Site-Directed Mutagenesis Of The Putative Processing Site Of Recombinant Human Immunodeficiency Virus Reverse Transcriptase, Susan M. Poppe

The human immunodeficiency virus (HIV) is the etiological agent of the acquired immune deficiency syndrome (AIDS). This retrovirus, encodes a RNA dependent DNA polymerase, reverse transcriptase (RT), which is essential to virus replication. Biochemical analysis indicated that RT is composed of 2 polypeptides of 66,000 and 51,000 daltons (p66 and p51) which combine to form a heterodimer. This heterodimer is thought to arise from the post-translational processing where p66 is made and either autocatalytic or proteolytic cleavage events result in a heterogeneous p66/p51. Site-directed mutagenesis was used at the proposed processing site to probe HIV-RT processing.

Neanthes virens were subjected to a toxicity test with 1.8, 3.4, 6.7 and 11.0 mg/1 Cd2+. Worms were withdrawn on Days 1, 4, 7, 10 and 14 from each of five replicate tanks and their caloric content determined by bomb calorimetry. Analyses of variance and covariance showed no significant differences in worm caloric content with withdrawal day or cadmium concentration.

In a second experimental series, a sediment toxicity test with Neanthes virens was conducted in contaminated sediments from two sites, as well as reference sediments from a control site. Caloric content of the worms was measured by bomb …

In the eastern United States, Dermacentor variabilis is the primary vector of Rickettsia rickettsii, the etiological agent for Rocky Mountain Spotted Fever. Cases of Rocky Mountain Spotted Fever in eastern United States increased steadily from 1959 to 1979 to a peak of one case per 200,000 people in 1979 (Morbidity and Mortality Weekly Report, 1985). In 1976, 98.9% of the reported cases of Rocky Mountain Spotted Fever were east of the 100th meridian with 1.1% occurring west of the 100th meridian (Burgdorfer, 1977). Cloning And Sequencing Of The Eco Rii Monomer Fragment Of Satellite Dna From The Mouse Mus Musculus, Timothy Patrick Boyle

Total mouse satellite DNA was isolated in cesium chloride gradients in the presence of the dye Hoechst 33258. This satellite fraction was digested with Bst Nl, an isoschizomer of the restriction enzyme Eco RII, and the 234 base pair monomer fragment was cloned into the plasmid vector pBS. Dideoxynucleotide sequencing of the cloned DNA yielded significant homology with the published consensus sequence of mouse satellite DNA (92%).

Cholesterol oleate was demonstrated to be the cuticular contact sex pheromone of Dermacentor variabilis. This pheromone which has been termed the Mounting Sex Pheromone (MSP) was also demonstrated to be present on the surface of D. andersoni, Amblyomma maculatum and A. americanum. This contact sex pheromone enables males excited and attracted by 2,6-dichlorophenol to identify the female as a potential mating partner. The MSP is the second in the series of three sex pheromones guiding the hierarchy of behavioral responses which constitute tick courtship behavior. Remipedes are an unusual group of troglobitic crustaceans that live exclusively in anchialine caves. Since their discovery in 1979, nine species have been described, seven of which are found in caves in the West Indies, one from the Yucatan peninsula of Mexico and one from the Canary Islands. Most of what is known about these animals has come mainly from taxonomic descriptions. Little has been published about their reproductive biology or ecology. The objectives of this dissertation were to investigate the reproductive biology and ecology of the remipedes inhabiting Sagittarius Cave on Grand Bahama Island, Bahamas.

Neuroendocrine mediators have been shown to influence a broad spectrum of immune functions suggesting the existence of a neuroimmune regulatory axis effecting feedback control of the immune response. Most studies in support of this paradigm have employed either intact animals or immune cells derived from blood and other tissues. Under these conditions it becomes difficult to cope with numerous variables associated with the complex milieu of the immune system and the interpretation of data. Most insect circulatory physiologists believe the insect heart to be myogenic. There is disagreement, however, about mechanisms regulating heart function. Modulation is considered to occur via either neural or neurohormonal mechanisms but there is little existing data of the type necessary for proving one mechanism or the other. Using the recording technique of impedance conversion, heartbeat patterns were established for intact Honey Bees during rest and also during nectar feeding and locomotion, activities that both result in modulating heartbeat. The pebble nest microhabitats of four species of Semotilus were compared. Pit/mound nests of Semotilus corporalis were significantly larger than the pit/ridge nests of Semotilus atromaculatus, Semotilus lumbee, and Semotilus thoreauianus. Nests of S. corporalis were in wider and deeper streams. Pits of S. lumbee nests were longest of the four species; those of S. corporalis were widest and deepest. Semotilus corporalis used a greater proportion of larger stones (23 mm or greater) in nest construction than the other three species. Purification of Epstein-Barr virus-associated deoxyribonuclease (EBV-DNase) from Raji and P3HR-1 cells treated with 12-0-tetradecanoylphorbol-13-acetate and sodium butyrate was performed by sequential ion-exchange column chromatography and fast protein liquid chromatography. The enzyme activity, protein concentration, yield, specific activity, purification profiles, and polypeptide patterns by electrophoretic analysis in each column purification step were determined. The characteristics of the partially isolated EBV-DNase were demonstrated by the enzyme activity, DNA binding affinity, and inhibition by the nasopharyngeal carcinoma patient sera and rabbit polyclonal antibodies against the partially purified EBV-DNase. A nonisotopic assay was developed as a new method in detecting the nuclease. …

Mouse monoclonal antibodies directed against Pichinde virus (PCV) were produced to evaluate their application as vehicles for the delivery of antiviral drugs or toxins to virus-infected cells. Four monoclonal antibodies, PC4.9A6, PC4.9D3, PC4.7C2, and PC4.8D3, were of the IgG2a subisotype and reacted with acetone-fixed and live PCV-infected Vero-76 cells. In vivo stained splenic macrophages derived from PCV-infected hamsters that had been injected with fluorescein-labeled PC4.9A6 (FITC9A6) demonstrated a 400% increase in total fluorescence over similarly treated, non-infected cells when analyzed by flow cytometry. The purpose of this study was to identify the effects of the neurotransmitter, serotonin (SE), on the immune function of peripheral blood mononuclear cells (PBMC) of normal, healthy subjects. This was done as a preliminary investigation to studies on the association of SE with immune changes in autistic subjects.

The PBMC isolated from normal male subjects were treated with various concentrations of SE for 48 hrs. Their incubation in SE at a concentration of 10-3 M induced about a 35% decrease in DNA synthesis. However, incubation of the cells in lower concentrations (10-4 to 10-10) of …

A number of toxicities associated with methylene chloride have been found in both human subjects and mice. However, relatively few studies have probed immunotoxicities of methylene chloride. In order to examine possible immunotoxicities or immunomodulating effects of methylene chloride, several tests of cellular immune function were performed using both human in in vitro studies and a mouse model.

Body weights and specific organ weights of thymus, spleen, liver, and kidney were normal in CD-1 mice given various concentrations of methylenechloride. However, a significantly reduced mitogenic response to phytohemagglutinin (PHA} and reduced interleukin-2 (IL-2} production was found in these methylene-chloride-treated mice. …

Because of the damaging effects of bluetongue disease on the livestock industry, interest exists in developing a means of inducing passive immunity in infected animals. Antibodies against VP2, the polypeptide responsible for immunoantigenicity in bluetongue virus (BTV), have been found to elicit neutralizing antibodies; however, production of this purified antigen is very costly and thus impractical for commercial purposes. Total viral protein, prepared by a simple, rapid, highly reproducible SDS/KCl method, was injected into rabbits and found to produce antibodies which are useful in immunoblots. The mitochondrial DNA restriction fragment patterns of killer whales (Orcinus orca) were investigated in order to determine the level of genetic differentiation that exists between killer whales from various geographic locations. Twenty one killer whales were examined, seventeen of which were captive killer whales that originated from the North Atlantic and Northeast Pacific Oceans. Two were captive-born animals and two were killer whales that stranded along the Northeast Pacific coast.

Prolactin (PRL) acts directly on milk secretory cell nuclei to stimulate lactation in rats. In the cow, PRL has been detected in Golgi apparatus, endoplasmic reticulum, and secretory vesicle subcellular fractions of mammary gland , but not nuclear fractions. However, growth hormone {GH) is the galactopoeitic hormone in cows, not PRL. The possibility of GH acting directly on the cow milk secretory cells, and PRL acting on their nuclei was investigated using an immunocytochemical method. The investigations described in this paper are an attempt to further define and quantify the interrelationship of nitrogen fixation and iron nutritional physiology in these two species. Chapter II will present and compare data on nutritional ratios of field collected Trichodesmium colonies and laboratory Anabaena cultures, with the intent of examining possible correlations between observed iron levels and protein nitrogen and chlorophyll concentrations, as well as nitrogen fixation rates. Mechanisms of how red-sided garter snakes (Thamnophis sirtalis parietalis) travel up to 18 km from summer feeding sites to hibernation dens are not understood. In this study, monthly and seasonal trailing behavior were investigated to determine whether red-sided garter snakes prefer to follow trails of snakes from the same den (den-mates) versus trails made by other conspecifics (non-den-mates). Snakes from five different hibernacula in Manitoba, Canada, were involved in the study. Eighteen were adults and 15 were subadults. Subadult red-sided garter snakes do not return to hibernacula until their second year of life, so their trailing behavior was of interest …

The feasibility of using solid-phase resins for monitoring concentrations of pesticides in water was examined. Efforts were made to ascertain relationships between concentrations of a pesticide (methoxychlor) in water and on a resin (AXD-2) under laboratory conditions. Solutions of pesticide in water were drawn through columns of resin. Methylene Chloride was used to absorb the pesticide from the resin. Gas chromatography and ultraviolet spectrophotometry were used to measure concentrations of pesticide.

The lateral geniculate nucleus (LGNd) receives chemically identified inputs from brain stem structures, the thalamus and visual cortex. The identity of the neurotransmitter(s) in the retinal input, however, is unknown.

To investigate the possibility that some amino acids and certain dipeptides, such as N-acetyl-aspartyl-glutamate (NAAG), fulfill this function, changes in their concentration were measured in the optic tract, parvocellular and magno-cellular segments of the LGNd, superior colliculus and visual cortex of six monkeys (Macaca fascicularis), seven days after right optic tractotomy. This study investigated the small aquatic ecosystems of Santa Ana National Wildlife Refuge, located in a predominantly agricultural area of the Lower Rio Grande Valley, Texas. Fishes and water quality data were collected for 13 months to determine the status of fish populations and the factors which influence their distributions on the refuge.

Sixteen fish species were collected during the study, however, more than 20 species are known to inhabit the Rio Grande in the vicinity of the refuge. Water source played a major role in influencing fish species richness in these aquatic ecosystems. Diverted river water offered the best …
