Bone cancer is an aggressive disease and has peak occurrence during physiological stimulation of growth and aging. Astronauts who undergo long-term space missions also acquire an increased risk of bone tissue degeneration and cancer. Few in-vitro models currently exist capable of reproducing the complex microenvironment of bone tissue to support multicellular activity in a three-dimensional structure. This limits the ability to understand disease progression and develop suitable treatment strategies. Identification of the mutations and genetic causes of the muscular diseases is performed for the muscular dystrophies, atrophies and other genetic diseases using current molecular high throughput techniques that precisely allocate the defected genes and detect the aberration in the expressed proteins responsible for any phenotype. Somatic stem cells have been used as a regenerative medicine tool to treat diseases like leukemia for decades, but they are limited in their ability to self-renew and differentiate. A better alternative, induced pluripotent stem cells (iPSCs), has the ability to generate cells from all three germ layers and avoid tissue rejection as well as bypass the ethical concerns related to embryonic stem cells. In 2006, Takahashi and Yamanaka discovered that only four genes were required to induce pluripotency: Oct4, Sox2, Klf4, and c-Myc (OSKM). The ability of RNA to store genetic information and to catalyze biochemical transformations led to the speculation of the existence of RNA world before the evolution of contemporary ribonucleoprotein (RNP) world. RNA sequencing (RNAseq) can be used to compare gene expression between different groups, identify mutations, and confirm gene knockouts. In RNAseq, RNA is extracted and built into a sequenceable library to analyze the transcriptome. Ribosomal RNA (rRNA) makes up roughly 90% of the total RNA extracted and does not provide useful transcriptome information, thus it must be removed in order to perform transcriptional analysis. At MBP Titan, the protocol used KAPA’s RiboErase Depletion kit using custom methanotroph oligonucleotides to specifically bind to rRNA for depletion. Distributed Bio uses computational immunology techniques to generate antibody libraries against a target and bio-panning to identify and select clones that best interact with the target. This study characterized the growth of bacteria isolated from ballast tank fluids in hexadecane, petroleum, plant, and algae-derived fuels. The study was performed to explore the capacity of ballast tank isolates to survive and grow within fuels that may be stored within ballast tanks. Results of the hexadecane analysis indicated that most isolates had higher viable cell counts in media supplemented with hexadecane. Members of Alteromonas, Pseudoalteromonas, and a single Brevundimonas species had viable cell counts that were one or several orders ofmagnitude greater than that of controls. In invertebrates gap junctions are formed by the innexin family of proteins. Remarkably, the genome of Hydra magnipapillata contains 17 innexin genes. This study focused on Hydra innexin-2 (h-Inx2) which is expressed in nerve cells and plays a role in contraction of the body column. The gene sequence of H-Inx2 was obtained from the National Center for Biotechnology Information (NCBI), the gene was synthesized externally and transferred to a vector suitable for expression in Xenopus oocytes (pcDNA3.1 CT-GFP TOPO). Invasion of Amur honeysuckle (Lonicera maackii) into riparian areas of headwater streams creates strong potential for alterations of terrestrial subsidies that may drive bottom-up effects on aquatic biota. In this study, we analyzed effects of L. maackii on terrestrial subsidies in stream sites that represented a gradient of invasion intensity in temperate deciduous forests of southwestern Ohio (USA). Leaf litter biomass, terrestrial and aquatic fine woody debris (0.5–9.9 cm diameter) volume and count, and terrestrial and aquatic coarse woody debris (>9.9 cm diameter) volume were analyzed. High-throughput forward genetic screenings are invaluable tools to systematically explore genetic interactions and to link gene disruption with disease contexts. The adaptation of CRISPR/Cas9 has improved the sensitivity and specificity of functional screenings. Despite this advance, there remains a long-standing need to improve functional screenings with smaller and more versatile pooled libraries. Capitalizing on the inherent multiplexing capability of a class 2 CRISPR enzyme AsCpf1, we developed a multiplexed, high throughput screening strategy that has avoided the usual trade-off between library size and library penetration, allowing library minimization without sacrificing gene targeting efficiency. Following an action potential in the presynaptic neuron there is evoked release of neurotransmitter into the synapse which activates ionotropic transmembrane receptors on the postsynaptic membrane that cause depolarizations in voltage that get recorded as excitatory postsynaptic potentials (EPSPs). In the absence of an action potential there is spontaneous release of neurotransmitter that postsynaptically gets recorded as miniature excitatory postsynaptic potentials (mEPSPs). The impact of humans on landscapes may differ across regions because anthropogenic intervention on the respective landscapes occurs in different human-induced contexts. This study was designed to assess the human-induced landscape evolution of the Citanduy Watershed, Java, Indonesia, and determine its anthropogenic signatures. Several approaches were used, namely anthropogenic landscape identification, anthropogenic soil development, sediment analysis, and monitoring of changes to Segara Anakan lagoon at the mouth of the Citanduy Rier. Comparison Of 3d Coral Photogrammetry And Coral Video Transect For Coral Lifeform Analysis Using Low-Cost Underwater Action Camera, Ahmad Zuhairi Bin, Mohd Jinah Muhammad Idzham Helmi Bin, Saad Shahbudin Bin

This research analysed the use of 3D Coral Photogrammetry (CP) and Coral Video Transect (CVT) images collected from SCUBA divers using a low-cost underwater action camera to examine the coral lifeform. A comparison was made between data sets obtained using both methods on nine transects with different coral lifeform compositions and percentage cover within an area of 4 × 7 m. The comparison of the statistical analysis for CPCe revealed that there were no significant differences (p < 0.05) between CP and CVT photos where dead corals (p = 0.006), sand (p = 0.011), and unknown (p = 0.002) are present. Additionally, the coral value (p = 0.131) between CP and CVT was not significant. CP was capable of producing prominent branching, massive, and plate coral morphology results. This suggests that survey methods using low-cost action cameras for 3D Coral Photogrammetry would yield appropriate results in terms of coral lifeform detection. Hypothetically, by improving camera quality, it will yield a higher accuracy of 3D coral images that are suitable for use in scientific research and management. Other benefits of using CP include the possibilities for future studies with 3D coral surveys using remotely operated vehicles, less field time, and 3D coral seabed information.

The adsorption kinetics of water from an azeotropic mixture of isopropyl alcohol and water using chemically activated natural zeolites with and without a binder (starch) was investigated. In addition, an investigation of the compressive strength of zeolite pellets was conducted to ascertain the performance of the adsorbent for long-term operation. Three parameters were applied in the zeolite-making process: the particle size of zeolite (20, 30, 50, and 80 mesh), the sintering temperature (550, 750, and 1,000°C), the compaction pressure (2, 4, and 6 tonnes), and the starch-to-zeolite weight ratio (0, 1:3, 1:5, and 1:7). Magnetosomes are intracellular magnetic nanocrystals composed of magnetite (Fe3O4) or greigite (Fe3S4), enveloped by a lipid bilayer membrane, produced by magnetotactic bacteria. Because of the stability of these structures in certain environments after cell death and lysis, magnetosome magnetite crystals contribute to the magnetization of sediments as well as providing a fossil record of ancient microbial ecosystems. The persistence or changes of the chemical and magnetic features of magnetosomes under certain conditions in different environments are important factors in biotechnology and paleomagnetism. The following thesis shall primarily explore the current research regarding the implementation of modifiable microscopic and nanoscopic polymer particles as novel pharmaceutical delivery systems. Polymethacrylate-based copolymers, such as EudragitTM, are sensitive to alterations in pH levels, becoming increasingly more soluble in response to an increase in pH. The selective solubility and modifiable nature of these polymers allows for greater flexibility of treatment options for patients (Patra et al., 2017). Durian ‘Kamajaya’ merupakan salah satu jenis durian lokal yang keberadaanya hampir punah sehingga perlu dilakukan konservasi, salah satunya yaitu dengan perbanyakan menggunakan kultur jaringan. Permasalahan yang muncul dalam kultur jaringan durian ini salah satunya adalah metode sterilisasi. Penelitian ini bertujuan untuk memperoleh metode sterilisasi yang tepat untuk perbanyakan tanaman durian ‘Kamajaya’ secara in vitro. Acute Myocardial infarction (AMI) is the leading cause of death worldwide. Diagnosis of AMI depends on presenting symptoms, electrocardiogram (ECG), and cardiac troponins (cTns). Troponins are not good markers for patients presenting at the first hour of chest pain, so there is still a need for a marker that can be detected within the first hour of presentation, with high specificity and sensitivity, guide the medical decisions and give good insight on the expected prognosis. In the Midwestern United States, there is a strong management impetus toward reforestation to replace trees lost to due to tree mortality and to establish forests on previously cleared properties. Here, we describe a public outreach and volunteer effort that aimed to generate positive, community-engaged restoration outcomes and report on a practical experiment involving seed handling. We obtained tree seeds that were donated by members of the community to local land managers. An in vitro study was planned to assess the effects of a homofermentative microbial inoculant on the fermentation parameters and nutritive value of corn silage. The inoculant was applied at concentrations of 5x104cfu/g of forage (T1), 1x105cfu/g of forage (T2) 1.5x105cfu/g of forage (T3) and a negative control group (T0) without bacterial inoculant in three replicates each. At day 3, 7, 45 and 90 of the experiment individual buckets were opened to characterize the material, quick acidification, dry matter recovery, and aerobic stability of silage respectively.
