Kunyit putih (Kaempferia rotunda L.) merupakan tanaman obat yang bermanfaat dan perlu dibudidayakan lebih luas, namun terkendala oleh penyediaan bibit bermutu yang terbatas sehingga menjadi langka. Perbanyakan tanaman dengan cara konvensional memerlukan bibit bermutu dalam jumlah dan waktu yang lama, sedangkan ketersediaan benih masih terbatas. Perbanyakan secara in vitro dapat mengatasi kelangkaan bibit kunyit putih yang bermutu tinggi dalam jumlah besar dan bebas patogen. Ketersediaan bibit unggul yang dihasilkan secara in vitro bergantung pada jenis dan konsentrasi sitokinin yang ditambahkan pada media kultur. Perbanyakan tanaman stroberi secara konvensional dilakukan dengan menggunakan stolon, tetapi kurang efektif serta kualitas bibit yang dihasilkan kurang baik akibat adanya akumulasi penyakit. Budidaya stroberi memerlukan adanya perbanyakan bibit secara massal, tetapi tidak mengubah kualitasnya. Multiplikasi in vitro menjadi solusi untuk penyediaan bibit berkualitas dalam jumlah besar. Kondisi cekaman kekeringan pada tanaman tomat dapat menyebabkan pertumbuhan dan produksi tanaman tomat menurun. Salah satu upaya yang dapat dilakukan untuk mengatasi masalah tersebut adalah dengan perakitan varietas tanaman baru yang tahan terhadap cekaman kekeringan. Beberapa hasil mutasi gen IAA pada tomat mutan Micro-Tom mampu menghasilkan tanaman yang toleran terhadap kondisi stress secara abiotik, yaitu pada galur iaa9-3 dan iaa9-5. Locoweeds are the most widespread poisonous plant problem in the world and have been reported in the Western United States since the 1800s, causing tremendous losses in livestock. Consumption of locoweeds by grazing animals stimulates the neurological disease, locoism, characterized by weight loss, ataxia, and lack of muscular coordination. The name locoweed is used for Astragalus and Oxytropis species known to contain swainsonine, the toxic principle produced by the plant endophytic fungus Undifilum. An efficient protocol was developed for rapid propagation and regeneration of the Dioscorea alata L. locally known as ‘ubi badak’ because of their rhinoceros-shaped tubers. In the present study, direct and indirect regeneration were induced to produce plantlets of Dioscorea alata L. For direct regeneration explants (leave, stem and node) were cultured on MS media supplemented with various combinations and concentrations of BAP and NAA to induce microshoots and roots formation. The optimum medium for microshoots formation was MS medium supplemented with 1.0 mg/l BAP and 0.5 mg/l NAA with the number of microshoot is 23.07±0.44 from node explant. While …

Cytokinin, long known as a phytohormone that regulates plant growth and development, has been recently recognized as an important regulator of stress responses. However, our current knowledge about the mechanisms by which cytokinin regulates stress responses is fragmentary, as many of the studies in this field yielded conflicting results. Most of the work described here has focused on analyses of the molecular mechanisms of cytokinin-dependent regulation of growth and development under stress conditions, with an emphasis on the role of cytokinin-dependent regulation of protein synthesis in development and stress tolerance.

The in vitro propagation is a promising method for the production of plants in species that are endangered, commercially valuable, and/or otherwise difficult to propagate through traditional horticultural methods. Short Tandem Target Mimic (STTM) is a commonly technique used for functional studies of a number of genes in several plant model systems. However, very little is known about application of STTM technique in tree species. In this study, STTM was applied to knock down microRNA396 (miR396) in transgenic poplar trees for the first time. STTM396 expression resulted in dramatic decrease in miR396 expression levels leading to taller plants with larger leaves and larger leaf cell size. To minimize water loss, among many adaptation strategies, plants close their stomata to reduce transpiration. The stomatal closure is regulated by light, internal CO2 concentration, and plant hormones, mainly abscisic acid. Maturase K (MatK) is the only group II intron encoded protein in the chloroplast of land plants. Maturases are prokaryotic enzymes that catalyze formation of the lariat structure needed for intron removal from precursor RNAs. The chloroplast maturase MatK, is a descendant of prokaryotic maturases, however, unlike its prokaryotic relatives, MatK is thought to catalyze excision of, not only its own intron, but also the introns of other group II introns in the plastome. The environmental fate and potential impacts of nanopesticides on agroecosystems under realistic agricultural conditions are poorly understood. As a result, the benefits and risks of these novel formulations compared to the conventional products are currently unclear. Here, we examined the effects of repeated realistic exposures of the Cu(OH)2 nanopesticide, Kocide 3000, on simulated agricultural pastureland in an outdoor mesocosm experiment over 1 year. Photosynthetic membranes provide much of the usable energy for life on earth. To produce photosynthetic membrane lipids, multiple transport steps are required, including fatty acid export from the chloroplast stroma to the endoplasmic reticulum, and lipid transport from the endoplasmic reticulum to the chloroplast envelope membranes. Transport of hydrophobic molecules through aqueous space is energetically unfavorable and must be catalyzed by dedicated enzymes, frequently on specialized membrane structures. Catharanthus roseus (Madagascar periwinkle) is the exclusive source of an array of terpenoid indole alkaloids (TIAs) that are used in the treatments of hypertension and certain types of cancer. TIA biosynthesis is under stringent spatiotemporal control and is induced by jasmonate (JA) and fungal elicitors. Tryptamine, derived from the indole branch, and secologanin from the iridoid branch are condensed to form the first TIA, strictosidine. Unusual monoenoic fatty acids (UMFA’s) and specialized metabolites called anacardic acids (AnAc) are produced in glandular trichomes of Pelargonium ´ hortorum (geranium). The UMFA’s, 16:1∆11 and 18:1∆13 are precursors for the synthesis of unsaturated AnAc 22:1n5and 24:1n5 that contribute to pest resistance in geraniums. UMFAs and their derived AnAc metabolites not only provide a useful biological marker that differentiates the biosynthetic pathway for unusual mononenes from the common fatty acids (i.e. stearic, palmitic, oleic, linoleic and linolenic) but also have industrial, medical and agricultural applications. Perbanyakan tanaman pisang secara konvensional dengan menggunakan bonggol atau anakan (sucker) secara terus menerus dapat menurunkan kualitas dan memerlukan waktu lama serta meningkatkan terjadinya serangan penyakit. Oleh karena itu, suatu upaya untuk menghasilkan bibit yang memiliki kualitas baik dalam waktu yang cepat dapat menggunakan kultur in vitro. Biofuels are a form of renewable energy derived from living matter, typically plants. The push for biofuels began in order to decrease the amount of carbon dioxide (CO2) released into the atmosphere, as biofuels are essentially carbon neutral. The idea is the same amount of CO2 the plants took in to perform photosynthesis will then be released in the burning of the biofuels. Algae is an excellent source of biofuels because it grows quickly and is versatile in terms of the type of fuel it can produce. Climatic adaptation is an example of a genotype-by-environment interaction (G×E) of fitness. Selection upon gene expression regulatory variation can contribute to adaptive phenotypic diversity; however, surprisingly few studies have examined how genome-wide patterns of gene expression G×E are manifested in response to environmental stress and other selective agents that cause climatic adaptation. Here, we characterize drought-responsive expression divergence between upland (drought-adapted) and lowland (mesic) ecotypes of the perennial C4 grass, Panicum hallii, in natural field conditions. Overall, we find that cis-regulatory elements contributed to gene expression divergence across 47% of genes, 7.2% of which exhibit drought-responsive G×E. …

Isoprenoids are one of the most diverse classes of natural products and are all derived from universal five carbon, prenyl precursors. Squalene and botryococcene are linear, hydrocarbon triterpenes (thirty carbon compounds with six prenyl units) that have industrial and medicinal values. Squalene is produced by all eukaryotes as it is the first committed precursor to sterols, while botryococcene is uniquely produced by the green algae, Botryococcus braunii (race B). Natural sources for these compounds exist, but there is a desire for more renewable production platforms. The primary cell wall is a highly organized multi-layered matrix of polysaccharides (cellulose, hemi-cellulose, and pectin). The ability of the rigid cell wall to sufficiently loosen to allow growth is a complex process that differs considerably between grasses monocots and dicots. Cellulose is the major structural component required for anisotropic cell expansion and is synthesized by CELLULOSE SYNTHASE A (CesA) proteins. Plants respond to environmental stress by altering their gene expression. Under stress conditions some genes are activated and some genes are repressed. Even though a lot of work has been done to understand mechanisms of gene activation under abiotic stress very little information is available on how stress responsive genes are kept repressed under normal growth conditions. Recent work has revealed that plants use transcriptional repression as common mechanism of gene repression. Transcriptional repression is achieved by recruitment co-repressor complexes to the target genes. Microalgae can be grown on municipal wastewater media to both treat the wastewater and produce feedstock for algae biofuel production. However the reliability of treatment must be demonstrated, as well as high areal algae productivity on recycled wastewater media and efficient sedimentation harvesting. Tujuan dari penelitian ini adalah untuk mendapatan metode perbanyakan tanaman nilam melalui teknik in vitro dengan meng-gunakan jenis eksplan serta jenis dan konsentrasi sitokinin yang berbeda. Percobaan dilaksanakan di Laboratorium Teknologi Benih Fakultas Pertanian Universitas Padjadjaran dari bulan Maret sampai dengan Juni 2012. Rancangan percobaan menggunakan Rancangan Acak Lengkap pola faktorial yang terdiri dua faktor dan tiga kali ulangan. Faktor pertama adalah tipe eksplan (mata tunas, pucuk, dan daun).
