Heme is an essential cofactor required for a plethora of cellular processes in eukaryotes. In metazoans the heme biosynthetic pathway is typically partitioned between the cytosol and mitochondria, with the first and final steps taking place in the mitochondrion. The pathway has been extensively studied and its biosynthetic enzymes structurally characterized to varying extents. Nevertheless, understanding of the regulation of heme synthesis and factors that influence this process in metazoans remains incomplete. Secondary fermentation of beer is traditionally used to condition and refine the sensory profile of a beer. During methods such as barrel aging or lagering, the yeast continues to produce flavor-active compounds. Some methods include the intentional inoculation of microorganisms or the utilization of natural consortia associated with a barrel, fruit, or additive for the generation of desired sensory profiles. Yeasts are single-cell, eukaryotic microorganisms classified in the fungi kingdom. They are generally around 10μm in size, have a nuclear membrane, a cell wall, and a cytoplasmic content. Yeasts are characterized as heterotrophs, which means they rely on organic material as sources of energy and nutrients. Not all yeasts are equal. There are about 60 genera and about 1,500 species of yeasts. Only a few are used commercially. Stop codon readthrough occurs via genetic and epigenetic mechanisms, resulting in a longer polypeptide chain at the C-terminus. Although these readthroughs may seem like an error made by translational mechanisms, evidence from yeast suggests that stop codon readthrough has an impact on various cellular processes. Readthrough has the potential to create genetic diversity, similar to RNA splicing, mRNA editing, and protein modification. The diversity created by translational readthrough may result in a beneficial change in phenotype, and thus have a role in evolution and adaptability. Organisms such as Saccharomyces cerevisiae can regulate the mechanisms of proteins through post-translational modification. These modifications play a vital role in functional proteomic activity because they can regulate protein activity, localization, and interaction with other cellular molecules. Such modifications include phosphorylation, methylation, and acetylation. The metabolic mechanisms of yeast became of keen interest to our lab because our lab noticed many stress defense proteins were being acetylated during stress heat shock. Saccharomyces cerevisiae is a microorganism that is commonly used in industries, but its productivity is significantly hindered by its inability to resist high stress levels found in industrial processes. By increasing stress tolerance in S. cerevisiae, its role in industrial processes could be maximized. The objective of this study is to determine if complementation of lipocalin genes into the wildtype and knockout strains of S. cerevisiae can increase stress tolerance against a variety of stressors. We identified the knockout yeast strains, ALD3, ALD4, PDX3, and ILV1, as being sensitive to various stressors including salinity, heat, oxidative, and osmotic stresses. Arabidopsis …

The root cause for their evolution comes from selective environmental pressures that see organisms with beneficial mutations survive otherwise deadly encounters or outperform members of its population who fail to adapt. Many microbreweries practice serial fermentation, or serial repitching. Serial repitching is the process by which one yeast culture is reused for multiple batches of beer. Each batch of subsequent beer is called a “pitch.” This technique helps breweries limit production costs. However, fermentation is difficult to predict throughout serial fermentation. This leads to beer quality issues such as fluctuation in alcohol production, buildup of unwanted flavor compounds, and decreased carbon dioxide production, which results in reduced profit. A total of 1,188 pigs (L337 × 1050; PIC; initial BW of 11.1 lb) were used in a 40-d trial to determine the effect of a combination of mannan oligosaccharides, yeast culture, and beta-glucans (AgriMune Liquid; AFI-Agri Feed International, L.L.C., Rockwell, IA) on growth performance, morbidity, and mortality of pigs during the nursery period. Pigs were weaned at approximately 21 d of age and placed in pens (27 pigs per pen) based on initial BW. As organisms respond to changes in their environment genetic variation between individuals can directly affect organismal trait phenotypes by altering gene expression. Historically, studies have focused on the effect of genetic variation on mRNA synthesis (transcription) and decay rates. Relatively few studies have probed the relationship between DNA variants and protein-specific regulation of individual genes, despite the plethora of evidence that RNA levels are often poor proxies for protein levels.
