Bawang putih merupakan komoditas penting di Indonesia. Indonesia merupakan negara terbesar pengimpor bawang putih. Permasalahan yang dihadapi dalam teknik produksi benih secara konvensional menjadi salah satu faktor pembatas tidak optimalnya pengembangan dan penyebaran benih tersebut. Produksi benih in vitro dapat dijadikan sebagai metode alternatif dalam perbanyakan benih sumber kultivar unggul. In all multicellular organisms, the fundamental processes of cell proliferation and cell death are crucial for growth regulation during organogenesis. Strict regulation of cell death is important to maintain tissue homeostasis by affecting processes like regulation of cell number, and elimination of unwanted/unfit cells. The developing Drosophila eye is a versatile model to study patterning and growth, where complex signaling pathways regulate growth and cell survival. However, the molecular mechanisms underlying regulation of these processes is not fully understood. Myo-inositol phosphates (phytates) are important biological molecules produced largely by plants to store phosphorus. Phytate is very abundant in many different soils making up a large portion of all soil phosphorus. This review assesses current phytase science from the perspective of its substrate, phytate, by examining the intricate relationship between the phytate-hydrolyzing enzymes and phytate as their substrate. Specifically, we examine available data on phytate’s structural features, distribution in nature and functional roles. The vast majority of clinical human listeriosis cases are caused by serotype 1/2a, 1/2b, 1/2c, and 4b isolates of Listeria monocytogenes. The ability of L. monocytogenes to establish a systemic listeriosis infection within a host organism relies on a combination of genes that are involved in cell recognition, internalization, evasion of host defenses, and in vitro survival and growth. This clinical audit identified the compliance rate of ultrasound users to the standard operating procedures (SOP); and identified the factors that led to a compliance rate of ultrasound human users. The review was conducted for six days in a private radiologic academy in Indonesia. Only 23 subjects selected randomly were audited. Of the 23 participants, there were 11 males and 12 females. Of the 11 males, seven were compliant, and three were non-compliant. Of the 12 females, eight were compliant, and four were non-compliant. Wound Healing Post-Mesh Repair ― An Observational Study, Lebbe M.N, Kularajasingham J, Dioso P

This study identified the best surgical mesh repair techniques for inguinal hernia and prevalence of wound healing post-mesh repair. The cross-sectional study design used cluster sampling for data collection. Of the 120 respondents, 48.3% preferred anterior tension-free mesh repair and 49.2% Lichtenstein mesh repair, both identified as the common surgical techniques in eastern Sri Lanka. About 82.5% of the respondents (n = 99) healed while 17% (n = 21) had recurrence of hernia after one month. Nevertheless, 2.5% of the total respondents said that the hernia repaired after one month but less than two months; and 97.5% of the interviewees …

This research identified evidence of knowledge and caring attitude among Sri Lankan nurses caring for dying patients; and identified the factors affecting nursing care for the dying in the eastern province of Sri Lanka. A quantitative cohort study design was used having questionnaires as the means of collecting the data. The purposive way of sampling selected 120 staff nurses employed in a government hospital as respondents who falls under the inclusion and exclusion criteria.There was significant evidence of caring attitude towards the dying (p = 0.005534) and knowledge on the end-of-life nursing care (p = 0.0004703) for the dying patients. …

The main goal of this study is to elaborate and evaluate the physicochemical properties of the synthetic biphasic calcium phosphate (BCP) powder: an associate compound of hydroxyapatite (HA): Ca10(PO4)6(OH)2 and beta-tricalcium phosphate (β-TCP): Ca3(PO4)2. The new compound BCP has two advantages: high bioactivity (HA) and fast biodegradation (β-TCP). The obtained powder of BCP was prepared by the precipitate method. XRD analysis confirmed the synthetic material contained both HA and β-TCP crystalline phases. Spermatogenesis is the process by which mature functional spermatozoa are formed, and is initiated in the stem cell niche of the testes. During the post-meiotic stage of spermatogenesis, spermiogenesis, transition proteins facilitate the transformation of chromatin from a histone-based nucleosome structure to a protamine-based nucleosome structure. This study is aimed at analyzing genomic, transcript, and protein sequences of transition proteins in 13 sequenced Drosophila species. The Drosophila melanogaster spermatid specific transition protein-like protein (Tpl94D) was used as the reference sequence in this study. Unusual monoenoic fatty acids (UMFA’s) and specialized metabolites called anacardic acids (AnAc) are produced in glandular trichomes of Pelargonium ´ hortorum (geranium). The UMFA’s, 16:1∆11 and 18:1∆13 are precursors for the synthesis of unsaturated AnAc 22:1n5and 24:1n5 that contribute to pest resistance in geraniums. UMFAs and their derived AnAc metabolites not only provide a useful biological marker that differentiates the biosynthetic pathway for unusual mononenes from the common fatty acids (i.e. stearic, palmitic, oleic, linoleic and linolenic) but also have industrial, medical and agricultural applications. In an effort to expand the pool of bacterium useful for biotechnology applications, Pseudomonas fluorescens, a common gram negative microbe, was examined for its ability to function in a recombinant setting. P. fluorescens is ubiquitous in nature and was initially identified as a soil bacterium found in dirt and is typically associated with plant material. Copper plays key catalytic and regulatory roles in biochemical processes essential for normal growth, development, and health. Defects in copper metabolism cause Menkes and Wilson’s disease, myeloneuropathy, and cardiovascular disease and are associated with other pathophysiological states. The ribosome is the quintessential antibacterial drug target, with many structurally and mechanistically distinct classes of antibacterial agents acting by inhibiting ribosome function. Detecting and quantifying ribosome inhibition by small molecules and investigating their binding modes and mechanisms of action are critical to antibacterial drug discovery and development efforts. To develop a ribosome inhibition assay that is operationally simple, yet provides direct information on the drug target and the mechanism of action, we have developed engineered E. coli strains harboring an orthogonal ribosome controlled green fluorescent protein reporter that produce fluorescent signal when the O-ribosome is inhibited. Target-based screening is one of the major approaches in drug discovery. Besides the intended target, unexpected drug off-target interactions often occur, and many of them have not been recognized and characterized. The off-target interactions can be responsible for either therapeutic or side effects. Thus, identifying the genome-wide off-targets of lead compounds or existing drugs will be critical for designing effective and safe drugs, and providing new opportunities for drug repurposing. In the fight against drug-resistant bacteria, accurate and high-throughput detection is essential. Here, a bimaterial microcantilever with an embedded microfluidic channel with internal surfaces chemically or physically functionalized with receptors selectively captures the bacteria passing through the channel. Bacterial adsorption inside the cantilever results in changes in the resonance frequency (mass) and cantilever deflection (adsorption stress). The excitation of trapped bacteria using infrared radiation (IR) causes the cantilever to deflect in proportion to the infrared absorption of the bacteria, providing a nanomechanical infrared spectrum for selective identification. Background: SENSITIVE TO FREEZING 2 (SFR2) is classified as a glycosyl hydrolase, and by using glycosyltransferase activity, it modifies membrane lipids to promote freeze tolerance.

Results: Although the active site of SFR2 is identical to hydrolases, adjacent loop regions contribute to its transferase activity.

Conclusion: Transferase activity evolved by modifications external to the core catalytic site.

Significance: Defined structure-function relationships will inform engineering of transferases and freeze tolerance.

Normal cellular physiology is critically dependent on numerous mitochondrial activities including energy conversion, cofactor and precursor metabolite synthesis, and regulation of ion and redox homeostasis. Advances in mitochondrial research during the last two decades provide solid evidence that these organelles are deeply integrated with the rest of the cell and multiple mechanisms are in place to monitor and communicate functional states of mitochondria. Large manufacturers produce blends of probiotics for supplementing the diets of agriculturally important animals. Perbanyakan tanaman pisang secara konvensional dengan menggunakan bonggol atau anakan (sucker) secara terus menerus dapat menurunkan kualitas dan memerlukan waktu lama serta meningkatkan terjadinya serangan penyakit. Oleh karena itu, suatu upaya untuk menghasilkan bibit yang memiliki kualitas baik dalam waktu yang cepat dapat menggunakan kultur in vitro. Percobaan ini bertujuan untuk memperoleh laju multiplikasi yang tinggi dengan penggunaan salah satu jenis dan konsentrasi sitokinin pada kultur in vitro tanaman pisang kepok kuning. In transcription, a universal step in gene expression, information from a DNA sequence is copied into RNA. A key component in gene expression is the promoter sequence, a region of DNA to which RNA polymerase binds during the initiation of transcription of downstream genes. Most bacterial promoters contain a -10 and a -35 sequence that are bound by the RNA polymerase. Some promoters also contain an Upstream Promoter (UP) element. Production and isolation of proteins are difficult, costly and time-consuming processes. The aim of this project is for the development of plasmids, which allow for streamlined production and isolation of proteins. To allow for modular insertion of varying segments of DNA we are using ‘recursive directional ligation by plasmid reconstruction’. This technique uses type II restriction endonucleases, which cut downstream from their recognition site allowing multiple insertions without losing a restriction site. Found in all living organisms, terpenoids make up the largest group of natural products and are essential compounds for many major processes, including photosynthesis, respiration, hormone production, and electron transport. Additionally, they have commercial and medical value in products including fragrances, cosmetics, and medicines. Terpenoids originate from the five-carbon building blocks isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP), which are synthesized by the mevalonic acid (MVA) and methylerithritol phosphate (MEP) pathways. An alternative MVA pathway was discovered in Archaea with the final two enzymes being phosphomevalonate decarboxylase (MPD) and isopentenyl phosphate kinase (IPK). The CRISPR-Cas system is an adaptive immune system found in bacteria which helps protect against the invasion of other microorganisms. This system induces double stranded breaks at precise genomic loci (1) in which repairs are initiated and insertions of a target are completed in the process. This mechanism can be used in eukaryotic cells in combination with sgRNAs (1) as a tool for genome editing.
