Early Events Of Lethal Action By Tobramycin In Pseudomonas Aeruginosa, Jane Elizabeth Raulston

The immediate activities of the aminoglycoside antibiotic, tobramycin, were investigated in Pseudomonas aeruginosa PAO1. The influence of carbon growth substrate and the antibiotic exposure environment in the magnitude of activity were examined. Lethality by 8 ug/ml tobramycin occurred rapidly (1 to 3 minutes). The release of specific cellular component into the supernatant was associated with lethality. This material was initially detected as an increase in UV-absorbance. Magnesium in the reaction mixture provided protection against lethality and leakage, but did not reverse lethal damage after a 3 minute tobramycin treatment. The loss of color results as a darker sheet. There are 3 major variables which cause blackening: moisture content, nip pressures, and roll temperatures. The increase in any of these variables results in blackening. In this experiment, the increase of moisture content produced the most significant blackening. Pulp viscosity and zero span tensile tests can be used to predict the ultimate fiber strength of pulp fibers. A project to compare the traditional test with the newer zero span tensile test was completed. In order to compare both tests for precision, bleached softwood kraft pulp was disintegrated and made into handsheets. The handsheets were exposed to ION HCL vapor from a few minutes to a few hours to get a range in fiber strength since the HCl degrades the cellulose. Ellagic acid (EA), a naturally occurring plant phenol, at concentrations of 5 to 50 pg/plate, inhibited rat liver S9 protein dependent benzo[a]pyrene (B[a]P)-induced mutagenesis in Salmonella typhimurium TA 100 by 30-81% and B[a]P 7,8-dihydrodiol (DHD)-induced mutagenesis by 29 to 75%. EA (5-40 juM) did not significantly affect the metabolism of B[a]P or B[a]P 7,8-DHD as determined by high performance liquid chromatographic analysis of the organosoluble fraction and by the quantification of watersoluble conjugates. This study describes in a quantitative manner the relationships between coating structure and optical properties. Scattering coefficient, gloss and pore volume were measured after applying different blends of polystyrene spheres using various amounts of binder to transparent Mylar film. The coatings were prepared using four proportions of two sizes of polystyrene spheres and varying amount of two binders, starch and latex respectively. The objective of this research was to evaluate receptor localization and distribution on normal and diabetic cells by electron microscopic analysis.

To achieve this objective, gold-adsorbed and ferritin-conjugated macromolecules were used to determine the location and concentration of specific receptor binding sites on the cells surfaces. To facilitate the use of these ultra-structural histocytochemical markers, normal and diabetic Chinese hamster and other nondiabetic cells were either grown in vitro on ultra thin plastic films or embedded in Lowicryl K4M and pre- or post-labelled with these marker molecules.

Pineal "synaptic" ribbons are a heterogeneous population of organelles. In addition to "synaptic" ribbons (SR) sensu stricto, which are electron-dense rods, "synaptic" spherules (SS) and intermediate forms (IMF) are present. The SS are electron-dense spherical structures, the diameter of which is two to three times the width of the SR. The IMF appear as irregularly formed structures. Relationships between dietary nutrients and plasma and fecal estrone, estradiol-17β, testosterone, and plasma prolactin levels were studied in young Seventh-day Adventist men: 18 nonvegetarians (NV), 20 lacto-ovovegetarians (LOV), and 15 vegans (V). Blood samples and 3 d dietary records were obtained. Contemporaneously collected diet composites and stool samples were analyzed for fiber. The vegetarians consumed significantly more fiber than the omnivores, while NVs and LOVs consumed more saturated fat than the Vs. Although plasma steroid hormone status did not differ, the Vs had significantly higher fecal estrogen levels than the NVs or LOVs. Plasma prolactin levels were significantly higher in …

The effects of topography on the scene radiance of LANDSAT thematic mapper (TM) image were examined in the context of the remote sensing of vegetation m Great Smoky Mountains National Park (GSMNP).

To simulate the image intensity component due to varying orientation of surface elements, two reflectance models were adopted to generate the synthetic images from a Digital Elevation Model (DEM). Visual and analytical procedures were developed to register the TM image to the DEM data.

Censusing of cave arthropods has been carried out in three areas of Carlsbad Cavern from 1984 to 1987, using pitfall traps. The areas represent a food rich area (Bat Cave, a guano community with high species richness), a food poor area (Sand Passage, an area deep within the cave, with low species richness), and an area with moderate food resources (Left Hand Tunnel, an area near the Underground Lunch Room). The Pacific members of the genus Bollmannia Jordan 1890 comprise seven species of gobies distributed from Baja California to Peru and west to the Galapagos Archipelago.

Previous authors recognized nine nominal species, all known only from their brief original accounts. Those accounts do not allow for proper discrimination of the species. All nominal species were re­ examined using meristic, morphometric, and pigment-pattern characteristics. Three of the nine nominal species are herein considered synonymous with previously described forms. Despite the extensive literature on salamander cranial morphology, few authors mention the structure or development of the mandibular symphysis. Adult specimens representing eight caudate families and larval Ambystoma opacum, representing nine developmental stages, were evaluated histologically to determine the structure and development of the mandibular symphysis.

From this analysis it was determined that early in the ontogeny of the mandibular symphysis Meckel's cartilages are separated by a band of undifferentiated cartilage. As development progresses, this band of undifferentiated cartilage gradually transforms into a wedge of fibrocartilage which fuses to Meckel's cartilages.

A large scale preparation of human chorionic gonadotropin (HCG) from the urine of pregnant women was accomplished by using kaolin adsorption, NH₄OH extraction and DEAE-cellulose ion exchange. A recovery about 81% of HCG and a specific activity in the 2,000-3,000 IU/mg range was achieved using this purification method.

Two HCG activity fractions, HCG I & HCG II, were isolated from DEAE-cellulose chromatography by a stepwise sodium chloride gradient elution. HCG I was eluted at 0.01-0.02 M NaCl while HCG II was eluted at 0.12-0.02 M NaCl.

Drugs such as adrenoceptor antagonists and certain centrally acting agents are known to lower blood pressure and thus serve as clinically useful antihypertensive agents. Norepinephrine 4, a natural ∝-adrenoceptor agonist, is the major agent associated with increased blood pressure in the periphery. Based on a careful structure-activity comparison, a series of substituted oxazolidines, 20, 21, 22 and 23, have been designed as possible antihypertensive candidates. Specifically, 22 and 23 have been synthesized to serve as potential norepinephrine antagonists.

To determine the effects of salinity on adult and larval survival, egg deposition, egg hatching and metamorphosis, shrimp were held in the laboratory and exposed to treatment salinities ranging from O to 35 ppt. Tests on adult survival indicate a 96 hour LC-50 between 30 and 35 ppt. Longer term tests on adult survival showed high mortality in salinities greater than 15 ppt. Shrimp became ovigerous in salinities of O - 20 ppt with egg hatching and metamorphosis limited to O - 5 ppt. A study of phytoplankton vertical distribution at six stations along the northeast continental shelf of the United States was conducted in April 1984.

A total of 168 taxa were identified from nine phylogenetic groups collected at depths up to 150 m. Highest cell concentrations and cell volumes were in the upper 30 m. Greatest cell concentrations were produced by diatoms and a pico-nanoplankton component, with cell volumes mainly the product of diatoms and dinoflagellates. A high degree of variability was observed between phylogenetic groups within each station, but less variability was noted for phylogenetic categories at different stations.

A method was developed to analyze Acetylene, Ethylene, Ethane, Propylene and Propane levels in the ambient air. Chlorinated compounds within the operation retention time, Methyl chloride, Methylene chloride and Vinyl chloride, were monitored also.

Four sampling sites are located at Carteret, Elizabeth, Newark and Montville in New Jersey. Samples were taken during the summer of 1988. Three-day food records from a large group of marathon runners were analyzed and compared to various standards of dietary quality. Nutrient intake by the runners exceeded two-thirds the Recommended Dietary Allowance for all nutrients except for vitamin D and zinc in female runners. Energy and nutrient intake levels in the marathon runners were higher than those of the general US population except for fat and zinc in both males and females, and vitamin B12 in females. In general this group of runners consumed diets that would be considered more optimal than those of the general population. However, both caloric intake …

Plasmid transformation is an essential step in many gene cloning experiments. In Salmonella tynhimurium, mutants of rough strain have shown improved transformation frequency previously. In this work a stable restriction-minus (r^-) and modification-plus (m⁺) strain and its derivatives were constructed. Effect of restriction barrier on transformation of plasmid pBR322 was tested. Several factors which may affect transformation frequency have been systematically examined to determine the optimum conditions for transformation in S. tvphimurium. A deleterious effect of spaceflight on human health is the loss of bone associated with the prolonged lack of gravitational stress on the skeleton. A potential for bone loss is indicated by the hypercalciuria and decreased heel bone density determined in Skylab astronauts. Histological studies of juvenile laboratory rats in actual spaceflight and in simulation models document a suppression of bone formation with little alteration of bone resorption. The mechanism for net bone loss needs to be defined in order to develop measures for its prevention or correction.

Light touch and low-frequency vibration sense are often impaired in diabetic patients. Meissner corpuscles are distally located mechanoreceptors that respond to these stimuli. Peripheral sensory neuropathy in diabetes progresses from distal to proximal, therefore, Meissner corpuscles are ideally suited to study diabetic neuropathy in the C57BL/Ks (db/db) mouse.

Corpuscles from the forepaw digital pads of 9 diabetic and 9 nondiabetic littermates were studied by electron microscopy. These were divided into 3 age groups; 2.5 months, 10 months, and 17 months old.

Enzyme II^mtl of Escherichia coli was purified from mtlA-overexpressing E. coli strain LJ1112 extraction of isolated membranes with deoxycholate, followed by hexyl agarose and ω-amino hexyl agarose chromatography in Lubrol PX. A 10-fold increase in total pure protein and 2.5-fold increase in specific activity over previously reported procedures was obtained. Tryptic fragments of Enzyme II^mtl were also purified and identified as the membrane and cytoplasmic domains by protein sequencing and SDS-PAGE. Zooplankton samples were collected from 166 Oregon lakes. The lakes included a wide range in size, trophic status, and water quality. Lakes were located throughout the state. Zooplankton were identified using standard taxonomic keys and counted. Seventy-four species were identified from the 200 total samples, including 32 species of cladocera, 22 copepods, and 11 rotifers. Two species of copepod were recorded for the first time in Oregon: Diaptomus mississippiensis and Diaptomus pallidus. Seven species had widespread distributions throughout most of the watersheds in the state. Seventeen other species had distributions restricted to eastern, central, or western Oregon.
