gastroenteritis worldwide. In the United States, HuNoV is not notifiable case-by-case. Instead, outbreaks are reported to national surveillance systems when two or more cases are epidemiologically linked. Beyond The Byproduct: Exploring Approaches For The Sustainable Valorization Of Underutilized Food Streams, Ian Phillip Israelsen

Commercial food processing creates side-streams that are often underutilized yet hold significant potential for valorization into value-added products for consumer use. This study investigated and compared potential protein recovery and lipid removal potential from buckwheat (BW; Fagopyrum esculentum Moench), brewer’s spent grain (BSG), and coffee silverskin (CSS) using a one-step solvent defatting and a pH-shift protein extraction method. The extraction efficiency yields of extracted protein from CSS were significantly lower than all other samples (p < 0.05). The defatted BW had the highest extracted protein content of all samples (p < 0.05). The sizes of the proteins in the starting materials were observed using LDS-PAGE. These sizes were consistent with published values and were mostly unchanged by defatting or the pH-shift process. The presence of 13S globulins and 2S albumins was confirmed under reducing conditions. Protein extracted from all native samples had lower total free essential amino acids than the starting materials. Native CSS and extracted protein followed this trend, but in defatted CSS and respective extracted protein samples, the extracted proteins had higher free essential amino acid content than the starting materials, sometimes of twofold or greater (p < 0.05). After defatting, the percentage of lipids recovered compared to the original lipid content of the samples was measured. The crude fat content of native and hexane-defatted BW flour was higher than BW flour defatted with ethanol (p < 0.05). The lipid extraction efficiency of all samples ranged from 4.6 ± 1.26%–43.20 ± 14.21%. The mass of the native and defatted starting materials were recorded at each step of processing, with reasonable losses assessed. The major non-polar lipid classes in each starting material were separated using thin-layer chromatography. All samples also displayed the presence of polar lipids. When comparing starting materials, BW and BSG was the most effective for protein extraction, and BSG has the most promise for lipid removal and potential for upcycling. All the examined starting materials show promise for use in food supplementation, especially as native flours. These products are plentiful and considered underutilized or side-stream products.

During the feedlot receiving period, newly weaned beef cattle face various stressors, including weaning, transportation, vaccination, and exposure to pathogens. Additionally, changes in diet and low dry matter intake contribute to nutrient deficiencies, heightening susceptibility to disease and performance declines. Therefore, we examined the effects of dietary supplementation with a multicomponent blend of prebiotics and probiotics on the health, immune status, metabolism, and performance of newly weaned beef steers during a 35-day receiving period. The purpose of this experiment is to determine the thermal kinetic parameters of Salmonella in moisture-enhanced, reconstructed ground chicken patties as affected by temperatures and salt concentrations. Salmonella is responsible for 35% of the foodborne illnesses associated with poultry products (Batz et al., 2011). Microwave-assisted deep eutectic solvent and gamma-valerolactone metallic chloride catalyzed conversion of xylan to furfural were investigated using a 2x3 factorial experimental design at two levels of percent microwave power, reaction time, and catalyst concentration. The levels of each factor studied were (20%, 60% microwave power, 2, 4 minutes, and 10, 20mg) respectively. The effect of three metallic chloride catalysts (LiCl, FeCl3.6H20, CuCl2) on the conversion of xylan to furfural was also investigated. Refined Liquid Smoke (RLS) (1%) was incorporated during osmotic dehydration of apple cubes (10 mm each side) to observe its effect on solid gain, water loss, yield, water activity, product quality, and consumer preference. Three different treatments, POD (pure osmotic dehydration, no RLS), LSISS (RLS inside sugar solution) and PTLS (pre-treatment with RLS), were compared in this study. Osmotic dehydration was carried out using 42 °Brix sugar solution, and the fruit to solution ratio was kept at 1:4. There is a growing concern regarding the microbial safety of farmers’ markets as fresh produce consumption in the United States is gaining popularity. The CDC reported 46% of foodborne illnesses are related to fresh produce. Consuming fresh produce raw removes a processing step, increasing the risk of cross-contamination, leading to illness. However, this risk can be mitigated by washing produce with antimicrobial solutions during post-harvest processes. Nanotechnology takes advantage of cellular biology’s natural nanoscale operations by interacting with biomolecules differently than soluble or bulk materials, often altering normal cellular processes such as metabolism or growth. To gain a better understanding of how copper nanoparticles hybridized on cellulose fibers called carboxymethyl cellulose (CMC) affected growth of Saccharomyces cerevisiae, the mechanisms of toxicity were explored. Multiple methodologies covering genetics, proteomics, metallomics, and metabolomics were used during this investigation. The work that lead to this dissertation discovered that these cellulosic copper nanoparticles had a unique toxicity compared to copper. During the late 19th and very early 20th centuries widespread deforestation occurred across the Appalachian region, USA. However, since the early 20th century, land cover rapidly changed from predominantly agricultural land use (72%; 1909) to forest. West Virginia (WV) is now the USA’s third most forested state by area (79%; 1989–present). It is well understood that land cover alterations feedback on climate with important implications for ecology, water resources, and watershed management.
