The Ethics Of Brain-Machine Interfaces, Devon J. Lynn Jan 2024

The Ethics Of Brain-Machine Interfaces, Devon J. Lynn

Honors Undergraduate Theses

Brain Machine Interfaces (BMI) are a rapidly developing technology that raise unique ethical issues that demand review. They have demonstrated impressive restorative potential, particularly for individuals living with epilepsy, and those who are locked in. Although BMIs have the potential to provide significant benefit to millions of users, further advancement of the technology should proceed cautiously, according to the guidelines outlined in this paper. Failure to adhere to ethical guidelines could lead to severe privacy concerns, and would violate moral principles of beneficence, virtue ethics, care ethics, and utilitarianism. Despite the moral risks, BMIs hold promise for reshaping future healthcare …

Further Optimization Of A Simplified Protocol For Tooth Sample Preparation For Analysis Using The Applied Biosystems™ Rapidhit™ Id Instrument, Yen H. Dang

Master of Science in Forensic Science Directed Research Projects

Due to the destructive nature of mass casualty incidents (MCIs) or mass fatality events (MFEs), tooth samples are often one of the only viable sources for Disaster Victim Identification (DVI) because of the incredible integrity of the enamel layer surrounding the tooth. However, the resilient structure of the tooth requires harsh chemicals and a labor-intensive sample preparation protocol to ensure successful STR profile development. Considering this difficulty, one of the promising resolutions is the Applied Biosystems RapidHIT™ ID ID platform, which is a fully automated system that is designed to perform cell lysis, DNA isolation, polymerase chain reaction (PCR) amplification, …

Examining The Effects Of Artemisia Annua On Neuro2a Cells, Wesley Lim Jan 2024

Examining The Effects Of Artemisia Annua On Neuro2a Cells, Wesley Lim

Honors Undergraduate Theses

Artemisia annua L. is a well-known medicinal herb used in Traditional Chinese Medicine for many centuries and contains artemisinin, which exhibits antimalarial properties [1] and is a potential treatment for SARS-CoV-2 [2]. However, the neurological effect of A. annua and its compounds are poorly understood. We hypothesize that A. annua extracts and artemisinin will provide protection against oxidative stress in mouse neuronal Neuro2A cells. Superoxide dismutase 1 (SOD1) is an anti-oxidant enzyme that protects against oxidative damage [3]. Growth Arrest and DNA Damage-inducible 45 alpha (Gadd45a) is involved in DNA repair and is a common biomarker for DNA damage[4]. SOD1 …

Hybridization Kinetics Of Four-Way Junctions Localized On A Dna Scaffold, Katherine N. Taylor Jan 2024

Hybridization Kinetics Of Four-Way Junctions Localized On A Dna Scaffold, Katherine N. Taylor

Honors Undergraduate Theses

DNA computing is an ever-growing field with scientists trying to design structures that optimize logic gate communication to develop fast, biologically compatible, computational structures. We hypothesize that by using the principles of DNA computing, it is possible to design a DNA tile capable of studying localized DNA hybridization that can differentiate between oligonucleotides of different structural conformations. This includes synthetically manipulating DNA into a nanostructure that can perform Boolean logic functions to calculate the different rates of hybridization. To test our hypothesis, we designed a DNA Tile that incorporated a 4WJ using YES logic. Linear and hairpin single-stranded (ss) DNA …

A Tool To Automate Neuropathological Assessment In Huntington Disease Mouse Models, Samuel A. Moldenhauer Jan 2024

A Tool To Automate Neuropathological Assessment In Huntington Disease Mouse Models, Samuel A. Moldenhauer

Honors Undergraduate Theses

As our life expectancy continues to rise, so does the prevalence of neurodegenerative diseases, such as Huntington disease (HD). Neurodegeneration leads to progressive regional brain atrophy, typically initiating prior to symptom onset. With the advancements of medical treatments designed to target neurodegeneration, researchers measure their impact on atrophy in animal models to assess their effectiveness. This is important because treatments designed to combat neuropathology are more likely to modify the disease itself, per contra to treatments designed to mask or treat symptoms. One method of brain region size quantification is magnetic resonance imaging (MRI), which while accurate, is prohibitively expensive. …

International Arab Journal of Dentistry

Introduction: Limited research has explored the mechanical characteristics of recently introduced multilayered monolithic zirconia (MZC) crowns with feather-edge margins design. Moreover, most of the conventional techniques for evaluating the mechanical behavior of brittle dental ceramics rely on destructive tests, making it impossible to precisely evaluate real strength values due to premature crack initiation detection failure.

Objectives: The main objective of the study was to assess the load capacity of translucent multilayered monolithic zirconia crowns with feather-edge margin thicknesses of 0.4 mm and 0.5 mm. Additionally, the research introduced a novel non-destructive acoustic emission testing (AET) technique in the …

Nudcl2 Is Required For Cytokinesis By Stabilizing Rcc2 With Hsp90 At The Midbody, Xiaoyang Xu, Yuliang Huang, Feng Yang, Xiaoxia Sun, Rijin Lin, Jiaxing Feng, Mingyang Yang, Jiaqi Shao, Xiaoqi Liu, Tianhua Zhou, Shanshan Xie, Yuehong Yang

Markey Cancer Center Faculty Publications

Cytokinesis is required for faithful division of cytoplasmic components and duplicated nuclei into two daughter cells. Midbody, a protein-dense organelle that forms at the intercellular bridge, is indispensable for success- ful cytokinesis. However, the regulatory mechanism of cytokinesis at the midbody still remains elusive. Here, we unveil a critical role for NudC-like protein 2 (NudCL2), a co-chaperone of heat shock protein 90 (Hsp90), in cytokinesis regulation by stabilizing regulator of chromosome condensation 2 (RCC2) at the midbody in mam- malian cells. NudCL2 localizes at the midbody, and its downregulation results in cytokinesis failure, multinu- cleation, and midbody disorganization. Using iTRAQ-based …

Mechanistic Analysis Of Four-Way Dna Junctions And Cytokine-Binding Aptamers For Therapeutic Interventions, Roaa S. Mahmoud

Theses and Dissertations

DNA is inherently dynamic and topologically diverse and can fold into many different structures. Besides the canonical Watson-Crick structure, other higher-order structures such as G-quadruplexes (G4), i-motifs (iM), and four-way DNA junctions are possible. Although these high-order DNA structures are known to form transiently, they are important due to the crucial roles they play in many cellular processes including DNA replication, recombination, and repair. Among these DNA structures, 4-way junctions (also known as Holliday junctions, HJ) which are formed during the repair of double-strand DNA breaks (DSBs) and interact with proteins have garnered significant attention due to their central role …

Exploring A Medicinal Strain Of White-Rot Fungus For Biofuel Applications, Kortney L. Lucius Jan 2024

Exploring A Medicinal Strain Of White-Rot Fungus For Biofuel Applications, Kortney L. Lucius

Masters Theses

The major limitation to producing biofuels from lignocellulosic biomass, such as miscanthus straw, is the difficulty in accessing fermentable carbohydrates encased in lignin. Traditional methods for separating lignin are economically and environmentally costly. Biological pretreatment using white-rot fungi, such as Trametes versicolor, has been demonstrated to potentially reduce the need for harsh chemical and physical pretreatment methods. Despite these efforts, the process of converting lignocellulose to ethanol remains financially challenging. To explore a potential co-product for the process, this study evaluated the efficacy of T. versicolor strain CM-101, known for producing the valuable medicinal macromolecule polysaccharide-K (PSK), as a …

Large T Antigen Mediated Target Gene Replication Improves Site-Specific Recombination Efficiency, Zening Wang, Chuan Chen, Xin Ge Jan 2024

Large T Antigen Mediated Target Gene Replication Improves Site-Specific Recombination Efficiency, Zening Wang, Chuan Chen, Xin Ge

Faculty, Staff and Student Publications

With advantages of high-fidelity, monoclonality and large cargo capacity, site-specific recombination (SSR) holds great promises for precise genomic modifications. However, broad applications of SSR have been hurdled by low integration efficiency, and the amount of donor DNA available in nucleus for SSR presents as a limiting factor. Inspired by the DNA replication mechanisms observed in double-stranded DNA virus SV40, we hypothesized that expression of SV40 large T antigen (TAg) can increase the copy number of the donor plasmid bearing an SV40 origin, and in consequence promote recombination events. This hypothesis was tested with dual recombinase-mediated cassette exchange (RMCE) in suspension …

Converting skin by-product of unicornfish (Naso reticulatus Randall, 2011) into a valuable commodity like collagen is a good strategy to explore alternative sources for land-based animals. Additionally, marine fish collagens are preferable due to their safer and great properties. This project aimed to compare extractability of collagens prepared with three organic acids (i.e., acetic, lactic and citric acids), and their characteristics were also discussed. Collagen extracted with citric acid (CSCUS) (7.10%) had a significantly higher yield (p

This study aimed to assess the soaking behaviour of MR 297 dried paddy under two conditions: room temperature and hot water soaking at 60°C. The experiment involved soaking dried MR297 paddy samples in water for 0.5 to 8 hours at room temperature and 0.5 to 5 hours for soaking at 60°C. The water absorption of MR 297 behaved differently at two different soaking conditions. The kinetic behaviour of the soaking process was also simulated using established 25 predictive models and incorporating 5 novel models presented in this work. The comparison of all prediction models involved using three statistical measures: the …

This study investigates the release behaviour of Butterfly Pea (BP) anthocyanins from Sago starch micro-composite (SMC) films in the presence of food stimulants. To comprehensively describe the release profiles of BP from SMC, six well-established kinetic release models were employed which includes Korsmeyer-Peppas, zero-order, first-order, second-order, Higuchi, and Hixson-Crowell models. These models played a crucial role in examining how the release fraction changes over time, facilitating a comprehensive understanding of the release kinetics of BP anthocyanins. Our data analysis revealed a strong conformity with the Korsmeyer-Peppas model, suggesting its applicability in describing the release of BP anthocyanins from SMC. The …

The composition of fats and oils significantly impacts the flavour, nutrition, and appearance of food. Excessive consumption of saturated fatty acids, primarily found in animal fats, raises the risk of coronary heart disease. To address this, plant-based fats are used as substitutes, with microalgae offering unique nutritional benefits as a fat source. This study evaluates the performance of structured plant-based fats, incorporating microalgae (Aurantiochytrium sp. SW1), as a fat substitute in chicken patties. Four formulations were tested: 0% (control), 5%, 7%, and 10% structured microalgae-based fat. Colour, texture, water-holding capacity, cooking loss, and microstructure in both raw and cooked samples …
