A Review On The Cytogenetic Integrity Of Micropropagated Plants [Review Paper], Eliaza Alumbro Peniton Jr Jun 2025

A Review On The Cytogenetic Integrity Of Micropropagated Plants [Review Paper], Eliaza Alumbro Peniton Jr

The Philippine Agricultural Scientist

Cytogenetic stability is essential for the survival of micropropagated plants. While micropropagation allows rapid propagation of plants with desirable phenotypes, it may also produce genetic variations that are, at times, cytogenetically observable. Factors affecting these variations are not fully understood, and micropropagation protocols are optimized to produce genetically stable regenerants. The genetic stability of micropropagated plants is traditionally assessed by comparing the phenomorphological traits of the regenerants to the mother plant. However, genetic variation does not often result in obvious morphological changes. Therefore, molecular and cytogenetic markers are typically used to evaluate the genetic variations of regenerants. Moreover, analyzing plant …

Effects Of Fibrogenic And Non Fibrogenic Nanomaterials On Cellular And Molecular Responses In Thp-1 Macrophage, Adedayo E. Oyerinde May 2025

Effects Of Fibrogenic And Non Fibrogenic Nanomaterials On Cellular And Molecular Responses In Thp-1 Macrophage, Adedayo E. Oyerinde

Biotechnology Theses

Traditional toxicological testing of nanomaterials is often low-throughput and time-consuming. Here, we aimed to develop an economically viable in vitro assay to prescreen nanomaterials with fibrogenic potential. To this end, we tested the generation of reactive oxygen species (ROS) as well as cytokines (pro- and anti-inflammatory) by macrophages following treatment with fibrogenic and non-fibrogenic nanomaterials. Both classes of nanomaterials induced ROS indistinguishably, suggesting that ROS production doesn’t predict nanomaterial fibrogenicity. We observed higher levels of IL1β and IL6 in macrophages treated with fibrogenic nanomaterials. Analysis of single-cell RNA-Sequencing data of fibrotic lungs from rodents and humans revealed overexpression of Brain …

Engineering Hiv-1 Env Protein To Elucidate In Situ Structural Dynamics Of Env, Narendra Kumar Gonepudi May 2025

Engineering Hiv-1 Env Protein To Elucidate In Situ Structural Dynamics Of Env, Narendra Kumar Gonepudi

Biotechnology Theses

The HIV-1 Envelope (Env) glycoprotein trimers undergo structural changes upon interacting with host receptor CD4 and coreceptors (CCR5/CXCR4) for virus entry. As the only surface-exposed viral protein, Env is a key antibody target but evades recognition through structural changes. Studying Env dynamics with respect to the coreceptor binding region (V3 loop) provides insights into virus entry and immune evasion. Fluorescence labeling via genetic code expansion (amber–TAG suppression) offers a minimally invasive approach for fluorescence microscope-based Env dynamics studies. Here, two V3-related dual-amber Env constructs (A135TAG-P308TAG, P308TAG-E395TAG) were generated by incorporating two non-canonical …

Myocardin Effector Proteins Affect Tgf-Beta-Induced Myofibroblast Differentiation In Ipf Cells, Allison Gonzalez

Biotechnology Theses

Idiopathic pulmonary fibrosis (IPF) is an incurable interstitial lung disease defined by the progressive, irreversible scarring of the tissues between the lung’s alveoli. To date, only two drugs have received approval for treatment. However, these medications are not considered cures since they do not stop the progression of the disease. A deeper understanding of the mechanisms underlying IPF could aid in discovering a cure. We recently found that myocardin (MyoCD) regulates SMAD2/3 transcription factors in the TGF-β signaling pathway and governs the genes involved in pleural fibrosis. However, it is unclear whether MyoCD plays a critical role in IPF by …

Rowan-Virtua Research Day

Cerium oxide nanoparticles (CNPs) are recognized for their ability to scavenge reactive oxygen species (ROS), making them promising candidates for mitigating oxidative damage in biomedical applications. However, many conventional synthesis methods limit the formation of Ce³⁺ states, which are critical for this antioxidant function. Building upon previous findings that glucose can promote Ce³⁺ formation, this study investigates the broader impact of both reducing (glucose, fructose, galactose) and non-reducing (dextran, sucrose, cyclodextrin) natural sugars on CNP synthesis. CNPs were synthesized in the presence of these sugars, and the surface densities of Ce³⁺ were measured. Results showed that reducing sugars significantly increased …

Antimicrobial Eats: How Food Additives Are Contributing To Obesity Via The Gut Microbiome, Olivia Mankos May 2025

Antimicrobial Eats: How Food Additives Are Contributing To Obesity Via The Gut Microbiome, Olivia Mankos

Senior Honors Theses

The typical modern diet is brimming with food additives used in food processing. Additives such as antibiotic residues, GMOs, pesticide residues, anti-caking agents, emulsifiers, artificial sweeteners, colorants, flavor enhancers, and preservatives. Most of these additives seem to shift the gut microbiome towards more pro-inflammatory signals. When all these additives are considered as an amalgam, such as with the modern processed diet, the ramifications on the gut microbiome are concerning. Obesity, which is characterized by low-level chronic inflammation, is likely exacerbated by these food additives. Consequently, most of these additives should be avoided or banned for optimal health.

School of Medicine Faculty Publications

The thoracic dystrophies are inherited skeletal conditions where abnormal embryonic development of the thoracic skeleton results in a narrow chest, pulmonary hypoplasia, and respiratory insufficiency, which can be severe or lethal. The majority of thoracic dystrophies are due to biallelic alterations in genes needed for normal ciliary function. However, despite the identification of over 20 genes as causal for the thoracic dystrophy phenotype, around 20% of patients remain without a molecular diagnosis. We present two unrelated families with a clinical diagnosis of thoracic dystrophy with associated respiratory insufficiency without a molecular diagnosis on previous genetic testing. Both harbor rare biallelic …

Metabolite Analysis To Understand Factors Limiting Microbial Fermentation At High Substrate Concentrations, Bishal D. Sharma Apr 2025

Metabolite Analysis To Understand Factors Limiting Microbial Fermentation At High Substrate Concentrations, Bishal D. Sharma

Dartmouth College Ph.D Dissertations

Clostridium thermocellum, a thermophilic anaerobic bacterium, holds promise as a biocatalyst for conversion of lignocellulosic biomass into biofuels. This microbe has been subjected to various metabolic engineering strategies, including the deletion of genes encoding secondary fermentation products such as H₂, formate, acetate, and lactate, as well as the expression of heterologous genes to redirect carbon flux toward ethanol production. Despite these efforts, achieving economically viable ethanol titers remains a challenge. We hypothesized that studying how intracellular metabolite concentrations change as fermentation stops could reveal metabolic bottlenecks or regulatory mechanisms limiting ethanol production at high titers. However, metabolomics studies at …
