Evaluation Of Intra-Species Diversity Of Oxalobacter Formigenes Strains Using Pulsed-Field Gel Electrophoresis (Pfge) And Multiplex Pcr, Nivedita Pareek

Oxalobacter formigenes is a beneficial, strict anaerobic gut bacterium that plays an important role in the prevention of kidney stone disease with a unique characteristic of using only oxalate as a carbon and energy source. To date, twenty-one strains of O. formigenes have been isolated which are further divided into two groups based on differences in the patterns of cell membrane lipids, cellular proteins, and nucleic acid fragments. The objective of the study was to evaluate intra-species diversity of O. formigenes strains using pulsed-field gel electrophoresis (PFGE) and multiplex PCR.

Large rivers are highly important systems; being exploited both commercially and recreationally. Because of this usage by humans, close monitoring of the ecology of theses rivers is of the utmost importance. The Long-Term Electrofishing project (LTEF) monitors the fish communities of the Illinois, Mississippi, Wabash, and Ohio rivers using day time, pulsed-DC electrofishing during the late Spring through the early Fall each year. Little information is available regarding the genetic structure and diversity of sportfishes in large rivers, which can be vital to continuing the sustainable exploitation of these fisheries. Additionally, impoundments often have vast impacts on riverine systems. These impacts include altering the flow regime, changing the nutrient load, and preventing migratory fish from reaching their spawning grounds. If fishes are prevented from reaching their spawning grounds, their genetic diversity and structuring could begin to change relative to historic measures, potentially decreasing their fitness and ability to survive. In 1971, a graduate student at Eastern Illinois University, James E. Wiedman, studied the lichen flora of Rocky Branch Nature Preserve, Clark County, Illinois. During his study he collected, identified, and recorded a total of 64 lichen species. Since 1971, Rocky Branch Nature Preserve has experienced a dramatic increase in use by people and a concomittant decline in the quality of lichen habitats. In order to assess the decline of suitable lichen habitats and the reduction of lichen species a second study of the lichens at Rocky Branch Nature Preserve was undertaken. The study was conducted to determine the effects of lowered genetic variance in the Western mosquitofish (Gambusia affinis) on the physiological parameters of critical thermal maximum and minimum pH. Mosquitofish were obtained from two small central Illinois ponds, Lost Pond and Shadow Pond, by seines and dip nets. Once caught, fish were returned to the laboratory and acclimated for a minimum of five days at a temperature of 27°C and a pH of 8.25. The Illinois Department of Natural Resources currently manages approximately 4,000 hectares of natural prairie. High human impacts coupled with small patch size result in challenging management decisions for prairie conservation. The ideal restored prairie would consist of a mosaic of habitats varying in vegetation from forb-grass dominated areas to oak savannas, with concurrent stages of vegetative succession occurring across the habitats. A study was conducted at the Middle Fork State Fish and Wildlife Area located in Vermillion County, IL, to analyze small mammal diversity and community composition among three different stages of prairie restoration.

Belmont Prairie is located in Downers Grove, DuPage County, Illinois, and is one of the few preserves of a native tall grass prairie. It is located in the Morainal section of the Northeastern Morainal Division, and harbors over 100 species of native prairie plants which are typical of the community type. Three vegetation zones were sampled for species composition and both frequency and relative frequency. These areas are as follows: Dry mesic upland area, sedge meadow, and transitional area.

The benthic insect populations from Lake Charleston and the Embarras River above and below Lake Charleston were examined twice monthly for seasonal differences in number of organisms, number of taxa and species diversity from October 13, 1976 to February 16, 1977. The water quality parameters of alkalinity, carbon dioxide, dissolved oxygen, pH and temperature were also examined for each site to determine if any parameter showed a sufficient deviation as to exert an influence on the benthic insect populations.
