Connections Between Mechanosensitive Ion Channel Msl10 And Er-Plasma Membrane Contact Sites, Jennette Marie Codjoe

Mechanosensitive (MS) ion channels are an evolutionarily conserved way for cells to sense mechanical forces and transduce them into ionic signals. A plasma membrane-localized MS channel from Arabidopsis thaliana, MscS-Like (MSL)10, senses cell swelling and initiates a signaling cascade that triggers programmed cell death. Whereas the channel properties of MSL10 have been well studied, how MSL10 signals remains largely unknown. I worked collaboratively to show that important lesions for cell death signaling in the cytosolic N- and C-terminal domains of MSL10 interact genetically. I also helped show that ionic flux through MSL10 is dispensable for signaling, which suggested that MSL10 …

Through oxygenic photosynthesis, cyanobacteria, algae, and plants convert light into chemical energy. However, highly energetic light often damages the photosynthetic apparatus, which can lead to the decline of photosynthetic activity (photoinhibition), and prolonged photoinhibition can lead to cell death. Cyanobacteria enact various protective mechanisms to mitigate photodamage, many of which have been conserved in plants. Repair of PSII reaction centers, decreased light absorption, quenching of excess absorbed energy, and biosynthesis of antioxidants all work to mitigate damage during high light. Plant roots are associated with a diverse microbial community of bacteria and fungi. The root microbiome communities associate with the root surface or penetrate the superficial layers of root tissues. It is of high agronomic interest to characterize root microbiomes and identify microbes with beneficial effects on plant defense against pathogens. Soybean taproot decline (TRD) is an emerging soil-borne disease caused by a fungus classified as Xylaria necrophora, challenging to control and with devastating effects on yield. Sustainable practices were evaluated for the production of two warm season vegetable crops, tomato and chile peppers. The first study investigated the plant vegetative growth, fruit production timing, yield components and fruit quality of three hybrid and three heirloom tomato cultivars grown in a high tunnel using grafted and non-grafted plants. Grafting the selected tomato cultivars with the two rootstock types did not alter total marketable yield of any cultivar, but affected overall stem diameter, fruit color, and β-carotene concentrations of tested tomato cultivars. The fragipan layer in the soil is a naturally occurring subsurface layer that restricts water percolation as well as inhibits root growth. A fragipan layer can be found almost ubiquitously across Southwestern Kentucky. This limits how agriculturalists manage soils in this area. It has been recently discovered by the University of Kentucky that Annual Ryegrass roots produce 3,4 dihydroxyphenylpropionic acid (DHPPA) that reacts with this hardpan to weaken the layer. Certain bacteria, such as Escherichia coli, Lactobacilli, and Bifidobacteria can also synthesize DHPPA through a breakdown process of chlorogenic acid. Increasing quantities of dust emitted from semi-arid soils, agricultural soils, and urban regions are blown to remote mountain lakes in the American West. Remote mountain lakes lacking local nutrient inputs and presenting simple food webs that are easily affected by climate changes. Dust can carry nutrients (e.g., nitrogen and phosphorus) to mountain lakes and potentially enhance algae growth and change algal communities. However, experimental tests of this hypothesis are lacking. Paraquat is the only herbicide labeled for postemergence control of Italian ryegrass in sugarcane. In 2017, sugarcane farmers in Louisiana reported Italian ryegrass control failures with paraquat applications. Seeds were collected from sugarcane fields in White Castle, St. Gabriel, and Bunkie, Louisiana where control was not achieved (reduced susceptibility). Additionally, Italian ryegrass seeds were collected from a sugarcane field in Welcome, LA with a history of paraquat use and with no reported control issues (susceptible population). Botrytis bunch rot, caused by Botrytis cinerea, is a fungal disease that primarily affects the fruit of wine grapes. Infection of fruit consequently results in reduced yields and wine quality. These factors lead to significant economic losses for growers which prompts the implementation of management practices to control the disease. The first article examines the demand side of CA and explores whether urban maize (Zea mays L.) consumers in the Democratic Republic of the Congo (DRC) would be willing to pay a premium for CA-produced maize flour. Crop yield often varies within a field of a single genetically uniform crop plant, with the causes presumed to be a mix of both biotic and abiotic factors. Manipulating crop root mycobiomes could potentially increase yield by reducing pathogen impacts and improving access to soil water and nutrients. This study aimed to identify different fungal inoculation treatments that could increase the growth of corn seedlings sown in low productivity soils to that in high productivity soils and shift the root mycobiome composition. To combat herbicide resistance among weeds, non-herbicide methods of control, such as cover crops, are becoming widely adapted. Experiments were conducted to determine how to effectively establish and manage cover crops in order to suppress tall waterhemp and Italian ryegrass and to assess their overall impact on soybean growth and yield. Various cover crop establishment methods were evaluated, and it was determined that interseeding at the R7 growth stage of soybean was the least effective method for proper cover crop establishment. Cover crops are instrumental in nutrient management and soil health. A field experiment was conducted from fall 2020 to summer 2021 to evaluate the efficiency of different cover crop termination techniques and supplemental nitrogen on sweet corn yield, ear quality and nitrogen use efficiency. The four termination types are herbicide and disk tillage (HDT), herbicide no-till (HNT), cultivation disk till (CDT), and roller crimped no-till (RCNT). Poa annua L. is ranked the most troublesome turfgrass weed but can also be a highly desirable turfgrass species. As the Latin name annua implies, the species is thought to persist via an annual life cycle; yet there are many reports in literature of P. annua persisting perennially. Cost-effective weed suppression is an important consideration for tomato growers. Growers often choose methods which minimize hand labor, as hand weeding can be prohibitively expensive. This project determined economic viability of high tunnel tomatoes treated with several methods of weed control, both organic and chemical. These methods included: 2-week hand weeding, 1-week hand weeding, preemergent, straw, landscape fabric, and untreated weedy control plots. These treatments were applied to randomized blocks in a high-tunnel. Weeding, planting, and harvest were all timed to determine labor and material costs of weed management strategy implementation. The muscadine (Vitis rotundifolia syn. Muscadinia rotundifolia) is a grape species native to the southeastern United States. Muscadines are one of three grape species in subgenus Muscadinia with a chromosome number of 2n=40 (V. rotundifolia, V. munsoniana, and V. popenoei), making them genetically distinct from the European wine and table grape (Vitis vinifera) and other species in subgenus Euvitis. Rooting hardwood cuttings from muscadine vines has traditionally been considered an exceptionally difficult task. Bacterial panicle blight (BPB) and sheath blight (SB) are major diseases of rice causing significant yield reduction in conducive conditions. Few cultivars have shown reliable resistance to these diseases. In this study, a recombinant inbred line (RIL) population derived from the U.S. cultivars Jupiter (moderately resistant) and Trenasse (susceptible) was investigated to identify genomic regions associated with BPB and SB resistance. Phenotypes in BPB and SB, as well as days to heading (DH), of the RIL population were evaluated in the field. DH was found to be correlated to BPB and SB diseases. BPB was also positively correlated to SB. …

A study was conducted to evaluate the performance of Ocimum basilicum L. in both aquaponic and soil systems in the UAE climatic condition. The experiments were conducted under greenhouse conditions. Plants were raised in aquaponic beds with ornamental fishes in different concentrations. At the same time, a set of plants were raised in pots under a greenhouse set-up. The studied parameters were morphological (total weight and leaf number, shoot and root length, number of branches), biochemical (chlorophyll a, chlorophyll b, total chlorophyll, anthocyanin, xanthophyll, protein, and phenol contents) and antioxidant levels (Ascorbic acid and tocopherol).
