In the last 15 years, crustacean fisheries have experienced billions of dollars in economic losses, primarily due to viral diseases caused by such pathogens as white spot syndrome virus (WSSV) in the Pacific white shrimp Litopenaeus vannamei and Asian tiger shrimp Penaeus monodon. To date, no effective measures are available to prevent or control disease outbreaks in these animals, despite their economic importance. Recently, double-stranded RNA-based vaccines have been shown to provide specific and robust protection against WSSV infection in cultured shrimp. However, the limited stability of double-stranded RNA is the most significant hurdle for the field application of these …

The use of skin virome offers a unique approach for human identification purposes in instances where a viable and statistically relevant human DNA profile is unavailable. The skin virome may act as an alternative DNA profile and/or an additional form of probative genetic material. To date, no study has attempted to investigate the human virome over a time series across various physical locations of the body to identify its diagnostic potential as a tool for human identification. For this study, we set out to evaluate the stability, diversity, and individualization of the human skin virome. An additional goal was to …

Intracellular Fate Of Bovine Milk Exosomes In Murine Bone Marrow-Derived Macrophages, Afsana Khanam, Janos Zempleni
Department of Nutrition and Health Sciences: Faculty Publications
Milk exosomes (MEs) and their microRNA cargos constitute novel bioactive food compounds, and bovine MEs (BMEs) are being considered for use in drug delivery. The internalization of MEs by macrophages and degradation in lysosomes (in all cells) limit biological activities. Here, we determined whether BMEs internalized by murine bone marrow-derived macrophages (BMDMs) retrofuse with the intralumenal membrane in the multivesicular body (MVB) for subsequent release into the extracellular space or whether BMEs are destined for degradation in lysosomes.
Conclusions: The retrofusion of BME-derived intralumenal MVBs and secretion of BME into the extracellular space is quantitatively minor in BMDMs. The majority …

The 1990s saw the rapid reemergence of malaria in Amazonia, where it remains an important public health priority in South America. The Amazonian International Center of Excellence in Malaria Research (ICEMR) was designed to take a multidisciplinary approach toward identifying novel malaria control and elimination strategies. Based on geographically and epidemiologically distinct sites in the Northeastern Peruvian and Western Brazilian Amazon regions, synergistic projects integrate malaria epidemiology, vector biology, and immunology. The Amazonian ICEMR’s overarching goal is to understand how human behavior and other sociodemographic features of human reservoirs of transmission— predominantly asymptomatically parasitemic people—interact with the major Amazonian malaria …

Diversity And Phylogenetic Relationships Of ‘Tetraphyllidean’ Clade 3 (Cestoda) Based On New Material From Orectolobiform Sharks In Australia And Taiwan, Janine N. Caira, Kirsten Jensen
Harold W. Manter Laboratory of Parasitology: Library Materials
In an effort to expand knowledge of Clade 3—one of the ten clades that compose the non-monophyletic order ‘Tetraphyllidea’ all current members of which parasitise orectolobiform sharks—we targeted species of orectolobiform sharks that had not previously been examined for ‘tetraphyllidean’ cestodes. That work led to the discovery of three new species off Australia and Taiwan. Ambitalveolus gen. n. was erected to accommodate these species. Ambitalveolus costelloae gen. n. et sp. n., Ambitalveolus kempi sp. n., and Ambitalveolus penghuensis sp. n. differ from one another in scolex size, genital pore position, and number of marginal loculi, proglottids, and testes. Among ‘tetraphyllideans’, …

Sesame is classified as a “major” food allergen for which mandatory disclosure is required. Understanding reaction thresholds and how these vary within the allergic population is crucial in providing appropriate dietary advice to patients, providing guidance to the food industry, and informing dosing regimens for oral food challenges (FC). However, the largest data series used to derive a threshold dose-distribution for sesame included blinded challenge data from just 40 individuals.1 Data from low-dose, open FC can be used to supplement that from blinded FC, reducing uncertainty in estimating threshold dose-distributions for allergenic foods which otherwise lack sufficient data.2 …
Circling The Drain: The Extinction Crisis And The Future Of Humanity, Rodolfo Dirzo, Gerardo Ceballos, Paul R. Ehrlich
Circling The Drain: The Extinction Crisis And The Future Of Humanity, Rodolfo Dirzo, Gerardo Ceballos, Paul R. Ehrlich
Harold W. Manter Laboratory of Parasitology: Library Materials
Humanity has triggered the sixth mass extinction episode since the beginning of the Phanerozoic. The complexity of this extinction crisis is centered on the intersection of two complex adaptive systems: human culture and ecosystem functioning, although the significance of this intersection is not properly appreciated. Human beings are part of biodiversity and elements in a global ecosystem. Civilization, and perhaps even the fate of our species, is utterly dependent on that ecosystem’s proper functioning, which society is increasingly degrading. The crisis seems rooted in three factors. First, relatively few people globally are aware of its existence. Second, most people who …
Death Scene Insect Succession In Nebraska: A Guidebook, Erin Bauer, Larry Barksdale, Emma Sidel, Justine Laviolette
Death Scene Insect Succession In Nebraska: A Guidebook, Erin Bauer, Larry Barksdale, Emma Sidel, Justine Laviolette
Department of Entomology: Faculty Publications
Insect behavior can be helpful to law enforcement in determining time of death, manner of death, location, and environment related to human or other animal victims found at a death scene. They may also provide clues about other aspects associated with an investigation (i.e., fly specks, suspect DNA). The study of how insects and related arthropods can aid in legal investigations is known as forensic entomology. Although this includes both civil applications, such as urban (i.e., maggots in mortuaries or insect structural damage) or stored product (i.e., illness from food contamination) entomology, this manual focuses on criminal applications, such as …
Beyond Homogenous Decision-Making Models: Role Of Brain Areas Interaction And Heterogeneity, Mohsen Rakhshan
Beyond Homogenous Decision-Making Models: Role Of Brain Areas Interaction And Heterogeneity, Mohsen Rakhshan
Dartmouth College Ph.D Dissertations
Most existing mechanistic models of decision making consider microcircuits within a homogenous brain area and fail to consider the distributed nature of this cognitive process. More specifically, these models do not take into account: a) how different types of information might influence choice in different stages of the process; and b) how interactions and the heterogeneity between circuits across different brain areas might be essential for the process. In this dissertation, I present a combination of experimental and modeling approaches to study distributed nature of the decision-making by examining both the distributed processing of information as well as the interaction …
Profiling Durable Anti-Tumor Memory T Cell Responses In Long-Term Melanoma Survivors, Jichang Han
Profiling Durable Anti-Tumor Memory T Cell Responses In Long-Term Melanoma Survivors, Jichang Han
Dartmouth College Ph.D Dissertations
While T-cell responses to cancer immunotherapy have been avidly studied,
long-lived memory has been poorly characterized. Of melanoma patients who
receive immunotherapy, long-term survivors are frequently found to develop
melanoma-associated vitiligo. Our prior work showed in a preclinical model that
vitiligo skin sustained a long-lived CD8+ resident memory T cell (TRM) population,
playing key roles in perpetuating anti-tumor immunity. However, the characteristics
and longevity of these memory T cells in melanoma survivors have not been defined.
In the present studies, we probed both the CD8+ and CD4+ T cell responses,
focusing on memory T cell responses in a cohort of …

Unlocking The Genomic Regions Associated With Seed Protein Contents In Turkishcommon Bean Germplasm Through Genome-Wide Association Study, Faheem Shahzad Baloch, Muhammad Azhar Nadeem
Turkish Journal of Agriculture and Forestry
Human body needs sufficient quantity of protein on daily basis for the normal functioning. Dietary protein in sufficient quantity is becoming a key issue for huge population of the world particularly in developing countries. Present investigation aimed to explore the phenotypic variation and to investigate the genomic regions associated with seed protein contents in Turkish common bean germplasm collected from 19 provinces. Field experiments were conducted according to augmented block design at two locations (Bolu and Sivas) for two consecutive years (2017 and 2018). Analysis of variance revealed highly significant environmental effects (p < 0.05) on seed protein contents within environments, while genotype by environment interaction revealed nonsignificant effects. Overall mean protein contents were 36.36%, while Balikesir-3 and Hakkari-38 yielded minimum (31.64%) and maximum (40.70%) protein contents. Among 19 provinces, accessions from Hakkari and Erzincan provinces disclosed maximum and minimum mean protein contents respectively. Scatter plot between seed protein contents and plant height divided the accessions according to their protein contents and plant height. A total of eight stable accessions were selected and can be recommended for future breeding activities. The implemented constellation plot separated the studied germplasm into two populations on the basis of their protein contents. Genotyping by sequencing resulted 7900 DArTseq markers was used for marker-trait association and a total of 11 markers showed significant association for protein contents. During this study, DArT-3365703 marker was identified in both locations (Sivas and Bolu) during 2017 and might be used for future common bean breeding. Physical map revealed the presence narrow regions between identified markers on Pv02 and Pv05. Therefore, these regions should be considered for future breeding activities. We are confident that results of present study will be helpful for marker-assisted breeding of common bean.

The Cappadocia Region in Central Anatolia, Turkey is rich in wild apricot genetic resources. In this study, a multivariate analysis method of metric and nonmetric multidimensional scaling analysis (MDS) was used to classify 43 wild apricot genotypes and 5 standard cultivars by using Euclidean distance. MDS was applied, based on dissimilarities, to quantitative traits of weight, height, width and thickness of fruit, stone and kernel weight, flesh/stone ratio, total soluble solids content, titratable acidity and pH in fruit juice, and sensory attributes of fruit size and shape, cavity depth, suture (cheek line), fruit apex, fruit attractiveness, skin pubescence, ground and …

The commercialization of mushrooms is critical because they have a short shelf life. For this reason, we investigated the effect of agar-agar and egg white protein on weight loss, polyphenol oxidase and catalase, protein content, total phenolics, total antioxidant capacity, respiration rate, ethylene production and storability of button mushrooms. Our findings suggested that the treatment of edible coating delayed remarkably senescence and maintained the quality of button mushroom. Egg white protein coating was more effective on protein content, total phenolics, antioxidant capacity, respiration rate, and ethylene production than agar-agar coating. On the other hand, agar-agar coating was more effective on …

Turkey has very rich potential in terms of wild edible fruit species, especially berries that grow naturally in different parts of the country as population or scattered shrubs. One of the most important wild edible fruits found in Turkey's flora is the European cranberrybush (EC). All EC plants are found as wild in nature and there are no commercial EC orchards in the country. In this study, a total of 22 seed propagated EC genotypes that naturally grown in the Akkışla district of Kayseri province located in the middle part of Turkey were assessed in terms of their morphological (fruit …

Salinity is one of the major environmental constraints affecting agriculture in major regions of the world. This study was conducted to evaluate the effect of exogenous nitric oxide (NO) treatments on two chickpea cultivars (Cagatay and Inci) under salt stress conditions. Different NO doses (0. 75 and 100 μM sodium nitroprusside (SNP)) as an NO source were applied to chickpea plants grown under saline (presence of 50 and 100 mM of NaCl) and nonsaline conditions. In this study, plant shoot fresh and dry weight, root fresh and dry weight, chlorophyll a, b, total chlorophyll, chlorophyll reading value (CRV), relative water …

The aim of the present study is to evaluate diversity among the Turkish local pea germplasm for mineral concentrations and agro-morphological traits for breeding programs. The experiment was conducted during 2018/2019 and 2019/2020 and arranged according to an augmented block design with 40 local genotypes and two standard check cultivars (Carina and Jof). Seed yield per plant was positively and significantly correlated with branches per plant, pods per plant, seeds per pod, harvest index, and 100-seed weight. The first three principal components (PC1) for agro-morphological traits accounted for 76.3% of the total variation. The principal first component was positively correlated …
