Background

Type 2 diabetes (T2D) is the most common form of diabetes in humans and is closely associated with dyslipidemia and obesity that magnifies the mortality and morbidity related to T2D. The genetic contribution to human T2D and related metabolic disorders is evident, and mostly follows polygenic inheritance. The TALLYHO/JngJ (TH) mice are a polygenic model for T2D characterized by obesity, hyperinsulinemia, impaired glucose uptake and tolerance, hyperlipidemia, and hyperglycemia.

Results

In order to determine the genetic factors that contribute to these T2D related characteristics in TH mice, we interbred TH mice with C57BL/6J (B6) mice. The parental, F1, and …

In horses, a painful and often debilitating disease known as laminitis can result in impaired function and, in severe cases, euthanasia. Equine Metabolic Syndrome (EMS) is a syndrome in horses that results in development of laminitis and is characterized by the presence of general and/or regional adiposity (“cresty neck”), aberrations in blood lipid concentrations, insulin resistance (IR) and/ or hyperinsulinemia. Therapies have focused on improving the state of obesity and insulin resistance with the goal of diminishing the likelihood of laminitis development. For over three decades, amphibian populations have been declining across the globe. Emerging infectious diseases are responsible for some of these declines. Ranaviruses have caused die-offs in wild amphibian populations on 4 continents, in 5 Canadian provinces, and in over 25 U.S. states. In order to understand host-pathogen dynamics, it is critical to establish baseline information on species susceptibility and the effects of natural stressors. The goal of my thesis research was to quantify the effects of anuran development and exposure to invertebrate predators on species-specific susceptibility to ranavirus. The overall aim of the studies described herein was to evaluate genetic variation in cattle fertility traits for development of genetic and genomic predictors in breeding strategies. Results from these experiments suggest that improvements in fertility through genetic selection are a possible approach to increase reproductive efficiency. Experiment 1 evaluated the development of genetic parameters associated with multiple ovulation and embryo transfer schemes in an attempt to assist producers in identifying animals with greater genetic merit for these protocols. Two experiments examined growth performance and physiological measures of stress in pre- and postweaned Angus steers (313 ± 24.5 kg; n = 48/Exp.), where steers were fitted with (YD) or without (ND) an anti-suckling device (Exp. 1), or provided (YS) or not provided (NS) a grain supplement (Exp. 2) for 7 d and weaned by fenceline (FS) or total separation (TS). Mastitis, or mammary gland inflammation, causes the greatest loss in profit for dairy producers. Mastitis susceptibility differs among cows due to environmental, physiological, and genetic factors. Prior research identified a genetic marker in a chemokine receptor, CXCR1, associated with mastitis susceptibility and decreased neutrophil migration. Current research seeks to identify reasons behind mastitis susceptibility by validating this model through in vivo challenge with Streptococcus uberis and studying specific mechanisms causing impaired neutrophil migration. Holstein cows with GG (n=19), GC (n=28), and CC (n=20) genotypes at CXCR1+777 were challenged intramammarily with S. uberis strain UT888. After challenge 68% of quarters from …

Appetite control is a major issue in normal growth and in suboptimal growth performance settings. A number of hormones, in particular leptin, activate or inhibit orexigenic or anorexigenic neurotransmitters within the arcuate nucleus of the hypothalamus, where feed intake regulation is integrated. Examples of appetite regulatory neurotransmitters are the stimulatory neurotransmitters neuropeptide Y (NPY), agouti-related protein (AgRP), orexin and melanin-concentrating hormone and the inhibitory neurotransmitter, melanocyte-stimulating hormone (MSH). Examination of messenger RNA (using in situ hybridization and realtime PCR) and proteins (using immunohistochemistry) for these neurotransmitters in ruminants has indicated that physiological regulation occurs in response to fasting for several …

Broiler breeders experience a natural decline in fertility levels as a flock ages. A male management practice such as double interspiking could be applied to counteract this decline. Our objective was to investigate the effects of double interspiking on flock fertility, testosterone concentrations, stress responses, and behavioral responses of broiler breeders in heat stressed environments. Two hundred and eighty-eight broiler breeder pullets and thirty-six broiler breeder roosters (Ross 708) were assigned to three groups at 21 weeks of age (WOA). The Department of Energy Oak Ridge Reservation (ORR) and the nearby adjoining City of Oak Ridge, Tennessee had experienced a rise in deer-vehicle collisions (DVCs) to the point where safety for employees and residents became a concern. I investigated the effect of hunting, land cover, road mileage, season, lunar phase, sex, and change in traffic patterns that coincide with work shifts on DVCs from 1975 - 2008. The study area was divided into grids of 1.5 km² each for administration and data recording by managing agencies. Regional Differences In Adipose Tissue Of The Sinclair Minipig, Suzanne Lauren Booker

Adipose tissue is an endocrine organ, and its homeostatic mechanisms in normal weight, overweight and obese subjects must be elucidated. We sought to determine the basal adipose tissue biology of visceral (VIF) and subcutaneous (SQF) fat depots in 8 month old Sinclair minipigs, an animal that has been shown to be physiologically similar to humans.

Metabolic analysis showed a decrease in LDL, white blood cells (WBC), and lymphocyte percentages as the minipigs aged from 6 to 8 months (p <0.0001 and = 0.0046 and 0.0165 respectively). There were no significant changes in triglycerides, HDL, VLDL, and neutrophil percentages. There was a trend in insulin increase (P=0.0722).

It was hypothesized that technical factors associated with in vitro production (IVP) of embryos may influence rate of blastocyst development of oocytes matured at 38.5 or 41.0 C. To test this hypothesis, a retrospective meta-analysis was performed. Simple linear regression was performed to analyze continuous variables and ANOVA for categorical variables. Interactions among factors and maturation temperature on blastocyst development were analyzed using dummy regression for continuous variables, and using a factorial treatment design and ANOVA for categorical variables. This study examined the impacts of a beef cattle newsletter, “Beef Cattle Time,” on Tennessee beef producers. The purpose of this study was to assess the impacts of “Beef Cattle Time” as perceived by Tennessee beef producers on the utilization, satisfaction, benefit, and future of this newsletter. A self-developed, seventy-six question, survey was mailed to 639 randomly selected Tennessee beef cattle producers. Two hundred seventy six (43%) participants responded. One hundred thirty-four (48.6%) reported reading “Beef Cattle Time” and 142 (51.4%) had never read “Beef Cattle Time.” The utilization of “Beef Cattle Time” as a source of information was found …

Part 1: Visitation of potential pollinators to Cornus florida and C. kousa flowers was assessed in East Tennessee in 2008 and 2009. Data regarding insect visitation rates to multiple trees per location were gathered throughout the flowering period. Diurnal and seasonal variations in visitation were recorded. Pollen coverage was assessed on portions of captured insect exoskeletons that were most likely to contact the stigma. Eleven families in four insect orders were collected from C. florida and 26 families in five orders from C. kousa. The most important pollinators in eastern Tennessee were bees in the Andrenidae and Halictidae. The most …

Previous research has demonstrated circulating concentrations of leptin increase in ewes during mid pregnancy then decline in late pregnancy and early lactation. The objective of this study was to more narrowly define the timing of changes in circulating concentrations of leptin with pregnancy in ewes. Katahdin ewes (n= 19) located at latitude 34.275 and longitude -85.183 (Mount Berry, GA) were utilized. Blood samples were collected weekly via jugular veinpuncture beginning immediately before ram exposure on September 23 and continuing until 4 weeks post-lambing. Leukemia inhibitory factor (LIF) has been suggested to function as a potent inhibitor of feed intake in rodents. These studies were designed to determine whether LIF was found in the ovine hypothalamus and whether LIF inhibited feed intake in sheep. Sheep hypothalami were used to clone LIF to indicate presence of the gene in the hypothalamus. The sequence was similar to published data. Another group of sheep were provided intraventricular (ICV) cannulas and injected with doses of LIF at 250, 500, 1000 and 2500 ng per sheep, ICV. Kisspeptin (KP), a neuroendocrine regulator of gonadotropin releasing hormone, has been hypothesized as an integrator of nutrition and hormones critical to metabolism and regulation of reproduction. Recent evidence suggests growth hormone (GH) secretion may be influenced by KP. The objective of this study was to determine if the GH stimulatory effect of KP is due to actions on the hypothalamus or anterior pituitary gland in ewes. Adult ovariectomized ewes (n=8) were fitted with intracerebroventricular (ICV) cannula to facilitate central administration of experimental treatments. Bacteria in milk, whether originating from the cow or from the environment, significantly impact the quality of dairy products and therefore consumer acceptance. Bacteria produce heatstable enzymes that can damage milk fat and milk protein. These enzymes are not affected by pasteurization and continue to cause damage in the final dairy product. High bacteria levels in raw milk result in off-flavors, bitter flavors, rancidity and reduced shelf life. The Pasteurized Milk Ordinance specifies safety standards of Grade A milk to protect public health, not to maximize product quality and shelf life. The PMO only sets limits on somatic cell count …

The level of mastitis infection in a dairy herd can have a significant impact on herd profitability. Losses due to mastitis include decreased milk production, increased treatment costs, discarded milk, premature culling, death, decreased genetic potential, decreased reproductive performance, load rejection due to violation of somatic cell counts (SCC) or antibiotic residues and loss of milk quality premiums (3, 9, 12). Most producers easily relate clinical mastitis with these losses. However, subclinical (or hidden) mastitis may actually cause more losses in a herd than clinical mastitis. According to records, in 1995, there were more than 1300 dairy farms in Tennessee. In 2007, the number declined to fewer than 600 dairies (9). There are several reasons for this mass exodus from the dairy industry in Tennessee: low milk prices, shortages of qualified labor, high feed costs, high fertilizer costs, high fuel costs, etc. In addition, many dairy producers in Tennessee and in the Southern Region are at risk of losing their milk market due to below average milk quality.
