Differential Expression Of Cyb5a In Chinese And European Pig Breeds Due To Genetic Variations In The Promoter Region..Pdf, Ying Bai, Jibin Zhang, Y Xue, Yl Peng, Gang Chen, Meiying Fang

Jibin Zhang

Cytochrome b5 (CYB5A) is an important electron transfer protein with homologues in a number of different organisms. In pigs, CYB5A is related to boar taint because of its role in androstenone biosynthesis. To determine the variety of CYB5A expression in pig breeds, genetic variations in the porcine CYB5A promoter region in both Chinese and European pig breeds were examined. Three single nucleotide polymorphisms (NC_010443.4:
g.165901487delG, g.165901767T>C and g.165902078C>T) were identified in the porcine CYB5A promoter region. These SNPs occurred in different frequencies in Chinese and European pigs. Chinese pigs were primarily haplotype B (denoted as delG-C-T: the position of nt …

Differential Expression Of Cell Cycle Regulators During Hyperplastic And Hypertrophic Growth Of Broiler Subcutaneous Adipose Tissue.Pdf, Jibin Zhang, Yeunsu Suh, Young Min Choi, Paula Chen, Michael Davis, Kichoon Lee

Jibin Zhang

Hyperplasticgrowth and hypertrophicgrowth within adiposetissue is tightly associated with cellcycle activity. In this study, CCNG2 and CDKN2C were found to be correlated with cellcycle inhibition during fat cell differentiation, whereas CCND3, CCNA1, and ANAPC5 were positively associated with cellcycle activity during fat cell proliferation after selection based on GEO datasets available on the NCBI website. The findings were validated through comparison of expressions of these genes among different tissues/fractions in broiler chickens and time points during primary cell culture using quantitative real-time PCR. Development of broilersubcutaneousadiposetissue was investigated on embryonic days 15 and 17 and on post-hatch days 0, 5, …

Secretoryfactors in animals play an important role in communication between different cells, tissues and organs. Especially, the secretoryfactors with specific expression in one tissue may reflect important functions and unique status of that tissue in an organism. In this study, we identified potential tissue-specificsecretoryfactors in the fat, muscle, heart, lung, kidney and liver in the mouse by analyzing microarray data from NCBI's Gene Expression Omnibus (GEO) public repository and searching and predicting their subcellular location in GeneCards and WoLF PSORT, and then confirmed tissue-specific expression of the genes using semi-quantitative PCR reactions. With this approach, we confirmed 11 lung, 7 …

Increase of fat cells (FCs) in adipose tissue is attributed to proliferation of preadipocytes or immature adipocytes in the early stage, as well as adipogenic differentiation in the later stage of adipose development. Although both events are involved in the FC increase, they are contrary to each other, because the former requires cell cycle activity, whereas the latter requires cell cycle withdrawal. Therefore, appropriate regulation of cell cycle inhibition is critical to adipogenesis. In order to explore the important cell cycle inhibitors and study their expression in adipogenesis, we adopted a strategy combining the Gene Expression Omnibus (GEO) database available …

A Comprehensive Microrna Expression Profile Of The Backfat Tissue From Castrated And Intact Full-Sib Pair Male Pigs.Pdf, Ying Bai, Jinming Huang, Gang Liu, Jibin Zhang, Jianying Wang, Chengkun Liu, Meiying Fang

Jibin Zhang

Background: It is widely known that castration has a significant effect on the accumulation of adipose tissue. microRNAs (miRNAs) are known to be involved in fat deposition and to be regulated by the androgen-induced androgen receptor (AR). However, there is little understanding of the relationship between miRNAs and fat deposition after castration. In this study, the high-throughput SOLiD sequencing approach was used to identify and characterize miRNA expression in backfat from intact and castrated full-sib male 23-week-old pigs. The patterns of adipogenesis and fat deposition were compared between castrated and intact male pigs.
Results: A total of 366 unique miRNA …

Three single nucleotide polymorphisms (SNPs) at -660bp (deletion of a single guanine base), -380bp (T>C), -69bp (C>T) were identified by sequencing cDNA of CYB5 genes in different Chinese and Foreign pigs. After subsequent detection using microsatellite, pyrosequencing and restriction fragment length polymorphism (RFLP) techniques in large populations of different pig breeds, we found that the mutations in all the SNP sites were favorable in Chinese pigs except LiCha black pigs which were crossed with Landrace pigs before, whereas the wild type nucleotides were favorable in Foreign pigs including Large White, Landrace and Duroc pigs. Therefore, we conclude that these SNPs …

Interactions Between Pieris Oleracea And Pieris Rapae (Lepidoptera: Pieridae) Butterflies, And The Biological Control Agents Cotesia Glomerata And Cotesia Rubecula (Hymenoptera: Braconidae)., Megan Herlihy Jan 2013

Interactions Between Pieris Oleracea And Pieris Rapae (Lepidoptera: Pieridae) Butterflies, And The Biological Control Agents Cotesia Glomerata And Cotesia Rubecula (Hymenoptera: Braconidae)., Megan Herlihy

Megan V Herlihy

Pieris oleracea, formerly Pieris napi, was once a widespread pierid butterfly in New England until the introduction of a biological control agent, Cotesia glomerata. It has been suggested that C. glomerata is responsible for the range reduction of P. oleracea. There are been several introductions of a second more specialized biological control agent, Cotesia rubecula, to the United States since the 1960’s. My first goal was to determine the current distribution and status of P. rapae parasitoids and the effectiveness of C. rubecula as a biological control agent since its release. The findings of a survey I conducted of the …

Distribution Of Cotesia Rubecula (Hymenoptera: Braconidae) And Its Displacement Of Cotesia Glomerata In Eastern North America, Megan V. Herlihy

Megan V Herlihy

A survey was conducted from May to Oct of 2011 of the parasitoid community of the imported cabbageworm, Pieris rapae (Lepidoptera: Pieridae), in cole crops in part of the eastern United States and southeastern Canada. The findings of our survey indicate that Cotesia rubecula (Hymenoptera: Braconidae) now occurs as far west as North Dakota and has become the dominant parasitoid of P. rapae in the northeastern and north central United States and adjacent parts of southeastern Canada, where it has displaced the previously common parasitoid Cotesia glomerata (Hymenoptera: Braconidae). Cotesia glomerata remains the dominant parasitoid in the mid-Atlantic states, from …

Phylogenetics Of Morus (Moraceae) Inferred From Its And Trnl-Trnf Sequence Data., Madhav Nepal

Madhav Nepal

Morus (Tribe Moreae, Moraceae) consists of ca. 13 species of trees distributed in Asia, Africa, Europe, and North, Central, and South America. The broad geographical distribution of the genus, overlapping ranges of many taxa, and documented hybridization between some species present interesting questions of taxonomy, phylogeny, and biogeography. Phylogenetic data for Morus also contribute to higher level taxonomic work in the family. We used sequence data from ITS of the nrDNA and the chloroplast trnL-trnF intergenic spacer to study phylogenetic relationships of Morus. Phylogenies based on separate data sets were not statistically incongruent, and the combined tree …

Expression Analysis Of Hif-1a And Hif-2a Genes In Tibetan Chicken Under Normoxia And Hypoxia.Pdf, Yunzhou Yang, Ying Bai, Xinxing Dong, Jibin Zhang, Meiying Fang

Jibin Zhang

Tibetan chicken is one of those chicken breeds that could adapt to high altitude and low oxygen pressure environment; it owes to an integrative genetic mechanism for hypoxia adaptability compared to lowland chicken breeds. HIF-1α and HIF-2α are the central factors playing important roles in maintaining organisms’s oxygen homeostasis. In this study, the expression of HIF-1α and HIF-2α genes were investigated by Real Time-PCR in Tibetan chicken and Dwarf chicken embryo brain tissue under normoxia (21% oxygen concentration) and hypoxia (13% oxygen concentration). The results showed that Tibetan chicken always had lower mortality than Dwarf chicken during the incubation. We …

The fat mass and obesity associated gene (FTO) has been widely reported to be associated with fat mass or fat deposition in different species. In the present study, we cloned both promoter and codingregions of the gene in pigs with over 5 Kb of sequence for the former region and 1,596 bp for the latter region. Comparative analysis of the promoter region among 20 species including pig revealed four conserved regions that harbor transcriptional factors involved in adipose differentiation. Using a pooled DNA sequencing approach, we discovered 39 single nucleotide polymorphisms (SNPs) in the pig FTO gene and four of them were …

3β-hydroxysteroid dehydrogenases (HSDs) and 17βHSDs are pivotal enzymes involved in synthesis, activation and deactivation of steroid hormones. Therefore, they play an important role to maintain homeostasis of hormone and regulate the synthesis and metabolism of hormones, and they are also related to metabolism of androstenone which can cause boar taint in male pigs. Through ligation of different PCR fragments of these genes from Large White pigs to pGL3-basic plasmid and subsequent transient transfection of constructed vectors into HepG2 cell culture, we found that the fragment from -1038bp to 45bp in porcine 3βHSD gene and the fragment from -966bp to 39bp …
