The purposes of these studies were to determine differences in total (TCF), medial (MCF) and lateral (LCF) tibiofemoral compartment compressive forces and related muscle forces between limbs (replaced, non-replaced, and control), and different slopes during uphill [0° (level), 5°, 10°], and downhill [0° (level), 5° 10°] using statistical parametric mapping (SPM). Static optimization was used to determine muscle and compressive forces for 9 patients with total knee arthroplasty (TKA) and 9 control participants during walking trials. Epigenetic mechanisms, including histone and DNA methylation and microRNAs, play key roles in mediating transcriptional changes during plant development and stress responses. However, how these interconnected epigenetic components regulate gene expression in a spatiotemporal fashion remains partially known. Here, I generated 15 transgenic Arabidopsis GUS reporter lines for genes involved in DNA methylation and demethylation pathways. The spatiotemporal expression patterns of these genes were profiled in various plant organs during development, exogenous phytohormone response, and plant-parasitic nematode pathogenesis. The analyses revealed unique and overlapping expression patterns in roots, shoots, and reproductive organs, emphasizing the importance of a DNA methylation—demethylation equilibrium. …

This dissertation assesses fundamental social factors that drive variation in calling and other behaviors of experimental flocks of Carolina chickadees. Specifically, I tested how group member familiarity and group composition affected individual behavior. In addition, I performed a direct experimental test of the Social Complexity Hypothesis for Communicative Complexity, which states that groups that are more socially complex will communicate with greater signal complexity than groups that are less social complex. Lignocellulosic biomass is a potential energy source for fuels, chemicals and materials production in a sustainable manner. A network of covalent and non-covalent bonds between the three main polymers of biomass, i.e., cellulose, hemicellulose and lignin, results in a compact structure that is resistant to chemical and biological attacks and therefore challenging for the efficient utilization of lignocellulosic biomass. Morbid obesity is linked to physical and psychological well-being. Bariatric surgery has shown tremendous success with rapid weight loss in the patient population with morbid obesity. These patients experience issues with weight regain post-surgery, which can be linked to psychological and social factors. Despite this, mental health counseling is rarely offered in bariatric surgery programs. Stratified storage of dormant seeds in soil can result in natural archives useful for studying evolutionary responses to environmental change. However, few studies leverage soil-stored seed banks as natural archives, in part because of concerns over attrition, bias, and sediment mixing. Here, I examine the persistent seed bank of Schoenoplectus americanus, a foundational brackish marsh sedge, to a) determine whether it can serve as a resource for reconstructing demographic and population genetic/genomic variation, b) whether and how evolution may be occurring across a century. Selective gene expression is crucial in maintaining the self-renewing and multipotent properties of stem cells. Mediator is a large, evolutionarily conserved, multisubunit protein complex that modulates gene expression by relaying signals from cell type-specific transcription factors to RNA polymerase II. In humans, this complex consists of 30 subunits arranged in four modules: head, middle, tail, and kinase. In our introduction, we show the state of the field of Mediator study with a focus on the critical kinase module. As they have adapted to these conditions, Angiosperms diverged in their niche breadth, morphological, physiological, biochemical, and phenological traits and evolved into the plant species known today. We present two models of the Adelges tsugae, the hemlock woolly adelgid, an invasive insect pest of Tsuga canadensis, eastern hemlock, in the eastern United States. An A. tsugae infestation often results in the death of T. canadensis within years, and has caused significant changes to hemlock forests. We construct two models composed of systems of ordinary differential equations with time dependent parameters to represent seasonality. In plants, the Leucine-Rich Repeat Receptor-like Kinases (LRR-RLKs) encode a family of membrane-bound receptors that transduce such cell-to-cell signals and are required for many aspects of plant development. Very little is known about the function of most of these genes, and the evolutionary history of the family is difficult to infer because of its size and complexity. Dissimilatory iron reduction by hyperthermophilic archaea occurs in many geothermal environments and typically relies on microbe-mineral interactions that transform various iron oxide minerals. However, the kinds of iron oxides that can be used, growth rates, extent of iron reduction, and the mineral transformations that occur due to this metabolism are poorly understood. Mental fatigue causes an increase in task-based EEG theta and alpha power and a decrease in performance (for a review, see Tran et al., 2020). However, little is known about the emergence of mental fatigue in resting state EEG recordings and whether the progression of mental fatigue over time is influenced by individual differences. Gait asymmetries are associated with a high incidence of lower back pain (LBP). Although there are several causes of gait asymmetry (i.e. amputation, injury, or deformities), lower back kinetic demands have not been quantified and suitably compared due to experimental limitations in these clinical populations. The most consequential of these pathways involves the nuclear translocation of NF-kB, a transcription factor that induces the transcription of multiple proteins associated with cell survival, inflammation, proliferation and death. It is activated when the fibroblast growth factor-inducible 14 kDa protein (Fn14), a trimeric receptor recruits its ligand, TWEAK. ”The increasing applications of emerging and fugitive contaminants (EFCs) and engineered nanoparticles (ENPs) attract significant research interest for their potential risks to human health and the environment.
