It is well known that arginine vasotocin (AVT) in birds is involved in physiological homeostasis such as cardiovascular, osmotic regulation as well as reproductive functions. Pertinent to these physiological functions, AVT immunoreactive (-ir) neurons in the hypothalamus have been found associated with hemorrhage, dehydration, oviposition and other physiological regulation. Evidence, however, suggests that AVT also plays significant roles in modulating behavior, memory, stress, and food intake. This dissertation research addresses the latter two neuroendocrine functions of AVT in detail within the chicken brain. The discovery of virus induced cancer has paved the way for further genetic studies. Rous Sarcoma Virus (RSV) is a tumor-causing virus that infects poultry. Poultry is one of the main sources of protein in the United States and in 2014 Americans ate approximately 100 lbs per person. However, consumption of poultry products is strongly associated with foodborne illness from Campylobacter. In the first study, chitosan, caprylic acid and their combination were evaluated as a coating treatment for the reduction of Campylobacter jejuni on poultry products. For the initial screening trials, chitosan of three different molecular weights (15-50 kDa, 190-310 kDa and 400-600 kDa) was evaluated at three concentrations (0.5%, 1.0%, 2.0%) and separately, caprylic acid was tested at 0.5%, 1.0% and 2.0% using chicken …

Poultry health has been traditionally maintained by hygenic measures, vaccinations, and the use of antibiotics. Modern husbandry management considers the use of probiotics as a natural way to protect birds against many everyday pathogens. Different strains of Bacillus spp. have proved to have beneficial effects in poultry production. However, the most used bacterium in commercial probiotics is Lactobacillus, a vegetative cell. Campylobacteriosis is one of the leading foodborne illnesses in United States, and is associated with the consumption of poultry and poultry products. Reducing Campylobacter in these species will reduce the burden of this disease. The following studies evaluated our hypothesis that transmission by the fecal-respiratory route may be a viable portal of entry for Salmonella and could explain some clinical impressions of relatively low-dose infectivity under field conditions in relation to the requisite high oral challenge dose that is typically required for infection of poultry through the oral route in laboratory studies. Initial field reports indicating tracheal sampling to be a sensitive tool for monitoring Salmonella infection in commercial flocks, suggested that tracheal contamination could be a good indicator of Salmonella infection under commercial conditions. Coccidiosis is a common enteric disease of turkeys that is caused by protozoan parasites belonging to the genus Eimeria. There are about seven species of Eimeria that affect turkeys and of these E. adenoeides is the most pathogenic and commonly recognized. Pulmonary Arterial Hypertension (PAH) is a sever disorder characterized by high pulmonary artery pressure and pulmonary vascular resistance along with specific histological changes. Idiopathic PAH is a human disease of unknown origin that showed common characteristics to PHS/ascites in chicken. PHS/ascites is a condition found in broilers causing right ventricular hypertrophy, valvular insufficiency, increased venous pressure, variable liver changes, accumulation of fluids in the abdominal cavity, and finally lung and heart failure. Growing broilers on wire flooring provides an excellent experimental model for reproducibly triggering significant levels of lameness. In Pilot Study #1 using broilers from Line C grown on wire flooring, adding the Biomin probiotic PoultryStar® to the feed reduced the percentage of lameness by half when compared with broilers that received the control diet alone. In Pilot Study # 2 using broilers from Line B grown on wire flooring, adding the PoultryStar® probiotic reduced the percentage of lameness to 8% when compared with 28% lameness in broilers that received the control diet alone. Campylobacter spp. has been identified as one of the leading causative agents of food borne diarrheal illness. Epidemiological evidence has shown that poultry is the main source for human infection. Poultry are asymptomatic carriers of Campylobacter within their gastrointestinal tract, with colonization levels reaching 106-108 cfu/g cecal content. Surveys of domestic poultry flocks have estimated that approximately 90% of flocks are positive for Campylobacter colonization. Risk assessment studies have determined that by reducing levels of Campylobacter colonization during rearing, incidences of human infection will be significantly reduced. This project is focused on determining the contributions of particular genetic loci to low sperm mobility in the chicken. Sperm mobility has been shown to be a major determinant of male fertility in broiler breeders. A whole genome SNP scan identified chromosomal regions (QTLs) that control sperm mobility. I used microsatellites from 4 chromosomal regions ( 15.151 on chromosome 6, 12.435 on chromosome Z, 12.341 on chromosome Z, and 17.214 on chromosome Z) to determine the association of these regions with sperm mobility in experimental lines divergently selected for mobility phenotype. The Smyth line (SL) of chickens is a well-established animal model for the study of spontaneous autoimmune vitiligo, displaying all the characteristics of human vitiligo. In humans, phenolic derivative 4-tertiary butyl phenol (4-TBP) has been shown to trigger vitiligo in susceptible individuals. In vitiligo-prone SL-chickens, live turkey herpesvirus (HVT) infection at hatch was identified as the most effective trigger of SL-vitiligo expression. The present study investigated the distribution of the vasotocin subtype four receptor (VT4R) in brain and pituitary gland of the chicken, Gallus gallus. The anterior pituitary cell types associated with the VT4R were also determined. Two polyclonal antibodies were raised in rabbit against a cocktail of peptides, 15 amino acids from the amino terminal region and 17 amino acids from the carboxy terminal region of VT4R receptor. The antibody was validated utilizing the Western blot and immunocytochemistry. In 1935, the National Poultry Improvement Plan (NPIP) was created to control Salmonella gallinarum and Salmonella pullorum . These two pathogens were devastating economically for poultry producers. Through cooperative efforts using vaccination and strict biosecurity, these two pathogens were eradicated from the United States. Currently, the NPIP program is targeting two other poultry pathogens, Salmonella enterica serovar Enteritidis and Mycoplasma . In the broiler industry it targets 2 specific Mycoplasma species (synoviae, gallisepticum). Excessive weight has been shown to have many negative effects, including double hierarchies, multiple ovulations, and fatty liver.
