Thiamine deficiency from the consumption of invasive, high-thiaminase prey fishes is considered to be a major barrier for lake trout restoration in the Great Lakes. In fishes, an understudied aspect of thiamine deficiency is its effect on cardiac function. I examined the effects of dietary thiaminase on cardiac function and morphology in lake trout, specifically as they relate to thermal tolerance. Two hatchery strains of lake trout (Seneca and Slate) were raised on a control or thiaminase diet for nine months. Hibernation, characterized by a seasonal reduction in metabolism and body temperature, allows animals to conserve energy when environmental conditions (e.g. temperature, food availability) are unfavourable. During hibernation, small mammals such as the 13-lined ground squirrel (Ictidomys tridecemlineatus) cycle between two distinct metabolic states: torpor, where metabolic rate is suppressed by >95% and body temperature falls to ~5 °C, and interbout euthermia (IBE), where metabolic rate and body temperature rapidly increase and are maintained at euthermic levels several hours. Suppression of metabolism during entrance into torpor is paralleled by rapid suppression of liver mitochondrial metabolism. In my thesis, I …

The crop pest Drosophila suzukii, native to Southeast Asia, has been found in Ontario since 2010. This raises concern with respect to it establishing permanent populations, however, little is known about its cold tolerance. I investigated the low-temperature tolerance, including phenotypic plasticity, of D. suzukii. While acclimation increased cold tolerance, there was no evidence of short-term cold-hardening. Chill coma occurs at -1.2 °C, which will limit winter activity. Cold shock decreased the reproductive output of females, but this negative effect may be mitigated by re-mating. Drosophila suzukii is chill-susceptible and 80 % of the flies die after exposure …

Anthropogenic forcing, such as ocean acidification caused by rising carbon dioxide emissions, and eutrophication due to increased nutrient loadings in run-off, are causing major changes to the biogeochemistry of the oceans. As a consequence, coastal phytoplankton are susceptible to altered biogeochemical environments. This study examined the effect of a lower pH and increased levels of nutrients on the common coastal harmful alga, Heterosigma akashiwo. Growth rates, maximal cell yields, neutral lipid accumulation and toxicity of cells grown under various pH and nutrients regimes were measured. Migratory birds complete amazing journeys between their breeding and wintering grounds. Each journey comprises a series of flights that last hours to days, followed by stopovers where fuel stores are replenished. Despite the long flights undertaken by migratory birds, where respiratory water losses are high for extended periods of time, birds are not dehydrated after flight. My studies demonstrate that birds maintain hydration by modulating rates of endogenous water production in response to rates of water loss. Although fat deposition during stopover in migrating passerine birds has been extensively studied, changes in lean mass during refuelling are not well understood. I used quantitative magnetic resonance (QMR) analysis to measure the deposition of fat and lean mass for both recaptured and single capture migrant passerines in spring and fall at Long Point, Ontario. Both the recapture analysis and single capture regression analysis indicated a substantial contribution of lean mass to overall increases in total body mass. Some of the variation in the relative deposition of fat and lean mass was explained by sex, age and season. I then …

During hibernation, daily torpor, and fasting, mammals reduce metabolic rate (MR) up to 99%, 95%, and 30%, respectively, compared to resting levels. Mitochondrial metabolic suppression likely contributes to this MR reduction, and the first objective of this study was to determine the relative contributions of active, regulated inhibition and passive thermal effects as body temperature (Tb) falls, to mitochondrial metabolic suppression, and to examine the mechanisms involved using top-down elasticity analysis and novel statistical approach.
