I grew up contemplating the concept of life. From my Catholic grandmother to my pediatrician mother, I always received mixed perspectives on the hot topics of abortions, vaccines, and embryo use. I found that the controversy lies in an ambiguous, mixed understanding of what constitutes a human life. From biochemical approaches to extraterrestrial life forms and spirituality there is no clear, coherent all-encompassing definition of life. However, that does not mean we can merely agree to disagree. The concept of a human life has drastic implications on our everyday lives. In vitro produced (IVP) bovine embryos provide enhanced genetic value and a means for studying early embryonic development. However, IVP embryos are developmentally compromised and are lower quality compared to in vivo derived (IVD) embryos. Consequently, after transfer pregnancy rates from IVP embryos are consistently lower, as they exhibit the highest amounts of pregnancy failure during the early embryonic phase (prior to day 28). Progesterone (P4) has been shown to influence bovine endometrial epithelial (EPI) and stromal fibroblast (SF) cells to secrete developmentally important molecules that promote early embryo development and conceptus elongation. Obesity-induced hyperlipidemia is one of the main factors for female infertility. Hyperlipidemia, specifically with high levels of palmitic acid (PA) and oleic acid (OA), interferes with preimplantation development. Autophagy is essential in early embryo development but, it is unknown whether hyperlipidemia affects autophagic mechanisms in preimplantation embryos. It was hypothesized that PA will alter autophagy in preimplantation mouse embryos and that the subsequent effects will be reversed by OA. PA impaired blastocyst development by arresting embryos at the 8-cell stage. PA also elevated early embryo autophagy by increasing autophagosome formation, decreasing maturation, and disrupting degradation. Co-treatment with OA showed developmental …

Pregnancy loss remains a major source of economic cost to the equine industry. Frequently, the exact causes of pregnancy loss remain unknown. It has been shown, in other species, that increased dietary protein leading to elevated blood urea nitrogen concentrations (BUN) can be a factor in decreased survival of the early embryo. Our studies provided novel information regarding the effects of elevated BUN on endometrium and embryos from mares as well as insights on changes in their gene expression. Changes to the embryonic environment that occur within the first few days of development can have significant effects on subsequent fetal development. The following studies investigate the effects of progesterone-deprivation during the early preimplantation period in the mare. The first study described embryo morphology (diameter, stage, and quality grade) collected from mares with induced aluteal cycles. The second study evaluated the effect of induced aluteal cycles on the future fertility of embryo collections as a potential method for cycle manipulation of embryo donor mares. Every year more than 75,000 pregnant women are exposed to teratogenic medications or general anesthesia during non-obstetric surgery in the US,1-4 and embryonic effects of general anesthesia are of particular interest in laboratory research and veterinary medicine. The mouse system is used to screen potential toxic effects of anesthetics used in egg retrieval for in vitro fertilization (IVF), or those of pharmacologic agents which may come in contact with the egg or early embryo. Elevated ambient temperatures negatively impacts pregnancy rates in agriculturally important females. An oocyte undergoing meiotic maturation exposed to a physiologically-relevant heat stress results in alterations of developmentally important processes and reportedly decreases subsequent developmental potential after fertilization. To address problems of reduced competence after exposure to heat stress for the first 12 h of meiotic maturation (hIVM), Payton (2009) assessed the transcriptome profiles of oocytes and associated cumulus cells by microarray analysis and reported heat-induced perturbations at the molecular level in the oocyte. Embryo cryopreservation is an integral part of assisted reproduction because it allows for future use of these embryos. Cryopreservation occurs when there are supernumerary embryos or when an embryo transfer cannot be performed.
There are two main methods to cryopreserve embryos. The most recent is vitrification, which uses high concentrations of cryoprotectants, a short time to cool and avoids ice crystals. The 'gold standard' is the slow-cool method, which uses low concentrations of cryoprotectants, a long time to cool embryos and produces extracellular ice crystals.

The focus of this study was to elucidate the molecular and cellular mechanisms whereby fibroblast growth factors (FGFs) mediate outgrowth of the limb. Specifically, we examined the epistatic relationship between FGF and Wnt/Planar cell polarity (PCP) signaling in establishing cell polarity as a mechanism for outgrowth. By implanting beads into embryonic limbs and lateral plate mesoderm, we established that FGF activates Wnt5a in a gradient fashion. Once it was established that Wnt5a was expressed at the right time and place to turn on PCP signaling, we investigated the ability of Wnt5a to influence cell migration and/or cell polarity. Leukemia inhibitory factor (LIF) is a pleiotropic cytokine which demonstrates perplexing physiological effects. It has been demonstrated that LIF is essential for implantation in mice. Little is known relating to the manner by which LIF effects pre-implantation and post-implantation development. The objectives of this project were to determine the effects LIF on pre-implantation development, to determine the effects that it may have on implantation rates, successful pregnancy rates, and resorption rates, and to determine the effects that LIF has on the skeletal development of mice. The upper optimum temperature for embryonic development of mountain whitefish is 6 C, and for the post yolk sac stage is 9 to 12 C. The need to know effects on all stages in the life cycle in cases of thermal pollution is demonstrated. Abnormalities caused by thermal pollution in hatched larvae were agape jaws, coloboma or fissure of the eye, monophthalmia or the presence of only one eye, monomicrophthalmia or the presence of one small eye and one case of twinning. High mortalities of eggs occurred at 9 C and higher.
