Biomass And Morphological Development Of Miscanthus Compared With Native Grass In South Dakota, Ehsan J. Al Ainizi
Biomass And Morphological Development Of Miscanthus Compared With Native Grass In South Dakota, Ehsan J. Al Ainizi
Electronic Theses and Dissertations
Little is known about Miscanthus for bioenergy in the northern Great Plains. This study compared 10-year-old stands of Miscanthus x giganteus to switchgrass (Panicum virgatum L.) and prairie cordgrass (Spartina pectinata Link) for biomass on marginal cropland in eastern South Dakota. This study was conducted during 2017 and 2018 at Brookings, SD. Day of year (DOY), species, and N fertility were evaluated for effects on biomass yield and related components (tillers m-2, plant height, and weed biomass). All three variables had significant effects on biomass yield, tiller m-2, and tiller height in both years. N rate had no effect on …
Crispr/Cas9 Whole Genome Screens Reveal Novel Regulators Of Endocytic Processes In Macrophages, Jared Wollman
Crispr/Cas9 Whole Genome Screens Reveal Novel Regulators Of Endocytic Processes In Macrophages, Jared Wollman
Electronic Theses and Dissertations
Macrophages maintain tissue homeostasis by identifying and eliminating threats within their tissue microenvironment. Pattern recognition receptors allow macrophages to recognize and internalize specific ligands while macropinocytosis allows the internalization of all extracellular solutes from their environment. Without pattern recognition receptors, pathogens may grow unchecked if they cannot be detected, and without macropinocytosis, macrophages struggle to detect and move around their environment. This thesis presents the results of CRISPR/Cas9 whole-genome screens used to identify the regulators of both endocytosis (Chapter 2) and macropinocytosis (Chapter 3). In Chapter 2, we report genes regulating dextran uptake in primary murine macrophages and reveal Mrc1-mediated …

Winter Patch Grazing, Patch Burn Grazing, And Bird Communities In Western South Dakota, Jennifer Lutze
Winter Patch Grazing, Patch Burn Grazing, And Bird Communities In Western South Dakota, Jennifer Lutze
Electronic Theses and Dissertations
Ground nesting bird species are on a considerable decline and research efforts are being made to increase these populations throughout the Great Plains. Ground nesting bird communities found in the Northern Great Plains are driven greatly by varying amounts of cover and area size. Past research implemented patch-burn grazing to increase structural heterogeneity and to increase grassland bird habitat in the tallgrass prairie. While bird populations were very responsive to this management in the Northern Great Plains fire management is viewed negatively, especially for Midwest cattle ranchers. We implemented research to determine if winter-patch grazing on mixed grass prairie could …
Characterization Of Bacterial Endophytes Isolated From Brassica Carinata And Their Potential Use To Decrease Nutrient Requirements In Crops, Alex Soupir
Electronic Theses and Dissertations
Bacterial endophytes have the capability to enhance plant growth by producing plant growth hormones, solubilizing phosphates, suppressing pathogenic fungi, and reducing plant stress hormones. These capabilities make them desirable limiting the amount of nutrients and pesticides that are applied to crops. Through these assays and isolations, it is possible to identify novel bacterial species. In-vitro testing had shown 9 of the 20 isolates possess the ability to produce indole-3-acetic acid (IAA) with Pantoea agglomerans BC09 producing a concentration of 30.2 ng/μl over 4 days. BC09, Bacillus subilis BC10, and Pantoea sp. BC12 were able to solubilize calcium phosphate, 7 endophytes …
Partitioning Variation In Measurements Of Beef Carcass Traits Made Using Ultrasound, Bradie M. Schmidt
Partitioning Variation In Measurements Of Beef Carcass Traits Made Using Ultrasound, Bradie M. Schmidt
Electronic Theses and Dissertations
Ultrasound technology provides cattle breeders a relatively quick, non-invasive, and economical way to gather carcass data on live animals. Ultrasound provides the means to accurately predict body composition and develop estimated breeding values; however, national cattle genetic evaluations assume homogenous additive genetic and residual variances. These assumptions may be violated when estimating genetic merit for carcass traits by ultrasound because of differences in variance due to scanning technician and image interpretation laboratory. The objective of this study was to partition the phenotypic variance of measurements of carcass traits that were made using ultrasound into components attributable to additive genetic effects, …
Molecular Facet Of Host-Pathogen Interactions In Fusarium Head Blight In Wheat ( Triticum Aestivum L.), Bimal Paudel
Molecular Facet Of Host-Pathogen Interactions In Fusarium Head Blight In Wheat ( Triticum Aestivum L.), Bimal Paudel
Electronic Theses and Dissertations
Fusarium head blight (FHB) is a severe disease of wheat (Triticum aestivum L.). It not only reduces the quantity of the harvested grains but also decreases the grain quality due to mycotoxins contamination, especially Deoxynivalenol (DON). Qfhb1 (or simply called Fhb1) is the most important quantitative trait locus (QTL) for FHB resistance. Our lab has previously identified wheat gene WFhb1-1 (aka. Wfhb1_c1) as a candidate for FHB resistance gene. Here we report that WFhb1-1 has been cloned. The gene (GenBank # KU304333.1) consists of a single exon, encoding a putative membrane protein of 127 amino acids. WFhb1-1 protein produced in …
The Relationship Between Food Rules And Childhood Obesity In Rural And Non-Rural Families, Anna Venjohn
The Relationship Between Food Rules And Childhood Obesity In Rural And Non-Rural Families, Anna Venjohn
Electronic Theses and Dissertations
Background: Childhood obesity is continuing to rise, leading to long-term health consequences. Research shows that rural populations have higher rates of childhood obesity. There is a lack of research on how the home environment may affect this health disparity. Parents often enforce food rules to control their child’s eating habits, but the difference between the rural and non-rural populations in enforcing these rules is unknown.
Purpose: The purpose of the present study was to determine if there is a significant difference in food rules between rural and non-rural school-aged children, and if these differences correlate to BMI categories.
Methods: Secondary …
Optimization Of Conditions For Heat Pretreatment And Enzymatic Predigestion Of Ddgs For Pigs, Kevin Jerez-Bogota
Optimization Of Conditions For Heat Pretreatment And Enzymatic Predigestion Of Ddgs For Pigs, Kevin Jerez-Bogota
Electronic Theses and Dissertations
The high fiber content of corn dried distillers’ grains with solubles (DDGS) can limit its utilization in swine diets. Pretreatment with heat and enzymatic predigestion of whole stillage (WS; slurry material that is dried into DDGS) can alleviate negative effects of dietary fiber and can improve digestive and fermentation characteristics of the feedstuff. However, optimal time and temperature of the heat pretreatment of WS, and best enzymes for predigestion of WS have not been identified. Experiments were conducted to identify optimal conditions for heat pretreatment and multienzyme predigestion of WS for pigs. First experiment was conducted to identify optimal temperature …
Genetic Study Of Alkane Production And Oxic Nitrogen Fixation In Anabaena Sp. Pcc 7120, Jaimie L. Gibbons
Genetic Study Of Alkane Production And Oxic Nitrogen Fixation In Anabaena Sp. Pcc 7120, Jaimie L. Gibbons
Electronic Theses and Dissertations
Anabaena sp. PCC 7120 is a filamentous, nitrogen-fixing cyanobacterium that uses spatial separation to perform photosynthesis and nitrogen fixation simultaneously. Under fixed nitrogen limiting conditions, Anabaena 7120 forms specialized cells, called heterocysts, to fix nitrogen. In this research, I sought to add to the knowledge surrounding the process of heterocystous nitrogen fixation in Anabaena 7120. Cyanobacteria universally produce alkanes, which have been suggested to play a role in helping the organism adapt to abiotic stress, such as diazotrophic conditions. In my first study, I sought to identify the genes required for production of the hydrocarbon heptadecane (C17H36). Through using a …

Phytoremediation Of Saline-Sodic Soils In East Central South Dakota Utilizing Perennial Grass Mixtures, Douglas J. Fiedler
Electronic Theses and Dissertations
Several decades of above average precipitation in South Dakota has increased the area of saline and sodic soils, which reduce crop yields and inhibit sensitive plant growth. Saline and sodic soils are difficult to remediate using traditional agricultural crops. Establishing salt tolerant perennial species may restore productivity to salt affected areas. Two perennial grass mixtures (mix 1: slender wheatgrass, beardless wildrye; mix 2: slender wheatgrass, western wheatgrass, green wheatgrass, creeping meadow foxtail) were dormant frost seeded along a topographic gradient in Clark Co., SD. Soils were Forman-Cresbard loam and a Cresbard-Cavour loam with surface electrical conductivity (EC1:1) that …

While the gut microbiome has been investigated more deeply and continues to be a great interest, the oral microbiome is in comparison a more recent subject with fewer reports on the topic. The purpose of this review paper is to highlight the main human systemic diseases associated with the oral microbiome and to discuss how our understanding of the oral microbiome’s effect …
Freestanding Emergency Departments: A Possible Solution To Rural Hospital Closure, Emily Steadman
Freestanding Emergency Departments: A Possible Solution To Rural Hospital Closure, Emily Steadman
Biology and Microbiology Graduate Students Plan B Research Projects
Freestanding emergency departments have become more and more common over the years and have started to pop up in many states across the country. A freestanding emergency department is a facility that provides healthcare in the form of emergency services separate from those at a hospital. FEDs can be seen as independent structures that are distinct from hospital-based emergency rooms and urgent facilities. Just as freestanding emergency departments have spread across the country in a bit of a frenzy, another prevalent issue has been sweeping the country, the closure of rural hospitals. Over the last decade rural hospitals have struggled …

Role Of Adiponectin In Neuroinflammation & Neuroprotection, Nathan Stadem
Biology and Microbiology Graduate Students Plan B Research Projects
Microglial cells play a significant role in defense of the brain. However, in certain disease states, their response can lead to inflammation that can be detrimental to the survival of brain cells and have harmful effects on a person’s brain activity. Currently, the treatment options for microglial induced neuroinflammation are relatively limited. Adiponectin has recently appeared as a potential moderator of neuroinflammation as well as a promoter of neuroprotection. Adiponectin is a naturally occurring cytokine that has been shown to play a role in a multitude of processes throughout the body including potential as a microglial induced neuroinflammation. Adiponectin’s role …
Hiv Vaccine Development: The Importance Of Gp120 Variable Loop 3 For The Survival Of The Human Immunodeficiency Virus And The Potency Of The Neutralizing Antibody To Bind To The Variable Loop 3, Riham Hussien
Biology and Microbiology Graduate Students Plan B Research Projects
The tendency for the Human Immunodeficiency Virus (HIV) to proliferate and mutate it’s trimeric envelop (Env), is one of the reasons why it’s a challenge for an antibody-based vaccine to be developed. Glycoprotein 160 (gp160,) which consists of glycoprotein 120 (gp120) and glycoprotein 41 (gp41), is the main focus for a research vaccine design. Gp120 consists of an outer membrane, inner membrane and bridging sheet. The protein is trimeric, and non-covalently binds to gp41. The outer domain is where the neutralizing antibody bind, such as 2G12, immunoglobulin G1b12, and anti-V3 antibodies. V3 stands for the Variable loop 3, gp120 has …

Intermittent Fasting (If) Promotes Longevity Through Alterations Of The Mammalian Target Of Rapamycin (Mtor) And The Epigenome, Tayt Boeckholt
Biology and Microbiology Graduate Students Plan B Research Projects
Many studies with a range of subjects from Nematodes to Homo sapiens have found intermittent fasting (IF) to significantly improve the cardiometabolic health of individuals, but how IF promotes longevity through epigenetic modulations remains a sparse understanding throughout the literature. The process of aging may be characterized by a loss of cellular identity sprouted from a disrupted epigenome rich with information for a cell, while also losing the ability to recycle ineffective cellular components. The nutrient-sensing kinase Mammalian Target of Rapamycin (mTOR) is disrupted during bouts of fasting which allows for the recycling of cellular components through increased autophagy. Furthermore, …

Antidepressant Effects On Rem Sleep, Mitchel Adams
Antidepressant Effects On Rem Sleep, Mitchel Adams
Biology and Microbiology Graduate Students Plan B Research Projects
Individuals who deal with depression are typically prescribed antidepressants in order to alleviate the depressive symptoms that they may be having. Polysomnographic sleep research has revealed that depression is associated with altered sleep architecture and distorted REM sleep quality.2 Specifically, increased REM sleep duration and density have proved to be markers that predict recurrence and relapse for depressed individuals. REM sleep serves to stimulate regions of the brain that are important with learning and memory consolidation. However, an increased duration of REM sleep in depressed individuals proves to be unfavorable. Antidepressants drugs aim to weaken sleep quality, which is …

In Planta Evaluation Of Gene Regulation By Soybean Auxin Response Factors, Pratiksha K C
Electronic Theses and Dissertations
Nitrogen (N) is an important plant nutrient but its abundance in the soil is not sufficient for profitable crop production. N input in the form of chemical fertilizers helps fill this need; however, an alternative for fertilizers is an immediate need due to the environmental pollution resulting from excessive use of fertilizers. Leguminous plants such as soybean (Glycine max) that form root nodules through symbiotic association with N-fixing rhizobia and therefore need little or no chemical N fertilizers. The plant hormone auxin plays a crucial role in determining the number of nodules and their rate of maturity in soybean. Auxin …

Influenza viruses are respiratory pathogens that cause significant mortality worldwide. The subtype of influenza A virus currently affecting worldwide equine populations is H3N8, leading to epidemics and transboundary pandemics. The individual gene segments of an isolate named A/equine/Montana/9564-1/2015 were phylogenetically characterized. BLASTn search revealed that the polymerase basic protein 1 (PB1), polymerase acidic (PA), hemagglutinin (HA), nucleoprotein (NP), and matrix (M) segments of this H3N8 isolate shared the highest percentage identity to A/equine/Tennessee/29A/2014 (H3N8) and the polymerase basic protein 2 (PB2), neuraminidase (NA), and non-structural protein (NS) segments to A/equine/Malaysia/M201/2015 (H3N8). Maximum likelihood phylogenetic trees constructed using H3N8 viral genomes …
Identification And Genomic Characterization Of Candidate Starch And Lactate Utilizing Bacteria From The Rumen Of Beef Cattle, Venkata Bandarupalli
Identification And Genomic Characterization Of Candidate Starch And Lactate Utilizing Bacteria From The Rumen Of Beef Cattle, Venkata Bandarupalli
Electronic Theses and Dissertations
The rumen functions as an anaerobic fermentation chamber where microbial communities ferment feedstuffs into SCFAs. These SCFAs are critical to the ruminant host since they fulfill 70% of its energy requirements. In intensive management systems, ruminants are fed with grain-rich diets, which contain high levels of starch that help meet the energy demands of high producing animals. When grain-based diets are fed to ruminants, ruminal amylolytic and lactate metabolizing bacteria predominate. Grain overload can cause rapid production and accumulation of lactate, resulting in proliferation of amylolytic bacteria and death of lactate metabolizing bacteria. Consequently, high lactate levels result in a …

Is Vegan Confectionary Sold In The United Kingdom Safe For Milk Allergy Sufferers?, Faye Harrison
Electronic Theses and Dissertations
Background
Milk allergy is the most common childhood allergy. Reading food labels is important to prevent an allergic reaction. By United Kingdom (UK) law, milk needs to be listed on the food label if it used as an ingredient. Products can be called ‘free from’ milk if manufacturers have undergone rigorous process and testing to ensure it is not contaminated with milk. Precautionary Allergen Labelling (PAL) like ‘may contain milk’ statements are unregulated and can be used at manufacturer’s discretion. Similarly, ‘Vegan’ products are not regulated by law and appear to be on the increase. Objective
The study aimed to …

Decadal Heat And Drought Drive Body Size Of North American Bison (Bison Bison) Along The Great Plains, Jeff Martin, Perry S. Barboza
Natural Resource Management Faculty Publications
Large grazers are visible and valuable indicators of the effects of projected changes in temperature and drought on grasslands. The grasslands of the Great Plains have supported the greatest number of bison (Bison bison; Linnaeus, 1758) since prehis-toric times. We tested the hypothesis that body mass (BM, kg) and asymptotic body mass (ABM, kg) of Bison decline with rising temperature and increasing drought over both temporal and spatial scales along the Great Plains. Temporally, we modeled the relationship of annual measures of BM and height (H, m) of 5,781 Bison at Wind Cave National Park (WICA) from 1966 to 2015. …
