Changes In Sodium Concentration In Cardiac Myocytes From Diabetic Rats, Alice Warley

The effects of streptozotocin induced diabetes on rats were studied. The animals showed an increase in blood glucose concentration and a loss of weight from both the body and the heart. Loss of weight from the heart was less severe leading to an increased heart to body weight ratio. Study of element concentrations by X-ray microanalysis showed that there was an increase in intracellular Na concentration in cardiac myocytes from the diabetic animals, but no change in Mg. These results agree with studies which show changes in Na/K ATPase after the onset of diabetes.

Apex regions of continuously growing incisors of Wistar rats were quickly dissected, shock-frozen in liquid nitrogen-cooled propane, freeze-dried at -80 °C and infiltrated with Spurr's resin . 400nm thick dry sections were cut with a diamond knife on an ultramicrotome . Relatively flat sections were transferred with an eye lash onto collodium coated aluminum grids. They were flattened with a glass stick and by placing another collodi um coated aluminum grid just on top of the first one, exerting a uniform pressure . The electrophysiologic technique (Reuss, L., Proc. Natl. Acad. Sci. USA 82:6014, 1985) was modified to measure changes in steady-state hepatocyte volume during osmotic stress. Hepatocytes in mouse liver slices were loaded with tetramethylammonium ion (TMA$\sp{+}$) during transient exposure of cells to nystatin. Intracellular TMA$\sp{+}$ activity (a$\sp{\rm i}\sb{\rm TMA}$) was measured with TMA$\sp{+}$-sensitive, double-barreled microelectrodes. Loading hepatocytes with TMA$\sp{+}$ did not change their membrane potential (V$\sb{\rm m}$), and under steady-state conditions a$\sp{\rm i}\sb{\rm TMA}$ remained constant over 4 min in single impalements. Micro-PIXE (proton-induced X-ray emission) analysis was performed on unfixed and anhydrously prepared sections from developing enamel and dentin from hamsters injected with a single dose of 20 mg NaF /kg body weight. Fluoride, apart from inducing the formation of the characteristic paired response in the enamel (i.e., a hyper- followed by a hypomineralized band in the secretory enamel), also induces the formation of sub-ameloblastic cystic lesions under the transitional and early secretory enamel accompanied by relatively intense hypermineralization of the underlying cystic enamel surface. The effect of dietary aluminum sulfate on Ca and P metabolism was studied using 1-day-oldmale broiler chicks. In Experiment 1, practical diets providing .90% Ca plus .45% available P (Pav), .90% Ca plus .78% Pav, 1.80% Ca plus .45% Pav, or 1.80% Ca plus .90% Pav were fed with 0 or .392% Al as aluminum sulfate for 21 days. Dietary treatments providing three levels of added Al (0, .196, or .392%) as aluminum sulfate and of available phosphorus (Pav) (.45, .68, or .78%) in a factorial arrangement were administered to day-old chicks in Experiment 1. Plasma inorganic phosphorus (Pi) was significantly (P < .05) elevated by increasing Pav and was decreased by Al. Body weight gain, feed intake, and the gain:feed ratio at Day 21 were significantly decreased by increased concentrations of Al, but were unaffected by the Pav concentrations. Decreases of 39 and 73% in weight gain and of 34 and 66% in feed intake resulted from feeding …

Milk coagulation consists of four overlapping phases: enzymic hydrolysis, micelle aggregation, gelation and syneresis. The objectives of this study were to determine the effects of added CaCl2 on milk coagulation and the relationship between enzymic hydrolysis and micelle aggregation with substrate at different concentrations.

Addition of CaCl2 to milk is widely practiced in industry and in laboratories. This changes calcium concentration, pH and ionic strength. It is impossible to separate these three variables and investigate each one independently.

Addition of low levels of CaCl2 shortens coagulation time and increases curd firming rate. Low levels of CaCl2 …

Oat hulls and dry-milled corn bran were loaded with calcium or potassium and made into either sectioned bulk specimens of intact tissue embedded in resin or into non-sectioned bulk specimens made from powdered-compressed tissue formed into disks without resin. Regression lines of X-ray count versus mineral concentration were similar for both Ca and K. X-ray count versus mineral concentration relationships were similar for intact oat hulls and powdered-compressed specimens of either oat hulls or corn bran. However, the relationship for intact corn bran embedded in resin was significantly different. While the reason for this difference is not known, the result …

The applications of x-ray microanalysis in research into silicon in higher plants are reviewed, recent developments are assessed, and new data are presented. Conventionally prepared material [air or freeze drying for scanning electron microscopy (SEM), and glutaraldehyde/osmium tetroxide fixation for transmission electron microscopy (TEM)) has been studied using both wavelength and energy dispersive microanalysis. These techniques are reliable provided that the deposited form of silica is the major focus of investigation. Progesterone and 17 alpha-hydroxyprogesterone (but not other steroids such as testosterone, corticosterone, beta-estradiol, estrone, dehydroepiandrosterone, 20 alpha-hydroxypregnen-3-one, androstenedione, and pregnenolone) were shown to cause an immediate increase, in free cytosolic calcium ([Ca2+]i) in both capacitated and noncapacitated human sperm, using the fluorescent indicator fura 2. Significant increases in [Ca2+]i were observed with 10 ng/ml progesterone, while maximum effects were seen with 1 microgram/ml progesterone. Two other steroids 11 beta-hydroxyprogesterone and 5 alpha-pregnane-3,20-dione exhibited significant activity to increase [Ca2+]i. This increase in [Ca2+]i elicited by progesterone was entirely due to Ca2+ influx from the extracellular medium since the increase in [Ca2+]i …
