Upon irradiation, marine polysaccharides undergo depolymerization, leading to formation of oligosaccharides that elicit various biological activities in plants. Taking a step further on the previously established growth-promoting activity of irradiated sodium alginate (ISA), structural rearrangements in ISA were analyzed using complementary techniques to develop an understanding of the structure-property relationship. The essential oil (EO) of vetiver (Vetiveria zizanioides L. Nash) immensely benefits the perfumery industry, proving itself as an economically important crop. Commercially, nanoparticles are in great demand in electronics, catalysis, chemistry, energy, and medicine. Today in nanoparticle production green synthesis, using biological material including medicinal plants as a starting material, is in vogue. In the present investigation the powder extract (stems and leaves) of an important medicinal plant, Verbesina encelioides, was used for the biosynthesis of bionanoparticles. The genus Dendrobium possesses horticultural importance. Dendrobium sonia-28 is an important ornamental orchid in the flower industry. Cryopreservation is a favoured long-term storage method for orchids with propagation problems. Protocorm-like bodies (PLBs) of Dendrobium sonia-28 were cryopreserved using the vitrification technique. Histology and scanning electron microscopy (SEM) observations were conducted on stock, non-cryopreserved (control), and cryopreserved PLBs of Dendrobium sonia-28 to detect cryoinjuries resulting from the vitrification protocol. Histological observations of control PLBs indicated that the preculture, osmoprotection, and dehydration steps were not physically damaging to the PLBs. To investigate DNA and DNA-protein assembly, nucleic acids were adsorbed to freshly cleaved mica in the presence of magnesium ions. The efficiency of DNA adhesion and the distribution of the molecules on the mica surface were checked by transmission electron microscopy. In addition, various kinds of DNA-protein interactions including DNA wrapping and DNA super-coiling were analyzed using electron microscopy. Scanning electron microscopy, when applied to the surfaces of the needles of Taxus spp. (yew) revealed features that appear useful in the taxonomy of this tree species which yields the important anticancer drug, taxol. For instance, all of the four North American species have 3-5 rows of stomata on one-half of the abaxial leaf surface, whereas all of the others, including those from Europe and Asia, have 7-10 rows of stomata. The relative quantity of plastic film residues and other man-made materials in composts prepared from municipal solid wastes (MSW), biosolids, yard wastes, and pine bark was evaluated utilizing light and scanning electron microscopy equipped with energy dispersive X-ray analyzer. MSW composts screened through a 4 mm screen contained significant but highly variable quantities of plastic film residues and other man-made materials that escaped detection with the unaided eye. The other composts were free of such particles. We conclude that the database for evaluation of man-made materials in MSW composts is inadequate and must be developed further.

Scanning force microscopy (SFM) and scanning electron microscopy (SEM) were used to examine the tissue structure of the electric organ of Torpedo marmorata in air and in liquid after applying fracturing and cryosectioning techniques and chemical fixation. The electric organ is organized in columns of stacked electrocytes, arranged in a honeycomb pattern. The columns were cut along a plane normal to the cell stack and thin sections were transferred to polylysine coated glass coverslips. Exposure to intense sound produces a well-defined "patch" lesion on the chick basilar papilla in which 30-35% of the short hair cells are lost. The present study compares various aspects of sensory hair bundle morphology on surviving hair cells in the patch lesion with hair bundles from matched locations on nonexposed control papilla immediately after removal from the exposure and 12-days post exposure. A systematic, comparative study of the accessory sex glands of the adult male rat after androgen withdrawal was carried out. The changes were investigated by using scanning electron microscopy at different intervals after surgical castration. The main common signs of epithelial cell involution were flattening of the cell surface, reduction of the size and number of microvilli, some blurring of the cell borders, cessation of secretory activity and diminution of the luminal volume of the glands. This study describes the morphology of the accessory sex glands of the adult male rat as observed by scanning electron microscopy (SEM). The purpose was to obtain a systematic and comparative SEM description of these glands and to evaluate different preparation techniques. A common morphological feature is polyhedral delineation of the cells, which exhibited a variable convexity of their apical surface. The cell apices were more or less studded with microvilli. The developmental time scale of the cochlea varies from species to species. We investigate here the condition of the neonatal cochlea in the chinchilla, a species increasingly used in auditory research. We have examined the morphology of cochlear hair cells using scanning microscopy, and the development of auditory function during the first postnatal month by monitoring auditory brainstem evoked responses (ABR). We find that although there were some outer hair cell kinocilia present in middle and apical areas, the hair cells otherwise were mature at 24 hours after birth. Furthermore, cochlear auditory thresholds are adult-like at birth. However, whilst there …

The neonatal line of the enamel in human deciduous teeth was observed by scanning electron microscopy using secondary and backscattered electron signals. The neonatal line containing irregular structures of enamel prisms with disordered crystal arrangements was basically formed by the abrupt bending of the prisms towards the root. Usually, the prisms gradually bent back again to regain their previous orientation, but the prisms in the inner and the surface layer sometimes ran straight ahead after bending. Although much information about chromosome structure and behaviour has been obtained using light microscopy, greater resolution is needed for a thorough understanding of chromosome organisation. Scanning electron microscopy (SEM) can provide valuable data about these three-dimensional organelles. The introduction of methods using osmium impregnation of methanol-acetic acid-fixed chromosome spreads revolutionised matters, producing life-like images of chromosomes. Nevertheless, it became clear that osmium impregnation introduced various artefacts, although the resulting images were still useful. Methanol-acetic acid-fixed chromosomes are, in fact, flattened on the glass substratum, and the 3-dimensional appearance obtained after osmium impregnation is the result of swelling during this process. …

Cardiotoxicity is an unexplained toxic manifestation of 5-fluorouracil (5-FU). Its possible mechanism could be a direct cytotoxic effect on the vascular endothelium. We have tested this hypothesis in an experimental study in rabbits, using scanning and transmission electron microscopic evaluation of endothelium in small arteries (the central artery of the ear). The perfusion fixation method at physiological pressure and temperature was used. Both local and systemic effects of 5-FU on endothelium were studied 1, 3, 7, 14 and 30 days after in vivo treatment with 5-FU. The aim of this study was to examine the influence of different incubation media on the morphology of the endothelium of great saphenous vein grafts and establish a suitable scoring system for the evaluation of damage caused by these media. Fifty specimens of saphenous veins from ten patients during elective aorto-coronary bypass surgery were used. Ten specimens served as controls; the others were assigned to test groups and exposed to heparinized whole blood, Bretschneider's HTK, human albumin or Ringer's solution. Seventy-one chick embryos of both sexes at the 35 Hamburger and Hamilton (H-H) developmental stage were processed for scanning electron microscopy of vascular corrosion casts and of critical point dried specimens, as well as transmission electron- and light microscopy, in order to study the angiogenic structures. The gonadal subepithelial capillary network was located at the level of the tunica albuginea under the covering epithelium. Single crystal gallium arsenide (GaAs) specimens were loaded to failure. Scanning electron microscope examination of fracture surfaces showed that GaAs fails in a brittle manner on {110} planes. Features on these fracture surfaces were used to identify preexisting (critical) flaws that potentially initiated fracture when loaded by tensile stresses. Critical flaws in each specimen were identified by comparison to an intentionally damaged control. The size and shape of critical defects were consistent with existing failure models.

Regeneration of the synovial lining in the synovectomized rabbit knee was studied using the scanning electron microscope. The resected synovia regenerated considerably 3 weeks after synovectomy. However, 44 weeks following excision, their surface morphology was still very different from that of the normal tissue of intact animals. The regenerated synovia were characterized by three main features: the large number of various patterns, the many fields harboring fibrillation and the almost total lack of a bubble layer (the latter was formerly shown to be predominant on normal, intact synovia). Scanning electron microscopy (SEM) of red blood cells in whole blood samples from rats was performed following acute γ-irradiation of animals with 0.25 to 1 Gy. Increased incidence of echinocytosis was observed and found to be dose- and time-dependent. At a higher radiation dose (1 Gy), echinocytosis was revealed within 5 minutes after treatment and persisted up to 3 weeks. The data demonstrate the applicability of SEM for detecting minimal radiation-induced lesions of red blood cells.

Backscattered electron (BSE) imaging was used to display heavy metal stained biological structures of various embedded specimens. Samples were fixed, stained and embedded in resin blocks as with preparation for the transmission electron microscope (TEM). Blocks were trimmed to center the specimens in a trapezoidal face of up to 5 mm2 and their sides painted with conductive silver paint leaving the face uncovered. Blocks were sputter coated with 6-8 nm of silver, chromium or aluminum, with aluminum providing the best specimen contrast in BSE mode. Fractal dimension has been used extensively as a descriptor of the rugged outlines of fine-particles. Potentially, it may be a useful parameter for characterizing the outlines of fine-particles which have been subjected to some form of chemical degradation. Here, fractal dimension values have been computed for the outlines of microscopic lead fine-particles both before and after weak hydrochloric acid dissolution experiments. Values obtained for the post-dissolution rugged profiles were greater than those of the pristine fracture grains which had a Euclidean form. The development of nerve fiber arrangements of the organ of Corti was studied in rabbits 1, 3, 5, 7 and 12-days-old using thick sections from celloidin-embedded cochleas which were examined under a scanning electron microscope. The arrangements of nerve fibers varied with developmental age. The tunnel spiral bundle was thick and loosely collected in the immature cochlea. The outer spiral fibers were recognized even in the narrow space of Nuel in the one-day-old cochlea. As Nuel's space is extending, the fibers course along the medial side of Deiters' cells. The microstructure of sintered polycrystalline MgO·1.5Al2O3 ceramic is strongly affected by the sintering conditions due to the precipitation of α-alumina at a temperature lower than about 1400°C. Consequently, the mechanical properties, depending on grain size distribution, are greatly influenced. Three different microstructures were obtained by a two step sintering process adopting an intermediate sintering temperature of 1050°C, 1100°C and 1300°C respectively, and a final sintering temperature of 1500°C. For the cell type diagnosis of leukemia in adult patients, particularly when the sampling of bone marrow is difficult, the study of peripheral blood leukocytes (PBLs) by immuno-electron microscopy provides significant information, as illustrated here in two cases of hairy cell leukemia and seven cases tentatively identified as megakaryoblastic leukemia (M7). Indirect immunogold labeling with the B-ly7 monoclonal antibody (CD103) proved valuable in confirming the diagnosis of hairy cell leukemia. Overgrown human gingival specimens were examined histologically and by scanning electron microscopy (SEM) to study structural changes caused by cyclosporine. The biopsy specimens were from organ transplant recipients receiving cyclosporine to suppress the rejection of the transplanted organ. The epithelium of the overgrown gingiva was thickened, acanthotic and parakeratotic. Retepegs were anastomosing and extending into connective tissue. The SEM examination of the outer surface of the attached gingival showed loss of cellular attachments and cells were exfoliating. Tissues from fourteen mammary glands of eight bitches were prepared for scanning electron microscopy of vascular corrosion casts and for histology to study the vasculature of the lactating and non-lactating teats. The densely-meshed mammary dermal capillary network formed ridges and troughs. The teat ducts were vascularized by a relatively densely-meshed capillary network which drained into veins longitudinally oriented to the ducts. Between eight and fifteen teat duct openings were seen on the tip of the teat, that were sometimes divided by a septum. The cerebelli of teleost fishes, primates and humans were processed for conventional and high resolution scanning electron microscopy (SEM) to study the outer and inner surfaces of axo-dendritic, glomerular and axo-somatic synapses. The cryofracture technique, either by slow or fast freezing, exposed the hidden neuronal surfaces of synaptic connections, selectively removing the glial ensheathment. Axo-dendritic junctions of climbing fibers and Golgi axonal ramifications were studied in gold-palladium and chromium coated samples. Chromium coating showed different mass density and topographic contrast between axonal and dendritic profiles. Dental hard tissue abnormalities have never been described as part of the symptoms associated with hyperphosphatasemia. Fourteen teeth obtained from a young man, who had a mild form of hyperphosphatasemia, were analyzed using scanning electron microscopy (SEM), secondary ion mass spectroscopy (SIMS), X-ray diffraction (XRD), and infrared (IR) spectroscopy. SEM revealed a thin enamel, presenting a prismatic structure with many pits, and atypical cementum and dentin showing numerous resorption areas. The X-ray diffractograms revealed poorly crystallinehydroxyapatite associated with α-tricalcium phosphate and magnesium hydroxide phases.
