Idiopathic pulmonary fibrosis (IPF) is a progressive interstitial lung disease characterized by fibroblast activation, excessive extracellular matrix (ECM) deposition, and irreversible respiratory decline. In this study, we investigated the role of myocardin and its downstream effectors, RGS5, DLGAP5, and TAGLN, in thrombin-induced myofibroblast differentiation and ECM remodeling using primary human IPF fibroblasts. Myocardin knockdown significantly attenuated thrombin-induced expression of profibrotic genes, including FN1, COL1A1, PAI-1, ACTA2, and CNN1, as well as the novel effectors RGS5, DLGAP5, and TAGLN, confirming myocardin as a central regulator of fibrotic signaling. Autoimmune disorders disproportionately affect females, a bias largely attributed to estrogen's modulation of B lymphocyte development and tolerance. Bisphenol A (BPA), a common plastic monomer, shows estrogenic activity and acts as an endocrine disruptor and epigenetic modifier. This thesis examines how BPA may increase autoimmune risk by mimicking estrogen such that it can excite estrogen receptors which rescue autoreactive B lymphocytes from apoptosis. While direct research linking BPA to B lymphocyte-mediated autoimmunity is limited, understanding BPA’s course of action in B lymphocytes is critical for addressing the rising prevalence of autoimmune diseases and its possible link to the increased and …

Endometriosis is a disease characterized by pelvic pain and the formation of endometrial-like tissue on the outside of the uterine lining. This condition affects nearly 10% of women across the globe and confirming whether someone has endometriosis requires visualization in surgery. While the specific causes of this disease remain unclear, the over-expression of the Galectins 3 and 9 in ectopic endometrial tissues suggests a role in the development and maintenance of the condition. Pleural fibrosis involves excessive extracellular matrix (ECM) deposition driven by chronic injury and inflammation. Transforming growth factor-β (TGF-β) induces mesothelial–mesenchymal transition (MesoMT), promoting a myofibroblast phenotype. Our laboratory identified myocardin (Myocd), a transcriptional coactivator, as a key mediator of MesoMT that regulates ECM-remodeling proteases. RNA-seq and qPCR analyses showed that ADAM12 and ADAM19 are upregulated by TGF-β but suppressed by Myocd knockdown in Human pleural mesothelial and IPF-derived cells. Silencing ADAM12 reduced fibrotic markers, including fibronectin and smooth muscle actin (α-SMA), whereas ADAM19 showed relatively minor effects. Peripheral Artery Disease (PAD) is characterized by reduced blood flow, ischemic injury, and fibrosis, which collectively impair tissue function. Current treatment strategies are limited in their ability to target fibrotic tissue and restore normal function effectively. This study evaluates the therapeutic potential of drug-loaded nanoparticles to reduce fibrosis in ischemic tissue. Nanoparticles were synthesized and characterized for in vivo application. Tissue samples from experimental groups, including saline control, blank nanoparticles, free drug, and drug-loaded nanoparticles, were analyzed using Masson’s Trichrome staining to quantify collagen deposition. Chronic diseases, such as inflammatory bowel diseases, require patients’ commitment to their treatment, which can be challenging over time. Edible plants could help address this challenge by acting as living factories that produce a range of drugs, including medicinal proteins. Patients could eat the plants because their cells have a sturdy outer layer that protects and helps deliver the medicine. Lettuce is an example of a common vegetable that can serve this purpose. Pleural injuries can disrupt the organization of the pleural surface, leading to pleural remodeling and fibrosis (PF), characterized by the proliferation of myofibroblasts expressing alpha-smooth muscle actin (α-SMA) and the deposition of extracellular matrix proteins, primarily due to the mesenchymal transition of pleural mesothelial cells (PMCs) (MesoMT). It is well established that various external stimuli, such as TGF-β, thrombin, and factor Xa (FXa), induce MesoMT. Our laboratory found that myocardin, a transcriptional co-activator, was dramatically up-regulated during MesoMT, and that gene silencing of myocardin (MyoCD) diminished MesoMT. Adipose-derived stromal vascular fraction (AD-SVF) is a promising autologous source for tissue engineering. Curcuma zedoaria Rosc. (white turmeric) is a rhizomatous medicinal plant in the Zingiberaceae family, widely used in traditional Asian medicine. In recent years, increasing scientific attention has focused on its diverse pharmacological properties and bioactive constituents. This mini review summarises current knowledge of the phytochemical composition and biological activities of C. zedoaria, with particular emphasis on its antimicrobial, antioxidant, anti-inflammatory, and anticancer effects. The sustainability of global palm oil production faces significant threats from a range of diseases and infections affecting palm oil crops. This study presents a comprehensive bibliometric analysis to map the scientific landscape and evolution of research related to palm oil diseases over the past decade. Using data from the Scopus database, 366 relevant documents were analyzed using R and VOSviewer to uncover trends in publication volume, citation impact, prolific authors, country contributions, and keyword co-occurrence networks. This project aims to develop a transgenic plant cell system that can produce a pharmaceutical protein using molecular cloning techniques. CRISPR-Cas9 is a biotechnology tool adapted from a bacterial defense mechanism to precisely edit DNA using a guide mRNA and a Cas9 protein. This project aims to create CRISPR-CAS9 mutations in the APETALA3 (AP3) gene of the model plant Arabidopsis thaliana. AP3 is a class B gene critical to the petal and stamen development of Arabidopsis flowers. Endolysins are proteins that degrade bacterial cell walls. They are encoded on the genome of bacteriophages (viruses that infect and kill bacteria). At the end of the lytic cycle of a bacteriophage, endolysins cut the peptidoglycan layer of the bacterial cell wall, allowing the release of viral progeny. Pseudomonas aeruginosa is a well-known multidrug-resistant human pathogen that causes serious problems, especially in patients diagnosed with cystic fibrosis (CF). The purpose of this research is to verify the mutation in the Arabidopsis thaliana APETALA3 (AP3) gene. By sequencing DNA, we confirmed the presence of an extra guanine in the mutant AP3 gene. These findings support that further investigation is needed into how the AP3 gene affects flower development in Arabidopsis thaliana.

Over the past decade, fungal research and its technological applications have expanded across multiple disciplines, including the emerging field of biohybrid systems. This thesis develops and evaluates a wireless, untethered mobile robot controlled by the action potential-like activity generated by Pleurotus ostreatus sporocarps under red, green, and blue optical stimulation. Light is applied to the sporocarps, the resulting electrical responses are recorded, and these signals are transmitted wirelessly to actuate the mobile robot. Both the action potential-like activity patterns and the robot’s movement trajectories were analyzed. Glioblastoma multiforme is the most common adult primary tumor with a grim median survival time of 14.6 months. It is characterized by rapid progression, diffuse infiltration, and rather poor clinical outcomes. The WHO classifies GBM as a grade 4 diffuse astrocytic tumor with distinctive histopathological features including high mitotic activity, marked cellular atypia, microvascular proliferation, and necrosis. Anaerobic ammonium oxidation (anammox) is the biological conversion of ammonium to nitrogen gas (N2) using nitrite as an electron acceptor. This study evaluated the performance of a membrane bioreactor (MBR) in nitrogen removal at room temperature using Candidatus Brocadia fulgida, an anammox bacterium previously cultivated from Lake Koto Baru, Tanah Datar, Indonesia. The MBR was operated for 48 days with a hydraulic retention time of 24 h under a continuous supply of ammonium and nitrite, with both substrates provided at influent concentrations of 70, 150, and 250 mg-N/L. Pediatric acquired subglottic stenosis is a high-risk airway condition that requires close coordination between anesthesiology and otorhinolaryngology teams, particularly during endoscopic interventions performed under shared-airway conditions. This report describes the interdisciplinary management and anesthetic–surgical coordination in a child with severe acquired subglottic stenosis undergoing endoscopic balloon dilation, highlighting key decision points and airway management strategies. A 2-year-old boy with a history of prolonged and repeated endotracheal intubations presented with persistent dyspnea and noisy breathing, consistent with fixed extrathoracic airway obstruction.
