Optimisation Of Biomass And Lipid Production Of A Tropical Thraustochytrid Aurantiochytrium Sp. Umacc-T023 In Submerged-Liquid Fermentation For Large-Scale Biodiesel Production, Sohedein Mohamad Nor Azzimi

A tropical thraustochytrid, Aurantiochytrium sp. UMACC-T023, was optimised for high biomass and lipid production in submerged-liquid fermentation (SLF). Biomass and lipid production were optimised based on glucose, polypeptone, and yeast extract concentration using response surface methodology (RSM). In RSM, the applied central composite design (CCD) showed that the optimisation model was significant for all variables studied. Yeast extract and polypeptone were associated with significant effects in UMACC-T023 biomass production with p < 0.05. The model was validated by employing the optimised media composition in shake flasks and in 1.4-L stirred-tank bioreactors. The optimised media composition for both biomass and lipid production was 10 g L-1 glucose, 15.08 g L-1 polypeptone, and 13.56 g L-1 yeast extract. Biomass production in the bioreactor increased 2.12-fold compared with the shake flask culture utilising the same optimised media composition. This result demonstrated that the optimized media composition can be utilised for large-scale lipid production of UMACC-T023 for potential biodiesel feedstock as it also fulfilled the international standards for biodiesel.

This thesis demonstrates efficacy of Octenyl Succinic Anhydride (OSA), as a biofilm sanitizer. Biofilms allow bacteria to adhere to solid surfaces with the use of excreted polymeric compounds. For example, surfaces found in food production or processing facilities such as the interior of a raw milk holding tank, are some of the most susceptible to biofilm contamination. When present, biofilms can cause a variety of negative effects, which include; reduction of product shelf life, corrosion, and outbreaks of foodborne illnesses. Quantitative understanding of biomolecular electrostatics, particularly involving multivalent ions and highly charged surfaces, remains lacking. Ion-modulated interactions between nucleic acids provide a model system in which electrostatics plays a dominant role. Using ordered DNA arrays neutralized by spherical cobalt3+ hexammine and Mg2+ ions, we investigate how the interstitial ions modulate DNA-DNA interactions. Using methods of ion counting, osmotic stress, and x-ray diffraction, we systematically determine thermodynamic quantities, including ion chemical potentials, ion partition, DNA osmotic pressure and force, and DNA-DNA spacing. Analyses of the multidimensional data provide quantitative insights into their interdependencies. The key finding of this study is that …

Somatic stem cells have been used as a regenerative medicine tool to treat diseases like leukemia for decades, but they are limited in their ability to self-renew and differentiate. A better alternative, induced pluripotent stem cells (iPSCs), has the ability to generate cells from all three germ layers and avoid tissue rejection as well as bypass the ethical concerns related to embryonic stem cells. In 2006, Takahashi and Yamanaka discovered that only four genes were required to induce pluripotency: Oct4, Sox2, Klf4, and c-Myc (OSKM). Magnetosomes are intracellular magnetic nanocrystals composed of magnetite (Fe3O4) or greigite (Fe3S4), enveloped by a lipid bilayer membrane, produced by magnetotactic bacteria. Because of the stability of these structures in certain environments after cell death and lysis, magnetosome magnetite crystals contribute to the magnetization of sediments as well as providing a fossil record of ancient microbial ecosystems. The persistence or changes of the chemical and magnetic features of magnetosomes under certain conditions in different environments are important factors in biotechnology and paleomagnetism. Polymethacrylate-based copolymers, such as EudragitTM, are sensitive to alterations in pH levels, becoming increasingly more soluble in response to an increase in pH. Post-translational modifications (PTMs) play a significant role in regulating cell signaling and regulating the inner cellular environment. The study of PTMs and their primary biological functions is critical for the development of new medicines targeting the pathways and biomolecules responsible for the PTMs.

This paper intends to explore the rationality and feasibility of modeling dispersed submicron particles in air by a kinetic-based method called the unified gas-kinetic scheme (UGKS) and apply it to the simulation of particle concentration under a transverse standing wave. A gas-particle coupling scheme is proposed where the gas phase is modeled by the two-dimensional linearized Euler equations (LEE) and, through the analogous behavior between the rarefied gas molecules and the air-suspended particles, a modified UGKS is adopted to estimate the particle dynamics. Distinct populations of steelhead in the wild are in decline. The propagation of steelhead in hatcheries has been used to boost population numbers for recreational fisheries and for use in conservation. However, hatchery breeding practices of steelhead can result in changes in genetic structure. I investigated the genetic structure of winter-run steelhead (Oncorhynchus mykiss) returning to the Mad River, California, where a hatchery has been used enhance production for recreational fisheries since 1971. Brucellosis is considered a significant health threat, it is an infectious disease caused by the bacteria Brucella, which can spread from animals to humans causing severe diseases. Through this project, we aim to develop a very highly sensitive biosensor to detect Brucella early before spreading. This sensor is based on surface plasmon resonance (SPR) technique, which is used to analyze kinetics of interaction between biomolecules. It can detect down to picomolar concentrations of some proteins.

Herein we report an in-depth study on the cytotoxic mechanism of action of four developmental cytotoxic copper(II) complexes: [Cu(phen)2]2+ (Cu-Phen); [Cu(DPQ)(Phen)]2+ (Cu-DPQ-Phen); [Cu(DPPZ)(Phen)]2+; and [Cu(DPPN)(Phen)]2+ (where Phen = 1,10-phenanthroline, DPQ = dipyrido[3,2-f:20,30-h]quinoxaline, DPPZ = dipyrido[3,2-a:20,30-c]phenazine, and DPPN = benzo[i]dipyrido[3,2-a:20,30-c]phenazine). This complex class is known for its DNA intercalative properties and recent evidence—derived from an in vivo proteomic study—supports the potential targeting of mitochondrial function. Therefore, we focused on mitochondrial-mediated apoptosis related to cytotoxic activity and the potential impact these agents have on mitochondrial function. งานวิจัยนี้มุ่งเน้นศึกษาปัจจัยที่มีผลต่อการเกิดตำหนิและหาแนวทางปรับปรุงกระบวนการผลิตเพื่อลดปริมาณตำหนิชนิดฟองอากาศและรีม ด้วยการนำตัวแปรจากกระบวนการผลิตได้แก่ การชั่งน้ำหนักส่วนผสม อุณหภูมิในการหลอม อุณหภูมิในการขึ้นรูปและองค์ประกอบทางเคมีของกระจก มาหาความสัมพันธ์กับปริมาณตำหนิที่เกิดขึ้นด้วยการวิเคราะห์การถดถอย จากผลการวิเคราะห์ตำหนิฟองอากาศด้วยเทคนิคแมสสเปกโทรเมตรีและเทคนิครามานสเปคโทรสโคปีพบว่าฟองอากาศส่วนใหญ่พบแก๊ส CO2 เป็นองค์ประกอบอยู่ที่ 30 ถึง 70 เปอร์เซ็นต์ ซึ่งเกิดจากการสลายตัวทางความร้อนของวัตถุดิบและการไล่ฟองอากาศที่ไม่สมบูรณ์ และจากความสัมพันธ์ของปริมาณฟองอากาศและตัวแปรจากกระบวนการผลิตพบว่า อุณหภูมิการหลอมที่สูงขึ้น ปริมาณ fining agent ที่น้อยลง และกำลังการผลิตที่เพิ่มขึ้นส่งผลให้ตำหนิชนิดฟองอากาศเพิ่มขึ้น จากการศึกษาตำหนิชนิดรีมโดยใช้กล้องจุลทรรศน์อิเล็กตรอนแบบส่องกราดร่วมกับการวัดการกระจายพลังงานของรังสีเอกซ์ในการวิเคราะห์องค์ประกอบธาตุของตำหนิชนิดรีมเทียบกับบริเวณโดยรอบโดยพบว่ารีมมีปริมาณธาตุ Si และ Na สูงกว่าบริเวณโดยรอบ ซึ่งเกิดจากการชั่งวัตถุดิบที่คลาดเคลื่อนไปจากสูตรการผลิต โดยเกิดจากวัตถุดิบติดค้างบริเวณกรวยรับวัตถุดิบ ส่งผลให้องค์ประกอบทางเคมีของส่วนผสมไม่สม่ำเสมอ ยิ่งความแตกต่างของน้ำหนักส่วนผสมจากสูตรการผลิตยิ่งมาก จะทำให้ปริมาณรีมเพิ่มสูงขึ้น ซึ่งสามารถความผิดพลาดจากการชั่งนี้สามารถแก้ไขได้โดยติดตั้งค้อนลมเพื่อเคาะวัตถุดิบให้ลงไปในโม่ผสมวัตถุดิบครบถ้วนตามสูตรการผลิต

โดยทั่วไปกระบวนการหลอมแก้วระดับอุตสาหกรรมต้องการพลังงานสูงสำหรับการเปลี่ยนแปลงจากวัตถุดิบเป็นน้ำแก้ว วัตถุประสงค์ในงานวิจัยนี้จะกล่าวถึงการพัฒนากระบวนการหลอมแก้วโซดาไลม์โดยวิธีปรับลำดับการผสมในระดับห้องปฏิบัติการ ซึ่งอาศัยกลไกของปฏิกิริยาสถานะของแข็งระหว่างทรายแก้วและโซเดียมคาร์บอเนตในช่วงแรกของการหลอม การเตรียมส่วนผสมแบบดั้งเดิมเป็นการผสมวัตถุดิบทั้งหมดภายในครั้งเดียว ในงานวิจัยนี้ต้องการปรับเปลี่ยนวิธีการเตรียมส่วนผสมให้เหมาะสมที่สุดต่อการหลอมโดยการปรับลำดับของการผสม ประกอบด้วยสองขั้นตอนการผสม ขั้นตอนแรกคือการผสมทรายแก้วและโซเดียมคาร์บอเนต จากนั้นจึงผสมวัตถุดิบที่เหลือเป็นขั้นตอนที่สอง เพื่อเปรียบเทียบและศึกษาผลของการเตรียมส่วนผสมทั้งสองวิธี ได้ทำการศึกษาสภาพและการเปลี่ยนแปลงระหว่างกระบวนการหลอมที่อุณหภูมิตั้งแต่ 600 ถึง 1000 องศาเซลเซียส นอกจากนี้มีผลของปริมาณวัตถุดิบที่หลอมไม่หมดบนพื้นผิวซึ่งยังคงลงเหลือในสภาพผลึก ณ สภาวะที่กำหนด เพื่อเปรียบเทียบแนวโน้มประสิทธิผลในการหลอมของส่วนผสมจากการผสมสองวิธี โดยภาพถ่ายผิวหน้าและโดยเทคนิคการเลี้ยวเบนรังสีเอกซ์ ผลการวิจัยพบว่าการจัดลำดับการผสมเป็นวิธีการผสมที่เหมาะสมกว่า ส่งผลต่ออัตราการหลอม พื้นผิวสัมผัสที่เพิ่มขึ้นระหว่างทรายแก้วและโซเดียมคาร์บอเนตจากการผสมขั้นตอนแรกนำไปสู่การเพิ่มอัตราการหลอมของทั้งระบบ ต่างจากวิธีการเดิมที่อาจมีวัตถุดิบอื่นเป็นตัวขัดขวางการเกิดปฏิกิริยาสถานะของแข็งดังกล่าว

In this study, Anacyclus pyrethrm Link (roots) is a medicinal plant from the Asteraceae family, endemic to Morocco and is widely used in traditional medicine; was investigated for their total phenolics, flavonoids contents and antioxidant activities. The quantitative estimation of total flavonoids and phenols by the colorimetric method showed that the extracts were prepared by fractionation of the crude hydromethanolic extract (ethyl acetate and n-butanol). Ciguatera fish poisoning (CFP) is a type of illness that causes human intoxication after consuming contaminated coral fishes. The neurotoxins, ciguatoxins, are produced by species of Gambierdiscus, one of the harmful benthic dinoflagellates. Identification of harmful dinoflagellate species is important to gain further insights into the global dispersion. In this study, morphological characteristics of species in the genus Gambierdiscus and their habitat preferences were investigated. Field sampling was undertaken at Pulau Rawa, Terengganu and Pulau Sibu, Johor. Single cells of Gambierdiscus were isolated and clonal cultures were established. Background: It is unclear whether improving feed efficiency by selection for low residual feed intake (RFI) compromises pigs’ immunocompetence. Here, we aimed at investigating whether pig lines divergently selected for RFI had different inflammatory responses to lipopolysaccharide (LPS) exposure, regarding to clinical presentations and transcriptomic changes in peripheral blood cells.
Results: LPS injection induced acute systemic inflammation in both the low-RFI and high-RFI line (n = 8 per line). At 4 h post injection (hpi), the low-RFI line had a significantly lower (p= 0.0075) mean rectal temperature compared to the high-RFI line. Continuous monitoring of bacterial growth in aqueous media is a crucial process in academic research as well as in the biotechnology industry. Bacterial growth is usually monitored by measuring the optical density of bacteria in liquid media, using benchtop spectrophotometers. Due to the large form factor of the existing spectrophotometers, they cannot be used for live monitoring of the bacteria inside bacterial incubation chambers. In this study, a hyperbranched polyester co-polymer was designed using a proprietary monomer and diethylene glycol or triethylene glycol as monomers. The synthesis was carried out using standard melt polymerization technique and catalyzed by p-Tolulenesulfonic acid. The progress of the reaction was monitored with respect to time and negative pressure, with samples being subjected to standard characterization protocols. The resulting polymers were purified using the solvent precipitation method and characterized using various chromatographic and spectroscopic methods including GPC, MALDI-TOF, and NMR. The fungal circadian clock photoreceptor Vivid (VVD) contains a photosensitive allosteric light, oxygen, voltage (LOV) domain that undergoes a large N-terminal conformational change. The mechanism by which a blue-light driven covalent bond formation leads to a global conformational change remains unclear, which hinders the further development of VVD as an optogenetic tool. One limitation in using antibodies is their long circulation time, which can take days to weeks to fully clear from blood circulation.
