A 7-year-old spayed female domestic shorthair cat was presented because of post-prandial regurgitation, gasping shortly after eating, weight loss, dyspnea, and tachypnea. Six months prior to the onset of these signs the cat had received nutritional support via gastrostomy and then nasogastric tubes. A diagnosis of esophageal diverticulum was made on the basis of clinical features, contrast esophagram, and endoscopy, and was considered a complication of tube feeding. Medical therapy, including parenteral fluids, ranitidine, metoclopramide, and cisapride, and a soft, bland low-fat diet resulted in improvement in the clinical signs within 1 week. The cat was discharged and recovery was …

The Guinea Pig Ileum Lacks The Direct, High-Potency, M2-Muscarinic, Contractile Mechanism Of The Mouse Ileum, Michael T. Griffin, Minoru Matsui, Rennolds S. Ostrom, Frederick J. Ehlert
Pharmacy Faculty Articles and Research
We explored whether the M2 muscarinic receptor in the guinea pig ileum elicits a highly potent, direct-contractile response, like that from the M3 muscarinic receptor knockout mouse. First, we characterized the irreversible receptor-blocking activity of 4-DAMP mustard in ileum from muscarinic receptor knockout mice to verify its M3 selectivity. Then, we used 4-DAMP mustard to inactivate M3 responses in the guinea pig ileum to attempt to reveal direct, M2 receptor-mediated contractions. The muscarinic agonist, oxotremorine-M, elicited potent contractions in ileum from wild-type, M2 receptor knockout, and M3 receptor knockout mice characterized by negative log EC50 (pEC50) values ± SEM of …
Arbuscular Mycorrhizal Fungi: Factors Affecting Soil Organic Matter Storage, Carl Lee Rosier
Arbuscular Mycorrhizal Fungi: Factors Affecting Soil Organic Matter Storage, Carl Lee Rosier
Graduate Student Theses, Dissertations, & Professional Papers
Arbuscular mycorrhizal fungi (AMF) are soil fungi forming symbiotic associations with majority of land plants. AMF alter soil organic matter (SOM) directly through stabilization of soil aggregates and indirectly providing a path in which plant fixed C02 is transferred below-ground. Understanding contributions of AMF to SOM via protein production and stabilization of soil aggregates will greatly aid our understanding of soil carbon sequestration, nutrient cycling and mitigation of soil erosion. The work presented in chapter 2 challenges the glomalin extraction process and assesses the accuracy of the Bradford and monoclonal-antibody ELISA detection methods. My results clarify the contribution of glomalin …

Ecology And Molecular Phylogenetics Of Hydnora (Hydnoraceae) In Southern Africa, Jay Francis Bolin
Biological Sciences Theses & Dissertations
The Hydnoraceae are a clade of root holoparasitic angiosperms that contain two small genera, Hydnora and Prosopanche. This study, focused on Hydnora, presents novel data regarding the pollination biology, germination ecology, parasite-host nutritional relationships, and the molecular systematics of this group. Experimental addition of the primary pollinator, Dermestes maculatus to Hydnora africana chamber flowers demonstrated beetle imprisonment during the carpellate stage. Changes in the inner surfaces of the androecial chamber allowed beetle escape after pollen release. Most beetles escaped, dusted with viable pollen, three days after pollen release. To investigate germination ecology, aqueous root extracts of host and …

Analysis Of N-Linked Oligosaccharides Of Prostate-Specific Antigen And Prostatic Acid Phosphatase In Prostatic Fluids, Krista Yaudes White
Theses and Dissertations in Biomedical Sciences
Presently, prostate cancer is the most common cancer afflicting men in the United States, with serum PSA being the "gold standard" protein biomarker used in the clinic for detecting and diagnosing prostate cancer. Nonetheless, serum PSA levels can also be elevated in non-cancerous conditions as well, such as benign prostatic hyperplasia (BPH). Due to this overlap, many unnecessary biopsies and radical prostatectomies occur, leading to patient distress. Despite recent advances to clinical assays which consider other clinical parameters, there is still a great need for improved clinical detection methodologies for prostate cancer, including improved biomarkers. Therefore, this research project aims …
Computational Studies On R67 Dihydrofolate Reductase: An Investigation Into Its Unique Binding Patterns, Chuanyin Shi
Computational Studies On R67 Dihydrofolate Reductase: An Investigation Into Its Unique Binding Patterns, Chuanyin Shi
Theses and Dissertations in Biomedical Sciences
R67 dihydrofolate reductase (R67 DHFR) is a plasmid encoded enzyme which catalyzes the reduction of dihydrofolate (DHF) to tetrahydrofolate (THF) using NADPH as a cofactor. R67 DHFR is a homo-tetramer and D2 symmetric. It contains only one active site, which spans the central channel of the enzyme. The active site can bind either two reactants (DHF), two cofactors (NADPH) or one of each (NADPH/DHF), which is the productive ternary complex (i.e. the complex which yields product). In order to favor formation of the productive complex, this enzyme exhibits binding cooperativity. Unlike other allosteric enzymes which achieve binding cooperativity through conformational …

This volume has about 760 pages, split into 25 chapters, from 41 contributors. First part of this book presents advances of Dezert-Smarandache Theory (DSmT) which is becoming one of the most comprehensive and flexible fusion theory based on belief functions. It can work in all fusion spaces: power set, hyper-power set, and super-power set, and has various fusion and conditioning rules that can be applied depending on each application. Some new generalized rules are introduced in this volume with codes for implementing some of them. For the qualitative fusion, the DSm Field and Linear Algebra of Refined Labels (FLARL) is …
The Effects Of Varying Seeding Rates Of Bouteloua Curtipendula And Mowing On Native Plant Establishment In A New Prairie Reconstruction, Ryan Leon Welch
The Effects Of Varying Seeding Rates Of Bouteloua Curtipendula And Mowing On Native Plant Establishment In A New Prairie Reconstruction, Ryan Leon Welch
Dissertations and Theses @ UNI
A major problem early in prairie reconstruction is weed competition. Research has shown that mowing in the first year can increase establishment and survival of prairie plants. The use of nurse crops (companion crops) has been suggested as an alternative to mowing for weed suppression. The goal of this study was to examine various seeding rates of B. curtipendula, as a nurse crop in mowed and unmowed plots to determine if it suppressed weeds and aided in the establishment of natives. We hypothesized that increasing the seeding rate of B curtipendula will reduce weed growth and promote an increase in …

Potential For Range Expansion Of The Invasive Green Mussel, Perna Viridis, In The Southeastern United States, Alyson Goodwin Urian
UNF Graduate Theses and Dissertations
Cold temperatures are thought to be among the most important determining factors of geographic distribution for tropical and sub-tropical marine invertebrates. The Asian green mussel, Perna viridis, has been introduced into coastal waters of Florida where its current distribution is hypothesized to be limited by low temperatures during winter. Lethal and sublethal effects (heat shock protein/ Hsp70 expression) of cold water and air temperatures were analyzed in two size classes of P. viridis from Florida in an effort to determine the effects of current and forecasted temperatures on the potential for range expansion. Mussels were exposed to water temperatures …
The Effect Of Zinc On L-Leucine Transport In Lobster Intestine, Anna Lynn Mullins
The Effect Of Zinc On L-Leucine Transport In Lobster Intestine, Anna Lynn Mullins
UNF Graduate Theses and Dissertations
The current study is a characterization of L-leucine absorption across the American lobster intestine in order to determine the likelihood of a bis-complex formation with zinc ([Leu]-Zn-[Leu]) as a possible substrate for PEPT1. This study required isolated lobster intestines to be mounted in a perfusion chamber in order to quantify 3H-L-leucine transport. It was hypothesized that an apical dipeptide transporter, PEPT1, was responsible for uptake of L-leucine via formation of a bis-complex with zinc ([Leu]-Zn-[Leu]) in the form of molecular mimicry, where the bis-complex mimics the normal dipeptide substrate of the proposed carrier system. It was found that L-leucine transport …

Vegetation Response To Restoration Treatments In A Former Pine Plantation In North Florida, Ryan C. Meyer
UNF Graduate Theses and Dissertations
As the human population in Florida continues to expand, development follows, and tree farms give way to homes and businesses. As parks are established, restoration of these semi-natural plantations may provide critical habitat for species conservation. This study evaluates vegetation response to restoration treatments at two study sites, formerly tree farms, now preserves in NE Florida. Treatments included thinning, clearing, or control (no treatment) within 10m^2 plots. Thinning reduced tree canopy to 20% (2-3 pines/plot) and removed all other vegetation; clearing treatments removed all biomass to bare soil; no herbicides were used. Within these plots richness and abundance was assessed …

A Phylogeographic Analysis Of Fiddler Crabs, Uca Longisignalis And U. Pugnax, Cydney Brooks
Dissertations and Theses @ UNI
Uca pugnax and U. longisignalis are fiddler crabs commonly found in the southeastern United States. These sibling species occupy distinct regions. Uca pugnax is found on the Atlantic and U. longisignalis on the Gulf coasts. Their distributions are separated by the Florida peninsula. Many different taxa with similar distributions in this region have been the focus of phylogeographic studies. In these studies, it appears there is no simple pattern to explain genetic variability. Previous studies of Uca pugnax and U. longisignalis reveal no significant morphological or physiological differences within either species across its respective region. The purpose of this study …
Resistance To And Virulence Of Fusarium Verticillioides, Tracy Lynn Bruns
Resistance To And Virulence Of Fusarium Verticillioides, Tracy Lynn Bruns
Dissertations and Theses @ UNI
The filamentous fungus Fusarium verticillioides is plant pathogen that is responsible for billions of dollars in crop loss world wide. Fusarium verticillioides is of concern as it causes serious infections in maize and other agronomically important crop plants worldwide. Toxins produced by this fungus make grains from infected crops unusable. Plant resistance (or lack thereof) to disease is and has been for centuries a problem in agricultural productivity. Recent advances in the understanding of plant disease resistance promises to allow us to discover ways to counter the invasive properties of Fusarium verticillioides. Currently, very little is known about the virulence …

Oxidative Stress, Calcium Homeostasis, And Altered Gene Expression In Human Lung Epithelial Cells Exposed To Zno Nanoparticles, Chuan-Chin Huang
Masters Theses
"The influence of 20 nm ZnO nanoparticles on oxidative stress, intracellular calcium homeostasis, and gene expression was studied in human bronchial epithelial cells (BEAS-2B). ZnO caused a concentration- and time-dependent cytotoxicity while elevating oxidative stress and causing membrane damage (cellular LDH release). There was a remarkably steep relationship between concentration and toxicity at concentrations from 5 to 10 μg/ml. Exposure to ZnO increased intracellular calcium levels in a concentration- and time-dependent manner. Treatment with the antioxidant N-acetylcysteine prevented cell loss and diminished the increase in intracellular calcium concentration, suggesting oxidative stress mediated cytotoxicity. Exposure to a sublethal concentration of ZnO …

Limits To Maize Productivity In The Western Corn-Belt: A Simulation Analysis For Fully Irrigated And Rainfed Conditions, Patricio Grassini, Haishun Yang, Kenneth G. Cassman
Department of Agronomy and Horticulture: Faculty Publications
Unlike the Central and Eastern U.S. Corn-Belt where maize is grown almost entirely under rainfed conditions, maize in the Western Corn-Belt is produced under both irrigated (3.2 million ha) and rainfed (4.1 million ha) conditions. Simulation modeling, regression, and boundary-function analysis were used to assess constraints to maize productivity in the Western Corn-Belt. Aboveground biomass, grain yield, and water balance were simulated for fully irrigated and rainfed crops, using 20-year weather records from 18 locations in combination with actual soil, planting date, plant population, and hybrid-maturity data. Mean values of meteorological variables were estimated for three growth periods (pre- and …

Recently, bmr6 plants were shown to have limited cinnamyl alcohol dehydrogenase activity (CAD; EC 1.1.1.195), the enzyme that catalyzes the conversion of hydroxycinnamoyl aldehydes (monolignals) to monolignols. A candidate gene approach was taken to identify Bmr6. Two CAD genes (Sb02g024190 and Sb04g005950) were identified in the sorghum genome based on similarity to known CAD genes and through DNA sequencing a nonsense mutation was discovered in Sb04g005950 that results in a truncated protein lacking the NADPH-binding and C-terminal catalytic domains. Immunoblotting confirmed that the Bmr6 protein was absent in protein extracts from bmr6 plants. Phylogenetic analysis indicated that Bmr6 is …

We describe and analyze a pedagogical experiment that introduced a broad and holistic perspective on complete farming systems, systemic learning tools, and a participatory learning strategy at an early stage in agronomy education. The paper describes the adventure of three students, who came from a conventional agronomy program at the Swedish University of Agricultural Sciences (SLU), who were frustrated with the lack of integrated approaches to the study of agricultural systems and a strong focus on molecular-level processes in their first year of education. They encountered a narrow focus in most courses and the overall curricula of agricultural education that …

Alternative Methods For Wetland Restoration In The Rainwater Basin, Nebraska, Usa, Nancy Nicolai, James L. Stubbendieck
Department of Agronomy and Horticulture: Faculty Publications
South-central Nebraska is recognized as a focal point of the mid-continent migratory waterfowl flyway. Substantial wetland alterations led to a critical need for restoration. Managers have restored wetlands by scraping with heavy earthmoving equipment to remove excess organic material and near-monocultures of reed canarygrass (Phalaris arundinacea L.) but managers report high costs and topsoil perturbation. Moderate livestock grazing was tested to compare results with those achieved with mechanical techniques. Advantages of grazing could include low costs and less soil perturbation. During the 2006 and 2007 growing seasons, we compared cover of bare ground and open water and plant species …

Buffalograss [Buchloe dactyloides (Nutt.) Engelm.] use as a fairway turfgrass is limited in northern portions of its adaptation zone by its extended winter dormancy and tan coloration in early spring and late fall. Cool-season grasses mixed with buffalograss could enhance turfgrass appearance and performance in fall and early spring. Research was conducted near Mead, NE, with eight buffalograss genotypes maintained under fairway conditions to determine the effect of blue fescue (Festuca ovina L. var. glauca Lam.) overseeding rate on turfgrass performance. Interactions were nonsignificant in most cases so main effects are emphasized. Differences were observed between seeding rates and …

Department of Agronomy and Horticulture: Faculty Publications
Penstemon seed often shows an inconsistent or a low germination percentage. Although most select cultivars are propagated by cuttings, for export to other countries, seed is preferred. Three experiments were conducted to determine if soaking seed in gibberellic acid (GA3) would increase seed germination of Penstemon digitalis cv. Husker Red. GA3 concentrations used were 0, 10, 50, 100, 200, and 500 mg∙L–1 (first experiment); 0, 500, 1000, and 1500 mg∙L–1 (second experiment); and 0, 500, and 1000 mg∙L–1 (third experiment). The first and second experiments were conducted in a growth chamber, whereas the third …
