The investigation in this paper aimed to describe periphytic diatom assemblage, species composition and distribution in tropical polluted Linggi (sensu stricto or s.s.) and Kundor rivers in Negeri Sembilan, Malaysia. Regardless of ecological and environmental conditions, diatoms were the numerically dominant flora among periphytic algae collected monthly over a period of 13 months at nine stations in the Linggi (s.s.) and Kundor rivers. The freshwater periphytic diatom samples were collected mainly using artificial substrates and supplemented with natural substrates. A team of healthcare professionals in the Department of Emergency and Medical Services demonstrated decision-making processes for a poisoned pediatric patient with a Do-not-resuscitate (DNR) order. At the same time, clinical guidelines for poison resuscitations were critically applied. Lastly, the case examined and evaluated emergency care, management, treatment and diagnostic laboratory tests for pediatric poisoned patients. Ideas on pertinent literatures of decision-making models, clinical guidelines on poison resuscitation and DNR policies were already known by the majority. The IDH1 mutation is an important diagnostic and prognostic biomarker used to characterize glioblastoma (GBM). Patients harboring the IDH1 mutation have improved overall survival following maximal resection. Knowledge of the IDH1 mutation status allows the surgeon to modify the surgical plan; however, no existing molecular test can provide this information intraoperatively. We designed a novel colorimetric peptide nucleic acid loop-mediated isothermal amplification (PNA-LAMP) method that rapidly detects the IDH1 R132H mutation in GBM. PNA-LAMP amplifies target DNA under isothermal conditions with high specificity and speed. The PNA prevents amplification of wild-type IDH1 DNA, while allowing amplification of the R132H variant …

The Dual Contouring algorithm (DC) is a grid-based process used to generate surface meshes from volumetric data. However, DC is unable to guarantee 2-manifold and watertight meshes due to the fact that it produces only one vertex for each grid cube. We present a modified Dual Contouring algorithm that is capable of overcoming this limitation. The proposed method decomposes an ambiguous grid cube into a set of tetrahedral cells and uses novel polygon generation rules that produce 2-manifold and watertight surface meshes with good-quality triangles. Background: We present our experience with optimization and diagnostic use of quantitative real-time polymerase chain reaction (PCR) targeting the lytA gene of Streptococcus pneumoniae for the detection of S. pneumoniae in whole blood of children ≥5 CFU/10 μl or 1 copy of DNA/2 μl of blood.
Methods: This assay was applied on 1912 whole blood specimens collected from children pneumonia, of which 35 specimens were lytA positive. The bacterial loads were determined through categorization of load into five different categories, i.e., very high load, high load, moderate load, low load, and very low load.

Processing of next-generation sequencing (NGS) data requires significant technical skills, involving installation, configuration, and execution of bioinformatics data pipelines, in addition to specialized postanalysis visualization and data mining software. In order to address some of these challenges, developers have leveraged virtualization containers toward seamless deployment of preconfigured bioinformatics software and pipelines on any computational platform. We present an approach for abstracting the complex data operations of multistep, bioinformatics pipelines for NGS data analysis. As examples, we have deployed 2 pipelines for RNA sequencing and chromatin immunoprecipitation sequencing, preconfigured within Docker virtualization containers we call Bio-Docklets. The changes in the global environmental conditions have placed great challenges on governments, industries, and societies. Business organizations are often blamed for being the sources of theseenvironmental problems. This study aimed to examine the factors influencing the perceived environmental performance of firms certified under the ISO 14001 in Malaysia. A total of 268 manufacturing companies responded to the questionnaires. In this study, structural equation modeling was applied to test the hypotheses. This study identifed X-ray laboratory users’ compliance with the innovated radiographic technique and evaluated it’s effectiveness in decreasing risks of repeating unnecessary X-ray exposure. The use of various steps in the standard operating procedures of radiography, proper radiographic technique and critique, practical protection measures, and radiographic positioning practice in X-ray laboratory at a private radiographic academy in Indonesia were audited. Thirty participants were used as the simple random sample. The demographic distribution was divided into, gender, nationality, and practical experience. In this study, the photochemical degradation via photo-Fenton process was carried out to degrade dyes in textile industrial wastewaters. Experimental design methodology was also applied for optimizing effects of factors which influencethe effective treatment such as ferric dose, hydroperoxide dosage, initial pH, reaction time and initial chemical oxygen demand (COD). Two independent variables namely colour and COD removal efficiencieswere used to evaluate the treatment yield. Under the optimal conditions, ca. 99% and ca. 88%, colour, and COD were removed, respectively.

Background: Biomass pretreatment using certain ionic liquids (ILs) is very efficient, generally producing a substrate that is amenable to saccharification with fermentable sugar yields approaching theoretical limits. Although promising, several challenges must be addressed before an IL pretreatment technology can become commercially viable. One of the most significant challenges is the affordable and scalable recovery and recycle of the IL itself. Pervaporation (PV) is a highly selective and scalable membrane separation process for quantitatively recovering volatile solutes or solvents directly from non-volatile solvents that could prove more versatile for IL dehydration.

Biofilms are communities of sessile microbes that are phenotypically distinct from their genetically identical, free-swimming counterparts. Biofilms initiate when bacteria attach to a solid surface. Attachment triggers intracellular signaling to change gene expression from the planktonic to the biofilm phenotype. For Pseudomonas aeruginosa, it has long been known that intracellular levels of the signal cyclic-di-GMP increase upon surface adhesion and that this is required to begin biofilm development. Extracellular polysaccharides are compounds secreted by microorganisms into the surrounding environment, and they are important for surface attachment and maintaining structural integrity within biofilms. The social nature of many extracellular polysaccharides remains unclear, and it has been suggested that they could function as either cooperative public goods or as traits that provide a competitive advantage. Here, we empirically tested the cooperative nature of the PSL polysaccharide, which is crucial for the formation of biofilms in Pseudomonas aeruginosa. The Marine Gastrobot sponsored by Dr. Christopher Kitts of the Cal Poly Center for Applications in Biotechnology was a research and development effort intended to explore the use of microbial fuel cell technology as a power source for underwater robots. Our team Ocean Locomotion succeeded in developing a first iteration of an underwater robotic platform suitable for microbial fuel cell integration. Cyanobacteria are photosynthetic organisms that efficiently capture light by utilizing the light-harvesting complexes called phycobilisomes. In many cyanobacteria, phycobilisomes are composed of an allophycocyanin core with phycocyanin and phycoerythrin (PE) rods radiating from the core. These phycobiliproteins have multiple bilin chromophores, such as phycoerythrobilin (PEB), covalently attached to specific cysteine (Cys) residues for efficient photosynthetic light capture. Chromophore ligation on phycobiliprotein subunits occurs through bilin lyase catalyzed reactions.

Cancer-based immunotherapy has led the evolution of biologics that can stimulate immune responses towards tumor eradication. The synthesis of small to intermediate size molecules with the targeting and effector functions of mAb may represent a novel class of immunotherapeutics that may overcome the limitations of their biological counterparts.Towards this objective, B7H6 has been identified as a protein ligand localized on the cell surface of transformed tumor cells. Eurygaster integriceps Puton, common name sunn Pest, is one of the primary sources of wheat crop wastes in North Africa, Middle East, and Eastern Europe. It feeds by injecting the wheat grain with an enzyme characterized as prolyl endoprotease (spPEP) that breaks down Gluten, the wheat’s main constitutive protein necessary for bread production (Darkoh et al., 2010). One of the primary drivers of cyanobacterial harmful algal blooms (cHABs) in freshwater systems is nutrient loading, particularly of nitrogen and phosphorus. There has been an increased focus on assessing the role of nitrogen (N) in freshwater lakes and rivers that suffer cHABs. Urea, a widely-used, N-rich fertilizer, is a source of interest due to its abundance in freshwater ecosystems, primarily caused by anthropogenic nutrient loading. A Gram negative species of bacteria, Campylobacter jejuni, is the leading cause of food poisoning worldwide. Humans often contract food poisoning after ingesting contaminated poultry. Detecting the presence of C. jejuni in poultry is difficult because it is part of the natural flora and does not cause symptomatic infection. In a related manner, Bordetella avium is a Gram negative species of bacteria that causes bordetellosis in poultry. Keratoconjunctivitis sicca (KCS), also known as dry-eye disease, causes a deficiency of tears in both humans and canines. Due to the lack of effective therapeutics for the treatment of dry-eye disease, there is a market potential for a novel secretion enhancing factor. Lacritin, a naturally occurring tear glycoprotein, increases basal tearing in rabbits when topically applied to the ocular surface and shows potential as a dry-eye therapeutic. Dionaea muscipula, Venus fly trap, is an endangered plant that is propagated primarily through tissue culture. Tissue cultured plants must be acclimatized to their new environments when removed from their test tubes and the ensuing stress can result in plant mortality. A plant growth promoting rhizobacterium, Burkholderia phytofirmans strain PsJN, has been utilized in biotic hardening of tissue cultured plants and in the amelioration of stresses, particularly drought stress in potted plants. Clostridium thermocellum is a Gram-positive anaerobe with the ability to hydrolyze and metabolize cellulose into biofuels such as ethanol, making it an attractive candidate for consolidated bioprocessing (CBP). At present, metabolic engineering in C. thermocellum is hindered due to the incomplete description of its metabolic repertoire and regulation within a predictive metabolic model. Genome-scale metabolic (GSM) models augmented with kinetic models of metabolism have been shown to be effective at recapitulating perturbed metabolic phenotypes.

One of the most effective methods of reducing pesticide application in vineyards is breeding hybrid varieties for disease resistance. The new varieties must be assessed for disease resistance and viticultural traits. This thesis focuses on seven new varieties from a cross of ‘Norton’ and ‘Cabernet Sauvignon’. ‘Norton’ was chosen because of its disease resistance and cold hardiness. ‘Cabernet Sauvignon’ was selected because of its high-quality wine making berries. Ciliopathies are a class of genetic diseases which affect the function of primary cilium within multiple organs throughout the body. Although ciliopathies are known to present high morbidity and mortality rates, there are no current cures and only a limited amount of treatments. A universal function of cilia is to regulate cellular activities through the transduction of signals. Disruption of the localization of proteins following the reception of a stimuli can be responsible for the phenotypic effects of dysfunctioning cilia in ciliopathies. Sand flies, like mosquitoes, ticks, fleas, and lice, transmit pathogens that cause disease in humans. Leishmaniasis, caused by pathogens transmitted by sand flies, kills tens of thousands of people every year. Insecticides have been used to control sand flies, but there is evidence of insecticide resistance in populations of sand flies around the world. The goal of this dissertation was to develop tools to maintain sand flies in the laboratory, develop the ability to identify insecticide-resistant populations of sand flies, and to investigate the genetic mechanisms of how sand flies become resistant to insecticides. Autoimmune diseases are characterized by highly specific immune responses against molecules in self-tissues. Different autoimmune diseases are characterized by distinct immune responses, making autoantibodies useful for diagnosis and prediction. In many diseases, the targets of autoantibodies are incompletely defined. Although the technologies for autoantibody discovery have advanced dramatically over the past decade, each of these techniques generates hundreds of possibilities, which are onerous and expensive to validate. This dissertation is built upon the fundamental idea that the pre-B cell receptor (pre-BCR) is important to leukemia cell survival and a logical therapeutic target in B cell precursor acute lymphoblastic leukemia (BCP-ALL). The pre-BCR is expressed early at a specific stage during B cell development where it plays a central role in survival of healthy B lymphocytes. This receptor is composed of the membrane heavy chain (mIgμ) associated with surrogate light chain components, 5 and VpreB. Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) is an exciting new genetic engineering technology that was found in the chromosomes of certain bacteria and archaea. Bacteriophage are viruses that insert their DNA and hijack the host cell’s machinery to make new phage that can go on to infect new cells. Some microorganisms use CRISPR as a defense mechanism to disrupt the bacteriophage DNA after it is inserted into the cell.
